Abstract:To evaluate the peptide profiling using stable peptide peaks of human follicular fluid (HFF) analyzed by MALDI-TOF MS as potential biomarkers for follicle maturation. Prospective study. 3 HFF samples were collected from 3 women undergoing in vitro fertilization. Oviduct factor infertility was the only indication. This study included clear HFF containing mature oocyte (Fig. 1).The study design was approved by the Ethical Review Committee of Women’s Hospital. 1uL HFF (100mg/mL) was digested by 2.0ug proteomics-grade trypsin. The samples were incubated for 6h at 37°C. Digestion reaction was stopped by adding 2uL 500mM HCl, followed by using ZipTip C18 to clean up samples. MALDI-TOF MS analysis was performed after spotting 2uL mixture of tryptic digests of HFF samples and MALDI matrix solution (10 g / L in a-cyano-4-hydroxycinnamic acid (ACN): tryfluoroacetic acid (TFA), 50: 0.05%) on a MALDI plate. Spectra were analyzed using the Data Explorer software. P14R (M r 1533.86) and Insulin B chain (M r 3494.65) were used for external calibration. Each experiment was independently performed 3 times and each sample was measured 7 times. The mass spectra of each sample revealed many intense signals within the mass range of m/z 800-3000 (Fig. 2). The reproducibility of numbers and intensities of peptide peaks (S/N>5) was determined in the 3 samples, each analyzed 7 times. The average number of peptide peaks per sample was 508.27±141.13 (coefficient of variance (CV) 27.77%). In the 100%, 75%, 50%, and 25% frequency categories, 73.67 (CV28.64%), 108.0(CV24.26%), 261.67 (CV54.19%), and 625.67 (CV25.95%) peaks on average were detected, respectively. After correction for the background, the 3 samples provided 33 peptide peaks in 75% of all spectra. In addition, 19 peptide peaks stabilized (100%) in all spectra. The mass range of the 19 peptide peaks was mainly within m/z 1000 -2000. Observations indicated that most of the stable peptide peaks were intense signals. The average CV of intensities of the 19 peaks was 72.84%. After normalization by relative intensities, the average CV decreased to 48.68 % (Fig. 3). Influence of clean up tryptic digests of samples on the mass spectra quality was tested. After clean up, improvement was made to the quality of spectra resulting in more intense peptide peaks (Fig. 4). In the present study, we demonstrated that direct analysis of HFF peptide mixtures by MALDI-TOF MS could provide large number of peptide peaks with high mass accuracy. The reproducibility of the numbers of peaks for peptide profiling is higher comparing to the intensities of peaks. Thus, a valuable method to analyze peptide profiling of HFF would only require the presence or absence of a peak and the combination of information from multiple measurements per sample. This method is technically possible to find stable and specific peptide peaks as biomarkers that are related to maturation of follicle. However, a large number of samples with follicles at different growth stages are needed to provide detailed information. To our knowledge, there are no previous reports on the analysis of MALDI-TOF MS measurement of HFF peptide mixtures.
