Cloning and Sequence Analysis of Growth Hormone Gene from Alpaca

Abstract:RT-PCR and PCR were used to get the growth hormone(GH) gene from alpaca.The results showed that the cDNA sequence with the complete CDS was GH protein of 812 bp and the DNA sequence was 1 800 bp.Comparison DNA to cDNA sequences suggested that the GH gene sequence included 5 exons and 4 introns,encoding 216aa,including signal protein of 26aa and matured protein of 190AA.Based on the analysis of the similarity of GH genes in different species on the level of cDNA,DNA and the deduced amino acid,there was the most homology to the camel and comparatively high homology to pig,horse,dog and cat.However,the promotor of alpaca GH gene was not specific TATA box,but was CATA box like camel.It was also showed that the alpaca GH have no apparently functionally difference from human and pig GH based on the analysis of the functional site of GH protein.
