Abstract:An NMR pulse sequence was used to study the signal features from intermolecular multiple-quantum coherences between spin 1/2 and quadrupolar nuclei in solution NMR for the first time. General theoretical expressions were derived from the raising and lowering operator formalism. Theoretical predictions for the relative signal intensities and the optimal pulse flip angles are in excellent agreement with experimental results. Both the magic angle gradient and 2D experiments validate that the signals are indeed from intermolecular heteronuclear multiple-quantum coherences. (c) 2008 Elsevier B. V. All rights reserved.

Abstract:The behaviors of molecular self-diffusion were simulated in complex spin systems with both intra-molecular scalar couplings and inter-molecular dipolar couplings in liquid nuclear magnetic resonance (NMR). The simulation algorithm was based on a combination of the non-linear Bloch equations, product operator matrix, and finite difference method. The simulated results reveal different diffusion behaviors of inter- and intra-molecular multiple-quantum coherences, coincident with theoretical predictions and experimental measurements. Compared with the Monte Carlo method, the finite difference method is more precise and efficient for simulating diffusion behaviors of multiple-quantum coherences.

Abstract:High-resolution nuclear magnetic resonance (NMR) spectroscopy with information on chemical shifts and J-coupling constants is a sensitive tool for studying physical, chemical, and biological properties of materials at the molecular level. In this paper, a pulse sequence is developed for acquiring high-resolution NMR spectra of liquid samples with J-scaling in inhomogeneous fields via two-dimensional intermolecular multiple-quantum coherence acquisitions. In the resulting one-dimensional projection spectra, apparent J-coupling constants were obtained with a scaling factor theoretically varying from zero (completely decoupled) to infinity relative to the original J-coupling constants while retaining information on chemical shifts, relative peak areas, and multiplet patterns. This allows either an accurate measurement of small J-coupling constants of weakly coupled spin systems or less crowded spectra for spin systems with J-splitting. Experimental observations and simulation results agree with theoretical analysis.

Abstract:Intermolecular multiple-quantum coherences(iMQCs) originated from distant dipolar interactions among spins in different molecules have generated tremendous interests due to its unique properties.Compared to the conventional single-quantum coherence(SQC),iMQCs have following features:firstly,iMQC signals are related to the spins within the range of dipolar correlation distance;secondly,iMQC signals are related to the distribution of local field and susceptibility;finally,iMQCs hold special relaxation and molecular diffusion properties.IMQCs have been widely applied in NMR spectroscopy and MRI.In this review,we firstly made a brief introduction on the research history of iMQC phenomena and related theoretical explanations.We then expatiated on our research in iMQC field during the past ten-plus years,from the early research on the iMQC properties to the iMQC applications in NMR spectroscopy and MRI.

Abstract:Great progresses have been made in the past decades in understanding intra-and inter-molecular multiple-quantum coherences(MQCs) and developing their applications in nuclear magnetic resonance.To develop further applications for MQCs,it is useful to understand their physical bases and ways of practical implementation.In this review,the principles of dipolar coupling are described firstly.Comprehensive descriptions of MQCs based on intra-and inter-molecular dipolar coupling are then summarized,respectively.Finally,the applications of intra-and inter-molecular MQCs in chemistry and biomedicine are presented.

Abstract:Strong and extremely homogeneous static magnetic field is usually required for high-resolution nuclear magnetic resonance (NMR). However, in the cases of in vivo and so on, the magnetic field inhomogeneity owing to magnetic susceptibility variation in samples is unavoidable and hard to eliminate by conventional methods such as shimming. Recently, intermolecular multiple quantum coherences (iMQCs) have been employed to eliminate inhomogeneous broadening and obtain high-resolution NMR spectra, especially for in vivo samples. Compared to other high-resolution NMR methods, iMQC method exhibits its unique feature and advantage. It simultaneously holds information of chemical shifts, multiplet structures, coupling constants, and relative peak areas. All the information is often used to analyze and characterize molecular structures in conventional one-dimensional NMR spectroscopy. In this work, recent technical developments including our results in this field are summarized; the high-resolution mechanism is analyzed and comparison with other methods based on interactions between spins is made; comments on the current situation and outlook on the research directions are also made.

Abstract:Intermolecular zero-quantum coherences (iZQCs) originating from distant dipolar interactions between spins in different molecules provide an innovative way for nuclear magnetic resonance (NMR) spectroscopy. Since the field experienced by iZQC signals is in the dipolar correlation distance, it is naturally to apply iZQCs for resolution enhancement in NMR spectroscopy, especially under the spatially and/or temporally varying magnetic fields. Two theoretical frames, classical distant dipolar field and quantum-mechanical intermolecular multiple-quantum coherence treatments, are available for the description of iZQC signal evolution. A variety of iZQC spectroscopic techniques have been established and most of them take advantage of two-dimensional acquisition to recover a one-dimensional high-resolution spectrum with information on chemical shifts, relative peak areas, J-coupling constants, and multiplet patterns. In this review, two comprehensive descriptions of an iZQC signal are presented first. The existing iZQC techniques are then systematically described. The details and underlying mechanisms of these techniques are discussed. Finally, the in vivo applications of iZQC spectroscopic techniques are given.

Abstract:Intermolecular multiple-quantum coherence (iMQC) is capable of improving NMR spectral resolution using a 2D shearing manipulation method. A pulse sequence termed CT-iDH, which combines intermolecular double-quantum filter (iDQF) with a modified constant-time (CT) scheme, is designed to achieve fast acquisition of high-resolution intermolecular zero-quantum coherences (iZQCs) and intermolecular double-quantum coherences (iDQCs) spectra without strong coupling artifacts. Furthermore, double-absorption lineshapes are first realized in 2D intermolecular multi-quantum coherences (iMQCs) spectra under inhomogeneous fields through a combination of iZQC and iDQC signals to double the resolution without loss of sensitivity. Theoretically the spectral linewidth can be further reduced by half compared to original iMQC high-resolution spectra. Several experiments were performed to test the feasibility of the new method and the improvements are evaluated quantitatively. The study suggests potential applications for in vivo spectroscopy.

Abstract:Intermolecular multiple-quantum coherences (iMQCs) have some intrinsic properties different from conventional single-quantum coherences in solution NMR. In this paper, we extended our study to heteronuclear iMQCs in IS (I=1/2, S=3/2) spin systems. A sample of sodium chloride (NaCl) water solution was taken as an example. Heteronuclear COSY revamped by asymmetric Z-gradient echo detection (CRAZED) experiments were performed. One- and two-dimensional heteronuclear iMQC spectra were obtained. The quantum-mechanical treatment was used to deduce the signal expressions. Magic angle experiments validate that the signals are indeed from intermolecular dipolar interaction and insensitive to the imperfection of radio-frequency (RF) flip angles. Both experimental results and theoretical analysis indicate that heteronuclear CRAZED experiment allows coherence transfer from spin-3/2 nuclei to spin-1/2, and vice verse. Furthermore, the dependences of iMQC signal intensities on RF pulse flip angles follow the same rules as those for heteronuclear IS (I=1/2, S=1/2 or 1) spin systems.

Abstract:This paper analyses the heteronuclear Cosy Revamped by Asymmetric Z-gradient Echo Detection pulse sequence. General theoretical expressions of the pulse sequence with arbitrary flip angles were derived by using dipolar field treatment and signals originating from heteronuclear intermolecular single-quantum coherences (iSQCs) in highly-polarized two spin-1/2 systems were mainly discussed in order to find the optimal flip angles. The results show that signals from heteronuclear iSQCs decay slower than those from intermolecular double-quantum coherences or intermolecular zero-quantum coherences. Magical angle experiments validate that the signals are from heteronuclear iSQCs and insensitive to the imperfection of radio-frequency flip angles. All experimental observations are in excellent agreement with theoretical predictions. The quantum-mechanical treatment leads to similar predictions to the dipolar field treatment.

Abstract:Based on the double-quantum-filtered technique, a three-pulse sequence was designed to effectively eliminate contamination of residual conventional single-quantum coherences (SQC) from intermolecular SQC signals in single-resonance spin-1/2 samples such as water. Analytical expressions were derived from the modified Bloch equations with dipolar fields. All experimental observations and numerical simulation results are in excellent agreement with the theoretical predictions. Compared to the CRAZED sequence, the sequence can effectively suppress residual conventional SQC signals even with great imperfection of radio-frequency pulse flip angles. For the first time, signals from pure intermolecular SQCs were selectively observed in forms of spectrum or image.

Abstract:High-resolution NMR spectroscopy is a powerful tool for analyzing molecular structures and compositions. Line-widths of conventional liquid NMR signals are directly proportional to the overall magnetic field inhomogeneity the sample experiences. In many circumstances, spatial and temporal homogeneity of the magnetic field is degraded. In this paper, a modified pulse sequence based on intermolecular double-quantum coherences (iDQCs) was proposed to obtain 1D high-resolution NMR spectra under inhomogeneous fields using 2D acquisition. Analytical expressions were derived from the intermolecular multiple-quantum coherence (iMQC) treatments. Both experimental and simulated spectra provide high-resolution 1D projection spectra similar to conventional 1D high-resolution spectra. Moreover, the apparent J coupling constants are threefold magnified, which allows a more accurate measurement of small J coupling constants.

Abstract:A heteronuclear CRAZED pulse sequence was utilized to obtain high-resolution NMR spectra via heteronuclear intermolecular zero-quantum coherences (iZQCs) or intermolecular double-quantum coherences (iDQCs) in inhomogeneous fields. It overcomes the difficulty of selective excitation in homonuclear CRAZED experiments when the chemical shifts of solvent and solute are close to each other. In the 1D spectrum extracted from the 2D data, chemical shifts, coupling constants, multiplet patterns, and relative peak areas are almost independent of the magnetic field inhomogeneity. Abstract:In three-dimensional magnetic resonance imaging,intermolecular multiple-quantum coherences(iMQC) come from distant-dipole field effects.In this paper,a fast,efficient numerical algorithm was employed to simulate dipolar field effects for heterogeneous three dimensional structures.When suitable imaging parameters were selected,images with unique contrast were obtained without using contrast agents.Simulated results demonstrated that the iMQC images have contrast fundamentally different from the conventional single-quantum coherence images.

Abstract:In highly polarized scalar-coupled liquid systems, nuclear magnetic resonance signals from multiple-quantum coherences (MQCs) may be formed by inter-molecular dipolar and/or intra-molecular scalar couplings. Selection of specific signals can simplify the spectra, which may help us understand the underlying physical mechanisms in complex coupled spin systems. In this paper, a pulse sequence with three selective radio-frequency (RF) pulses and phase cycling was designed for this purpose. For an IpSq (p, q=1,2,3,…) spin system, there are three kinds of MQC signals, which originate from intra-molecular I–S, inter-molecular I–S, and inter-molecular S–S (or I–I) coherences. These three kinds of signals can be detected separately by proper phase cycling of RF pulses, which is independent of coupling constants. The intra- and inter-molecular MQC signals can also be detected separately with specific preparation periods, but this method is sensitive to coupling constants. Our theoretical predictions are in good agreement with experimental observations. The method proposed herein can be extended to heteronuclear cases.

Abstract:Although the theories and potential applications of intermolecular multiple-quantum coherences (iMQCs) have been under active investigations for over a decade, discussion of iMQC NMR signal formation was mainly confined in the time domain. In this paper, a full line-shape theory was developed to describe iMQC signals in the frequency domain. Relevant features of the line shape, such as peak height, linewidth, and phase, were investigated in detail. Predictions based on the theory agree well with experimental and simulated results. Since radiation-damping effects always couple with iMQCs in highly polarized liquid-state NMR systems, and strongly radiation-damped signals have many spectral characteristics similar to those of iMQCs, a detailed comparison was also made between them from different spectral aspects. With detailed comparison of peak height, linewidth, and phase, this work demonstrates that the iMQC and radiation-damping phenomena result from two completely different physical mechanisms despite that both present similar signal features and coexist in highly polarized liquid-state NMR systems. Abstract:A new pulse sequence based on intermolecular double-quantum coherences was proposed to obtain one-dimensional high-resolution liquid NMR spectra in inhomogeneous magnetic fields via Hadamard encoding. In contrast with the conventional intermolecular multiple-quantum coherences method with a two-dimensional acquisition to obtain one one-dimensional high-resolution spectrum, the new method can provide relatively high-resolution spectra directly through one-dimensional acquisition, and can greatly improve the signal-to-noise ratio of the spectrum within a relatively short acquisition time. Theoretical derivation was performed and analytical expressions of the resulting signals are given. Solution samples in purposely de-shimmed magnetic fields and pig brain tissue samples were tested. The experimental results demonstrate that this sequence can retain useful structural information, even when the field inhomogeneity is sufficiently severe to erase almost all spectral information with conventional one-dimensional single-quantum coherence techniques, and good solvent suppression can be achieved. This method may provide a promising technique for applications in in vivo and in vitro NMR. Copyright (c) 2012 John Wiley & Sons, Ltd.

Abstract:An NMR pulse sequence was designed to accurately measure the diffusion rates of intermolecular zero-quantum coherences (ZQCs) and double-quantum coherences (DQCs) in two-component spin systems. The Gaussian phase distribution approximation was employed to describe the behavior of intermolecular ZQCs and DQCs as they underwent unrestricted isotropic diffusion. Our experimental and theoretical results demonstrate that intermolecular ZQCs and DQCs evolve with the same diffusion rates. Uncorrelated phases between individual spins on different molecules in intermolecular ZQCs and DQCs are suggested as the cause for their unconventional diffusion behavior. The differences in diffusion rates between inter- and intramolecular ZQCs or DQCs are explained.

Abstract:High-resolution nuclear magnetic resonance spectroscopy is widely used to analyze molecular structures and compositions. Here, we present a pulse sequence that simultaneously detects both intermolecular zero- (iZQC) and double-quantum coherence (iDQC) signals in inhomogeneous fields, offering high-resolution spectra in absolute-value mode after data processing. In comparison with pure iZQC or iDQC spectra, the proposed sequence can provide nearly doubled spectral resolutions and suppressions of strong coupling artifacts. It is the first time that iZQC and iDQC signals can be simultaneously obtained in one single measurement.
