Abstract:ObjectiveTo investigate the involvement of L-selectin and its ligand, peripheral lymph node addressin (PNAd) in human embryo implantation.DesignProspective study.Materials and methodsL-selectin expression was examined by immunofluorescence in 20 preimplantation embryos (morula, n=8; blastocyst, n=12) and 5 cases of chorion during the first trimester of pregnancy. A total of 76 cases of endometrium from normal menstrual cycle (12 from healthy women, in proliferative phase; 64 from sterile women, of whom, 28 in early secretory phase, 36 in mid-late secretory phase) and 13 cases of deciduas during the first trimester of pregnancy were analyzed for PNAd expression by immunohistochemistry and Western blot analysis to assessed its spatio-temporal expression pattern. Sixty-four sterile women who underwent in vitro fertilization and embryo transfer (IVF-ET) were divided into pregnant and nonpregnant group according to whether resulted in clinical pregnancy. Cycle parameters of IVF-ET and PNAd expression level were compared between these two groups. Parametric variables underwent Student t test and Pearson correlation analysis, and the categorical variables underwent chi-square test and Spearman correlation analysis. Relative protein levels of each molecule of PNAd between groups were evaluated by rank-sum test.ResultsTabled 1ConclusionL-selectin and its ligand, PNAd are involved in maternal-fetal interface adhesion during implantation.
