Abstract:Objective To investigate the effect of PSUPER-siRNA-IGFIR plasmid in inhibiting the growth of human liver cancer cell SMMC7721.Methods Cancer xenograft was established by transplanting SMMC7721 cell to nude mice,and the PSUPER-siRNA-IGFIR plasmid was constructed and the graft cancer was treated with PSUPER-siRNA-IGFIR plasmid.The effect on tumor growth was monitored.Results PSUPER-siRNA-IGFIR plasmid can inhibit the growth of human liver cancer cell SMMC7721.The expression of IGFIR and MVD in human liver cancer cell SMMC7721 was decreased significantly in the gene transfected group as compared to that of control group.Conclusions The PSUPER-siRNA-IGFIR plasmid can inhibit the growth of human transfected liver-cancer in nude mice.

Abstract:Objective To study the inhibitory effect of small interference RNA(siRNA) targeting insulin-like growth factor-1 receptor(IGF-1R) on the growth of glioma. Methods Transcription-polymerase chain reaction and Western blot were used to analyze the down-regulation of IGF-1R expression.The invasiveness of C_(6) cell in vitro was measured by the matrigel invasion assays. Flow cytometry was performed to analyze cell apoptosis.The ability of the tumorigenicity in the nude mice was measured. Results The expression of IGF-1R was suppressed effectively by siRNA targeting IGF-1R.The expression of IGF-1R mRNA and IGF-1R protein decreased by 60% and 50% respectively.Cell apoptosis was induced from 1.29% to 15.88%(P0.01).The ability of tumorigenicity in the nude mice was reduced. Conclusion Down-regulation of IGF-1R by siRNA can suppress glioma cell growth markedly.

Abstract:OBJECTIVE:To study the effects of RNA interference (RNAi)-mediated insulin-like growth factor I receptor (IGF-IR) gene silencing on human lung cancer cells. METHODS:Plasmids expressing IGF-IR shRNA1 and IGF-IR shRNA2 were constructed. Human non-small cell lung cancer cells of the line A549 were cultured and transfected with sequence-specific shRNA. RT-PCR was used to monitor the IGF-IR mRNA expression. Western blotting was used to detect the expression of IGF-IR, bcl-2 and caspase-3, associated with apoptosis, and IGF-IR signaling pathways-associated proteins, total and phospho-ERK1/2 and Akt. MTT assay and flow cytometry were used to examine the cell activity and cell cycle. Twelve nude mice were injected subcutaneously with A549 cells, 20 days later the mice were randomly divided into 3 groups to be injected into the tumor with IGF-IR, PBS, or blank plasmid respectively 4 times with the interval of 5 days. Five days after the 4th injection the mice were killed and the tumors were taken out. TUNNEL assay was used to detect the apoptotic cell in the tumor. RESULTS:RT-PCR showed that the IGF-IR mRNA expression level of the A549 cells transfected with IGF-IR shRNA1 was only 24% +/- 4% that of the A549 cells transfected with blank plasmid (P < 0.05); however, the IGF-IR mRNA expression level of the A549 cells transfected with IGF-IR shRNA2 was 78% +/- 5% that of the A549 cells transfected with blank plasmid (P > 0.05). The IGF-IR protein expression level of the A549 cells of the IGF-IR shRNA1 group was only 10.2% +/- 2.8% that of the A549 cells of the blank plasmid group (P < 0.05). Western blotting showed that the protein expression levels of bcl-2 and caspase-3p20 of the A549 cells of the IGF-IR shRNA1 group were 46% +/- 6% and 156% +/- 8% those of the negative controls (both P < 0.05); however, the protein expression levels of bcl-2 and caspase-3p20 of the A549 cells of the IGF-IR shRNA2 group were not different from those of the negative control cells. The Akt kinase and ERK phosphorylation levels of the A549 cells of the IGF-IR shRNA1 group were 10% and 36% +/- 3% those of the negative control cells respectively (both P < 0.05). Since 48h after the transfection the active cell number of the IGF-IR shRNA1 group was 64% +/- 7% that of the negative group (P < 0.05), and this decrease effect lasted to 72 h after (67% +/- 6% that of the negative cells, P < 0.05). 48 h after the transfection the percentage of cells at G(0)/G(1) phase of the IGF-IR shRNA1 group was 77.5%, significantly higher than that of the negative control group, and the percentages of the cells at S and G(2)/M phases of the IGF-IR shRNA1 group were 15.7% and 7.3% respectively, both significantly lower than those of the negative control group (23.0% and 29.9% respectively). Since the second injection the tumor size of the mice of IGF-IR shRNA group was 40% - 50% that of the PBS group (P < 0.05), and the tumor size of the mice of the PBS group was 90% that of the control group. TUNNEL assay showed that the number of apoptotic cells in the tumors of the IGF-IR shRNA1 group mice was 118 +/- 8/high power, significantly higher than that of the control group (70 +/- 9, P < 0.05). CONCLUSION:RNAi technique effectively inhibits the expression of IGF-IR, thus decreasing the NSCLC cell proliferation inducing apoptosis and inhibiting the tumor growth.

Abstract:Cancer invasion and metastasis, involving a variety of pathological processes and cytophysiological changes, contribute to the high mortality of lung cancer. The type 1 insulin-like growth factor receptor (IGF-1R), associated with cancer progression and invasion, is a potential anti-invasion and anti-metastasis target in lung cancer. To inhibit the invasive properties of lung cancer cells, we successfully down-regulated IGF-1R gene expression in A549 human lung cancer cells by small interfering RNA (siRNA) technology, and evaluated its effects on invasion-related gene expression, tumor cell in vitro invasion, and metastasis in xenograft nude mice. A549 cells transfected with a plasmid expressing hairpin siRNA for IGF-1R showed a significantly decreased IGF-1R expression at the mRNA level as well as the protein level. In biological assays, transfected A549 cells showed a significant reduction of cell-matrix adhesion, migration and invasion. Consistent with these results, we found that down-regulation of IGR-1R concomitantly accompanied by a large reduction in invasion-related gene expressions, including MMP-2, MMP-9, u-PA, and IGF-1R specific downstream p-Akt. Direct tail vein injections of plasmid expressing hairpin siRNA for IGF-1R significantly inhibited the formation of lung metastases in nude mice. Our results showed the therapeutic potential of siRNA as a method for gene therapy in inhibiting lung cancer invasion and metastasis.

Abstract:Type I insulin-like growth factor receptor (IGF-IR), which is frequently overexpressed in a variety of human cancers including lung cancer, mediates cancer cell proliferation and tumor growth. In this study, we used a human U6 promoter-driven DNA-template approach to induce hairpin RNA (hpRNA)-triggered RNAi to silence IGF-IR gene expression in the human lung cancer cell line A549, and then evaluate its effects on apoptosis, apoptosis-related gene expression, and the growth of tumor cells in vitro and in nude mice. IGF-IR expression levels were found to markedly decrease in cells transfected with a plasmid expressing hairpin siRNA for IGF-IR (by more than 78.9%). Down-regulation of IGR-IR concomitantly accompanied reduction of bcl-2 as well as pERK and pAkt levels, activation of caspase-3, apoptosis and growth inhibition of A549 cells in vitro. Direct intratumoral injections of plasmid DNA expressing hpRNA for IGF-IR significantly regressed pre-established tumors in nude mice. Our results support the therapeutic potential of RNAi as a method for gene therapy in treating lung cancer.
Experimental Study of Suppressing Effect of Sirna Targeting IGF-1 on Growth of Glioma in Vitro and in Vivo

[Inhibitory Effects of Iressa on Hepatocellular Carcinoma Hep-3B and HepG2 Cell Xenografts in Nude Mice].

Abstract:BACKGROUND & OBJECTIVE:Iressa, a new molecular targeting medicine, has some effects on advanced non-small cell lung cancer, but its effect on hepatocellular carcinoma (HCC) has seldom been reported. This study was to observe the efficacy and adverse effect of Iressa on HCC Hep-3B and HepG2 cell xenografts in nude mice.METHODS:The nude mice with Hep-3B and HepG2 cell xenografts were randomized into control group (8 mice), low-dose group (8 mice), and high-dose group (8 mice), and received daily oral gavage of 5% glucose solution, 100 mg/kg Iressa, and 200 mg/kg Iressa, respectively, by 5 days each week for 3 weeks. The mice were killed 2 days after treatment. Inhibitory rate (IR) of tumor growth was calculated. Tumor volume, mice weight, and spleen index (SI) were compared between groups.RESULTS:Body weight and tumor volume of the mice were caparable before treatment. IRs of Hep-3B cell xenograft growth were 32.77% in low-dose group and 46.99% in high-dose group; IRs of HepG2 cell xenograft growth were 68.57% and 75.24%, respectively. Body weight and SI of nude mice with either Hep-3B or HepG2 cell xenografts were significantly lower in high-dose group than in control group, but no obvious difference was observed between low-dose group and control group.CONCLUSION:Iressa has obvious inhibitory effects on growth of both Hep-3B and HepG2 cell xenografts in nude mice, but high-dose (200 mg/kg) of Iressa may cause body weight lose and immunologic inhibition.
[Knockdown of Cyclin A2 Expression by Small Interfering RNA in MG-63 Cells].

Abstract:OBJECTIVE:To study the inhibitory effect of small interference RNA (siRNA) targeting cyclin A2 gene on the growth of osteosarcoma MG-63 and human normal skin fibroblast HSF cells and to explore whether cyclin A2 siRNAs could become a useful tool in the treatment of osteosarcoma.METHODS:Three pairs of siRNAs targeting cyclin A2 mRNA and a pair of nonsense siRNA were designed according to the current criteria. SiRNAs were chemically synthesized and purified. The siRNAs were transfected into MG-63 cells and HSF cells via oligofectamine. The cells transfected with nonsense siRNA served as negative control group and those only treated with PBS as blank control group. Quantitative fluorescence RT-PCR, Western-blot, MTT assay, reverse transcriptase (RT)-PCR, flow cytometry and clone forming test were employed to evaluate the efficacy of RNA interference. At the same time, the mRNA expression of PCNA and cyclin B1 in siRNA-treated MG-63 cells were examined.RESULTS:Although all three siRNAs could reduce the cyclin A2 expression, siRNA, appeared to be the most effective. After 48 h treatment with siRNA1, cyclin A2 mRNA and protein expression in MG-63 cells was significantly reduced by nearly 80% as compared with that of the blank control group, whereas the negative and blank control groups had similar expression levels. MG-63 cells treated with siRNA1 were arrested at G0/G1 phase by 80.1% and the proliferation of these tumor cells was suppressed 48 h after transfection. Furthermore, MG-63 cells showed a decreased colony forming ability after siRNA1 treatment. In addition, the cyclin A2-depleted MG-63 cells showed decreased levels of PCNA and cyclin B1. In contrast, although cyclin A2 expression in HSF reduced by nearly 60% after treatment by siRNA1 for 48h, these cells exhibited only a slight change in cell cycling, and neither clear inhibition of proliferation nor impaired colony forming ability was observed.CONCLUSION:Cyclin A2 is critical for proliferation of MG-63 cells. Cyclin A2-siRNAs can induce obvious inhibition of cyclin A2 mRNA and protein expression in MG-63 and HSF cells, which consequently down-regulate the proliferation of MG-63 cells. There is little effect on the proliferation of siRNA-treated HSF cells. Those results indicate that siRNAs against cyclin A2 may become a potential antiproliferative tool in future antitumor therapy.
Influences of Over-Expression of VEGF-C and Its Inhibition on Biological Characteristics of NCI-H446 Human Small-Cell Lung Cancer Cells

Abstract:Objective: To investigate the influences of vascular endothelial growth factor C (VEGF-C) on cell proliferation, invasion and apoptosis of NCI-H446 human small-cell lung cancer (SCLC) cells in vitro. Methods: The plasmids for VEGF-C overexpression (pHRi-VEGF-C) and small RNA interference targeting VEGF-C (pHRi-siVEGF-C) were constructed. Then the NCI-H446 cells were infected with the lentiviral packaging plasmids pHRi (an empty vector), pHRi-VEGF-C and pHRi-siVEGF-C, respectively. The expression of VEGF-C was examined by Western blotting; the cell proliferation was detected by MTT method; the invasive ability was detected by Transwell assay; and the apoptosis rate was determined by flow cytometry (FCM). Results: The expression level of VEGF-C protein in the NCI-H446 cells was significantly increased after lentiviral packaging plasmid pHRi-VEGF-C infection while it was significantly decreased after lentiviral packaging plasmid pHRi-siVEGF-C infection. The cell proliferation was significantly increased after the NCI-H446 cells were infected with pHRi-VEGF-C for five days as compared with that of the NCI-H446 cells infected with pHRi-siVEGF-C or pHRi (1.481±0.056, 0.838±0.035 and 1.146±0.07; P < 0.05). The average number of invasive cells in each visual field was significantly reduced in the pHRi-siVEGF-C-infected NCI-H446 cells as compared with that of the pHRi-VEGF-C- or pHRi-infected NCI-H446 cells (39.78±0.77, 84.87±1.27 and 60.82±1.05; P < 0.01). The number of apoptotic cells was also reduced by 90% in the pHRi-VEGF-C-infected NCI-H446 cells as compared with that of the pHRi-infected NCI-H446 cells (P < 0.01). Conclusion: The overexpression of VEGF-C can obviously promote the proliferative and invasive abilities of NCI-H446 cells, and it also can reduce the apoptosis. Meanwhile, these effects can be inhibited by small RNA interference targeting VEGF-C. The results suggest that VEGF-C may become a target of gene treatment for small-cell lung cancer. [Growth Inhibition of MG-63 Cells by Cyclin A2 Gene-Specific Small Interfering RNA].

Abstract:OBJECTIVE:To study the impact of small interference RNA (siRNA) targeting cyclin A2 gene on the growth of MG-63 and HSF cells and to explore whether cyclin A2 siRNAs could become a useful tool in the treatment of osteosarcoma.METHODS:One pair of siRNA targeting the cyclin A2 mRNA and a pair of nonsense siRNA were designed according the current criteria. SiRNA was chemically synthesized and purified. The siRNA was transfected into osteosarcoma cell line MG-63 and normal human skin fibroblast (HSF) cells via oligofectamine. Cells transfected with nonsense siRNA served as the negative control and those only treated with PBS as the blank control group. Quantitative fluorescence RT-PCR, Western-blot, MTT assay, reverse transcriptase (RT)-PCR, flow cytometry and colony-forming test were employed to evaluate the efficacy of RNA interference. At the same time, the mRNA expression of PCNA and cyclin B1 in siRNA treated MG-63 cells were examined.RESULTS:1 nmol/L, 10 nmol/L, 50 nmol/L and 100 nmol/L cyclin A2-siRNA can reduced cyclin A2 mRNA and protein expression respectively by 9.43%, 56.35%, 79.17% and 84.30% as compared with that of the blank control group, whereas the negative and blank control groups had similar expression levels. After 48 h treatment with 10 nmol/L siRNA, MG-63 cells were arrested in G0/G1 phase and the proliferation of this tumor cell was suppressed by 39.06% 48 h after transfection. Furthermore, the treated MG-63 cells showed less colony-forming ability. Increasing the siRNA concentration to 50 nmol/L can further inhibit the proliferation of MG-63 cells by 54.94%. In addition, the cyclin A2-depleted MG-63 cells showed decreased levels of PCNA and cyclin B1. In contrast, although cyclin A2 mRNA and protein expression in HSF reduced 58.13% 48 h after treatment by 50 nmol/L siRNA, these cells exhibited only a slight change in cell cycle, and no clear inhibition of proliferation and impaired plate colony-forming ability was observed.CONCLUSION:Cyclin A2 gene maybe served as a potential target for tumor therapy. RNA interference induces obvious inhibition of cyclin A2 mRNA and protein expression in MG-63 and HSF cells, which consequently downregulate the proliferation of MG-63 cells. There is few inhibitory effect on the proliferation by siRNAs for HSF cells. These results indicate that siRNAs against cyclin A2 could become a potential antiproliferative tool in future antitumor therapy.
Hvegf Sirna Inhibits Initial Growth of A549 Tumor Cells in Nude Mice and Angiogenesis on CAM Assay

Abstract:OBJECTIVE:To investigate the effect of small interfering RNA (siRNA) targeting human vascular endothelial growth factor (hVEGF) on A549 cell growth in nude mice and angiogenesis on chorioallantoic membrane (CAM) assay.METHODS:Three pairs of hVEGF siRNA-plasmid and non-silencing-plasmid were constructed, and transfected into A549 cells through lipofectamine 2000, respectively. The most effective pair of hVEGF siRNA-plasmid was selected by ELISA and real-time RT-PCR. A549 cells transfected with selected hVEGF siRNA- plasmid, A549 cells transfected with non-silencing-plasmid and A549 cells without transfection were inoculated into nude mice, respectively. Chick embryos were randomly divided into four groups and CAM was treated by different solutions for 48 h: culture media DMEM as negative control group,un-transfected A549 cell culture supernatants as positive control group, hVEGF siRNA A549 cell culture supernatants as hVEGF siRNA group and nonsilencing siRNA A549 cell culture supernatants as non-silencing siRNA group. The CAMs were harvested on d12 for microscopic assays.RESULT:Compared with control group, hVEGF siRNA-plasmid induced 48% reduction in hVEGF secretion by A549 cells accompanied by 70% reduction in hVEGF mRNA. Compared with non-silencing siRNA group, the mean tumor volume of murine xenograft was reduced by 58% in hVEGF siRNA group; time for xenografts growing to 50 mm(3)was delayed by 5.4 d. hVEGF contents in xenograft were reduced by 54%; but mean doubling time of tumors and the growth rate of tumors were not significantly reduced. In CAM assays, hVEGF content was zero in negative group, and in hVEGF siRNA group that was 40%-44% of non-silencing siRNA group or positive group; vessels branch points of CAM in hVEGF siRNA group or non-silencing siRNA group or positive group were increased by 45%-55% compared with negative group; total vessel length of CAM in hVEGF siRNA group was increased by 53% compared with negative group, while in non-silencing siRNA group or positive group that was increased by 97% or 99%. Compared with negative control group, the proliferation of microvessels was increased when cell culture supernatant with hVEGF was added in hVEGF siRNA group, significant proliferated vessels were observed in non-silencing siRNA group or positive group.CONCLUSION:A plasmid-mediated hVEGF siRNA has been constructed and verified, which can effectively downregulate the expression of hVEGF in human A549 cells, resulting in the inhibition of angiogenesis. hVEGF siRNA can delay initial growth of A549 tumor xenograft but not reduce the growth rate.
Experimental Research on the Inhibitory Effect of IFN-Lambda 3 Combined with Sorafenib on the Growth of Liver Cancer Transplanted into Nude Mice

Abstract:Objective: To investigate the effect of sorafenib combined with interferon-lambda 3 on the growth of liver cancer transplanted into nude mice. Methods: Female nude mice of 4-5 weeks of age that passed quarantine were selected and fed for 1-2 weeks before experimental operation. The cell suspension of human hepatoma cell line SMMC-7721 was inoculated into the right cervical axillary fossa with a syringe. The tumor-bearing mice were randomly divided into a control group and an experimental group. The control group received normal saline whereas the experimental group was further divided into three other groups: IFN-lambda 3 treatment group, sorafenib treatment group, and IFN-lambda 3 combined with sorafenib treatment group. The situation of nude mice was analyzed. At the end of the experiment, the volume of allogeneic transplanted tumor was measured, and the morphology of tumor cells, the expression of proliferating protein Ki-67, as well as the number of apoptotic cells were observed by hematoxylin and eosin (H&E) staining, immunohistochemistry (IHC), and TUNEL staining. Results: The tumor cell volume of the IFN-lambda 3 treatment group, sorafenib treatment group, and IFN-lambda 3 combined with sorafenib treatment group decreased, which was statistically significant compared with the control group (p < 0.05). The increment rate of proliferating protein Ki-67 in the transplanted tumor tissue of the three drug groups was significantly lower than that of the control group (p < 0.05). IFN-lambda 3 combined with sorafenib had the greatest effect on the expression level of Ki-67 protein. Compared with the control group, the expression rate was significantly lower (p < 0.05). In terms of cell apoptosis, IFN-lambda 3 and/or sorafenib, as well as the combination of the two, showed statistically significant differences compared with the control group (p < 0.05). The rate of cell apoptosis was the highest in the IFN-lambda 3 combined with sorafenib group. Conclusion: IFN-lambda 3 combined with sorafenib can inhibit the growth and proliferation of human hepatocellular carcinoma cells in nude mice and promote the apoptosis of hepatocellular carcinoma cells, which proves that IFN-lambda 3 combined with sorafenib can treat hepatocellular carcinoma in vivo.
Effect on Growth of Human Hepatoma Cells by Antisense Gene of Type II Insulin-Like Growth Factor Receptor Transfection

Abstract:Objective To study the growth inhibition of human hepatoma cells tranfected with type Ⅱ insulin like growth factor receptor (IGF ⅡR) antisense gene. Methods We constructed the recombinant eucaryotic expression plasmids of IGF ⅡR sense and antisense genes, which were transfected into SMMC 7721 human hepatoma cell line using calcium phosphate coprecipitation, and selected in DMEM supplemented with 500 μg/ml G418(Geneticin) for 2～3 weeks. Then the total number of colonies formed was counted. The effect of IGF ⅡR antisense gene transfection on endogenous IGF ⅡR mRNA levels was examined by Northern blot, and the growth rate and the ability of the transfected cells to form colonies in soft agar medium were also examined. Results The hepatoma cells transfected with IGF ⅡR antisense gene exhibited significant reduction in endogenous IGF ⅡR messenger RNA(mRNA) levels and loss of their ability for anchorage independent growth in soft agar. The IGF ⅡR antisense gene supressed the formation of colonies of SMMC 7721 cells resistant to G418 without alteration on cell growth curve. Conclusion The IGF ⅡR antisense gene expression effectively blocks IGF Ⅱ to IGF ⅡR autocrine and/or paracrine signal transduction pathway and reverses certain aspects of the cell malignant phenotype.
Effects of IGF1R Sirna on Chemosensitivity to Gemcitabine in Human Pancreatic Cancer Cells

Abstract:Objective To construct the lentiviral expression vector for RNA interference of insulin like growth factor-1 receptor (IGF1R) gene in human pancreatic cancer cells and to evaluate its effect on chemosensitivity to gemcitabine.Methods Three sequences of RNA interference targeting IGF1R gene were designed, synthesized and cloned into the pFU-GW-RNAi vector. After transfection of 293T cells with lentiviral vector, the lentivirus was produced and the titer of virus was tested. Panc-1 cells were infected with the lentivirus and the expression of IGF1R mRNA in Panc-1 cells was detected by real-time quantitative polymerase chain reaction (PCR). The changes of gemcitabine sensitivity after RNA interference were examined by methyl thiazol tetrazolium (MTT) assay. In vivo nude mice tumorigenicity model was used to determine the effect of RNA interference targeting IGF1R gene combined with or without gemcitabine on growth inhibition of xenograft tumors.Results A recombinant lentiviral vector expressing shRNA against IGF1R gene was obtained. After RNA interference, the median inhibition concentration (IC50) of gemcitabine against Panc-1 cells was reduced from (0.774±0.001) mg/L to (0.330±0.003) mg/L, indicating that the sensitivity to gemcitabine was increased by 2.35 times after interference. In vivo xenograft formation assay resulted in a significant growth-inhibiting effect by IGF1R RNA interference. Furthermore, the RNA interference targeting IGF1R gene enhanced the antiproliferative effect of gemcitabine on nude mice xenografts.Conclusion The sensitivity of Panc-1 cells to gemcitabine could be enhanced by RNA interference targeting IGF1R gene.

Abstract:Objective To investigate the effect of IGF1R gene silencing by SiRNA on the proliferation and apoptosis in human adrenocortical carcinoma cell line SW13 .Methods The specific SiRNA oligos for human IGF1R were synthesized , and transfected into SW13 cells by lipofectamine 2000 .The expression of IGF1R mRNA was detected by Realtime PCR after being incubated for 24 h .Western blot was used to detect the IGF1R protein .The proliferation and apoptosis of SW13 cells were evaluated by MTT and flow cytometry respectively .Results The inhibition efficiency of SiRNA for IGF 1R in the mR‐NA level was high and the same result was observed in the protein level by Western blot .MTT and flow cytometry showed that IGF1R gene silencing might lead to the inhibition of cell growth and enhancement of cell apoptosis .Conclusion IGF1R gene silencing of SW13 cells may induce apoptosis and inhibit proliferation of these cells .

Abstract:Emerging evidence confirms that insulin-like growth factor -II (IGF-II), oncogenes C-myc and N-ras are an essential regulator for development and growth in hepatocellular carcinoma (HCC). Although our previous study also indicated that IGF-II might upregulate levels of oncogenes C-myc and N-ras in hepatoma carcinoma cells, the molecular mechanism had not been fully elucidated. Herein, we successfully silenced IGF-II expression in SMCC-7721 cells by small RNA interference. Functional analysis showed that knockdown of IGF-II significantly suppressed growth and proliferation of SMMC-7721 cells and decreased C-myc and N-ras mRNA and protein levels. And this function was mediated by the FAK/PI3K/Akt signaling pathway. Taken together, IGF-II siRNA inactivates the FAK/PI3K/Akt signaling pathway, and further reduces cell proliferation, N-ras and C-myc levels in SMMC-7721 cells. Especially, understanding the relationship between IGF-II and oncogenes N-ras and C-myc in cancer cells will provide novel clues for clinic HCC treatment in the future.

Abstract:OBJECTIVE:To investigate the effect of recombinant adenovirus (phosphatidylinositol-3-kinases(PI3K)(I()-RNAi-AD which blocks the class I( PI3K signaling pathway on gastric carcinoma cells xenografts in nude mice.METHODS:Subcutaneous tumor models of nude mice were established with SGC7901 cells and randomly divided into PI3K(I()-RNAi-AD group, NC-RNAi-GFP-AD group and control group. The tumor size and the inhibitory rate of tumor growth on days 3, 6, and 9 after cell transplantation were measured. The expression of TNF-α, COX2, P53, PCNA, E-cadherin and nm23/DNPK in tumor tissues were detected by immunohistochemistry.RESULTS:Tumor growth was significantly inhibited in the PI3K(I()-RNAi-AD group(14.2%, 21.0%, and 28.1%) on days 3, 6, 9 compared with NC-RNAi-GFP-AD group(1.3%, 1.9%, and 2.0%, all P<0.05). The expressions of TNF-α, P53, E-cadherin and nm23/DNPK were up-regulated, and the expressions of COX2 and PCNA were down-regulated in the PI3K(I()-RNAi-AD group by immunohistochemical staining(all P<0.05).CONCLUSIONS:PI3K(I()-RNAi-AD can inhibit the growth of SGC7901 cell transplantation tumor in vivo in nude mice by inhibiting cell growth, reducing the capacity of tumor invasion and inhibiting tumor angiogenesis.

Abstract:Objective To investigate the inhibitory effect of lentivirus-mediated Bmi1-shRNA on transplanted human non-small cell lung cancer(NSCLC)by intratumoral injection.MethodsRecombinant lentivirus containing Bmi1-shRNA was constructed.A model of subcutaneous implanted tumor was generated through injecting human lung cancer cell A549(107)into the NOD/SCID mice.The 27 established mice with a mass at a diameter of 4 to 5 mm were divided into 3 groups randomly and equally.Recombinant lentivirus containing Bmi1-shRNA(5×108 TU/ml,0.1 ml),lentivirus containing NC-shRNA(5×108 TU/ml,0.1 ml,as negative controls)and PBS(0.1 ml,as blank control)was given the mice of the 3 groups respectively,once per 3 d,for 4 weeks.The size of the tumor mass were measured every 3 d.The mice were sacrificed in the end of 4 weeks' treatment.The expression of GFP in the xenografts was detected through laser scanning confocal microscope.RT-PCR and Western blot analysis were used to test the mRNA and protein expressions of Bmi1 in the xenografts.Results GFP was expressed in the tumor mass of the recombinant lentivirus groups,but not in the blank control.The average tumor volume in Bmi1-shRNA lentivirus group was significantly lower than that in the negative control and blank control groups(P<0.05)with an inhibitory rate of 52.9 % and 57.9% respectively.RT-PCR and Western blotting showed that the mRNA and protein expression of Bmi1 was decreased in the Bmi1-shRNA lentivirus group(P<0.05).ConclusionIntratumoral injection of recombinant Bmi1-shRNA Lentivirus inhibits the growth and proliferation of A549 xenografts in the NOD/SCID mice,suggesting that Bmi1 might represent a therapeutic target in the genotherapies of non-small cell lung cancer.

Abstract:Liver cancer is one of the most common malignant cancers worldwide. Systemic chemotherapy remains the major treatment option, but with severe adverse effects. Combinations of systemic with targeted treatments may provide effective therapeutics. The objectives of this study were to demonstrate if insulin-like growth factor-I receptor (IGF-IR) might serve as a functional target for liver cancer treatment and to investigate the chemo-sensitizing activity of IGF-IR downregulation. IGF-IR knockdown was achieved by stable transfection of liver cancer cells with IGF-IR small interfering RNA (siRNA). IGF-IR knockdown resulted in reduced growth, clonogenic survival, adhesion and migration of liver cancer cells, and increased sensitivities of liver cancer cells to apoptosis-inducing agents and chemotherapeutic drugs in vitro. In the animal studies, both IGF-IR knockdown and adriamycin (ADM) treatment significantly reduced the growth of liver tumors. IGF-IR knockdown enhanced the effect of ADM on tumor growth by further reducing tumor angiogenesis and inducing tumor cell apoptosis. The final tumor sizes in the IGFIR-siRNA, ADM-treated EGFP, and ADM-treated IGFIR-siRNA groups were significantly reduced by 52.5%, 33.8%, and 86.3%, respectively, compared with that in the EGFP control, suggesting that the ADM and the IGF-IR knockdown inhibit the growth of liver tumors in a synergistic manner. These results support that IGF-IR may serve as a functional molecular target for liver cancer treatment, and that the combination of systemic chemotherapy with targeted IGF-IR suppression may provide an effective treatment strategy for liver cancer.
Suppression of Insulin-Like Growth Factor-1 Receptor by RNA Interference Inhibits Cell Growth in Vitro and Induces Chemosensitization of HO8910PM Cell to Cisplatin

Abstract:OBJECTIVE:To assess the effect of suppression of insulin-like growth factor-1 receptor (IGF1R) in HO8910PM cell line by small interference RNA (siRNA).METHODS:Transfection of siRNA using lipofectamine 2000 was conducted to silence IGF1R gene expression, the expression levels of IGF1R mRNA and protein were evaluated, and the effects on the cell cycles at 48 hours of transfection were assessed by real-time PCR, western blot and flow cytometry (FCM) assay respectively. The cell growth was detected by cell counting kit-8 (CCK-8) at 24, 48, 72, 96 hours of transfection. After 24 hours of transfection, the cells were cultured with different concentrations of cisplatin (DDP) for 24 hours, the cell growth inhibition rate was evaluated by CCK-8. Following incubation with 10 microg/ml DDP for 24 hours after 24 hours of transfection, the apoptosis cells and the protein expression level of apoptosis-related gene, B cell leukemia/lymphoma 2 (Bcl-2), were identified by FCM and western blot respectively.RESULTS:(1) Expression levels of IGF1R mRNA and protein were markedly decreased respectively at 48 hours of transfection IGF1R siRNA. (2) Suppression of IGF1R accompanied the reduction of cell growth at 48, 72, 96 hours of transfection with IGF1R siRNA, absorbance were 1.71+/-0.13, 2.32+/-0.23, 2.79+/-0.28 respectively (P<0.01). (3) IGF1R siRNA induces arrest of G2 phase, the G2 phase rate of cells were 24.37% (P<0.05). (4) Following treatment with 2.5, 5, 10, 20 microg/ml DDP for 24 hours after 24 hours of transfection, the cell growth inhibition rates were (25.94+/-0.08)%, (40.25+/-0.05)%, (59.48+/-0.03)% and (74.18+/-0.08)% respectively (P<0.01). (5) Treatment with 10 microg/ml DDP for 24 hours after 24 hours of transfection, induces 17.95% of cells apoptosis (P<0.05), and decreases Bcl-2 protein level.CONCLUSION:RNA interference of IGF1R gene induces the IGF1R silence in HO8910PM cell line significantly, inhibits cell growth in vitro, arrests the G2 phase, and enhances the chemosensitization to DDP.
