Study on a Method of Microwave Dilapidation/hydrolyzation-fluorescamine Derivatization for Fast Detection of Total Bacterial Count in Specific Water System

Abstract:A chemical method for fast detection of the total bacterial count in a specific water system has been investigated.Microwave radiation had been applied to break down the cell walls of bacteria and assist the hydrolyzation of the proteins to amino acids.The amino acids produced were derivated by fluorescamine(FLA) for the fluorescence determination.The fluorescence intensity of the derivatives was measured for the calculation of the total bacterial count.The optimal experimental conditions for microwave dilapidating and hydrolyzing were as follows:4 mol/L HCl in E.coli sample,400 W microwave for 10 min.The optimal experimental conditions for derivating were:pH:2.0~2.2,1.0 mL 10-3 mol/L FLA solution in 3 mL sample(molecular ratio of FLA to amino acids higher than 1301),and reaction temperature:4℃.The relative standard deviation of the proposed method was 3.64%(n=6).For an E.coli sample,the determination result with the proposed method was in good agreement with that of standard method.The proposed method could be further developed as the fast detection for total bacterial count from samples of a specific water system.
