AhR Activation Stimulates Chemokine Production in an in Vitro Model of Endometriosis

Abstract:Objective: The industrial contaminant TCDD (tetrachorodibenzo-p-dioxin) is associated with inflammatory disorders of the reproductive tract in women and other mammals. The current studies were performed to investigate the effect of TCDD on RANTES (Regulated upon Activation, Normal T cell Expressed and Secreted) expression in an in vitro model of endometriosis. Design: Controlled laboratory study. Materials/Methods: Primary human endometrial stromal cells cultured in the absence and presence of 10 nM TCDD were lysed after 48 hours. The presence of AhR (Aryl hydrocarbon Receptor) was verified by western blotting and the receptors were localized in cell nuclei by immunocytochemistry. RANTES mRNA and protein were quantified by RT-PCR and ELISA, respectively. To quantify the transcriptional regulation by TCDD, human RANTES promoter-luciferase reporter constructs were transfected into endometrial stromal cells and luciferase activity was measured. The results were compared using non-parametric Wilcoxon tests. Two tailed tests with P < 0.05 were considered significant. Results: Semi-quantitative RT-PCR and ELISA indicated an up-regulation of RANTES mRNA and protein in endometrial stromal cells incubated with TCDD for 2 days. Treatment with TCDD for 24 hours increased the luciferase activity by 2–4 fold (P < 0.05) in stromal cells of both normal (n = 2) and endometriosis patients (n = 4). When AhR were overexpressed in these cells, the luciferase activity increased 5–19 fold (P < 0.01). Control transfections using TNF-alpha and NF-kB response element reporters indicated that these pathways were not activated by TCDD in human endometrial stromal cells. Conclusions: We demonstrated for the first time that TCDD up-regulates the expression of RANTES, which may provide a possible link between dioxin exposure, macrophage recruitment and endometriosis. TCDD up-regulation of RANTES gene expression appears to be independent of TNF-alpha and NF-kB activation.
