Effects of different dosed butorphanol intrathecal pretreatment on praxiology in rat model of formalin-induced inflammatory pain

Abstract:Objectives To study effect of different dosed intrathecal pretreatment of butorphanol on rat model of formalin-induced inflammatory pain,in order to explore the mechanism of antinociceptic effect. Methods 15 SD rats were intrathecally pretreated with left palmaris paw subcutaneous formalin injection. The rats were then divided into 3 groups: low-dose of butorphanol (12.5μg),high-dose of butorphanol (25μg),and control group. The formalin-induced behavior of rats was recorded continuously for 60 min. The rats were sacrificed 1.5 h after the formalin injection,and the expression of NMDA receptors of the fifth lumbar spinal cord was determined by immunohistochemistry. Results High-dosed butorphanol inhibited both phases of formalin response and lowered the expression of spinal cord NMDA receptors,while the expressions of both the early and later phases were not significantly affected in the low-dose group and the control group. Conclusion The results provided evidence that intrathecal injection of butorphanol could produce a dose-dependent anti-nociception in rat model of formalin-induced inflammatory pain. The mechanisms of analgesia may be generated from butorphanol's restrain to the stimulation of NMDA receptors. However,further studies are required to explain whether the inhibition mechanism of NMDA receptors is associated with stimulating к or μ receptors by butorphanol.
