Expression and Clinical Significance of p53 and MDM2 in Gastric Adenocarcinoma

Abstract:Objective To investigate the expression and clinical significance of p53 and MDM2 in gastric adenocarcinoma.Methods The protein expressions of p53 and MDM2 in 94 gastric adenocarcinoma tissues were detected by immunohistochemical staining,and the relationship between the two proteins was analyzed.Results Mutant-p53 protein was detected in 36%(34/94) of primary gastric adenocarcinoma,and the percentage of p53-positive cells was highly variable ranging from 10% to 99%(median,30%).MDM2 protein was located in the cytoplasm and nucleus of gastric adenocarcinoma cells,and the positive rate of MDM2 expression was 62.8%(59/94).The expression level of MDM2 in wildtype-p53 gastric adenocarcinoma tissues(negative immunohistochemical staining) was higher than that in mutant-p53 tissues(positive immunohistochemical staining).Conclusion Variable expressions of p53 and MDM2 were detected in gastric adenocarcinoma tissues,and high expression level of MDM2 protein in part of gastric adenocarcinoma cells may be one of the main reasons that contributes to the limitation of wildtype-p53 accumulation and activation.
