Preparation of in Vitro Transcript and Transfection of Giardia Canis Trophozoites Free from Giardiavirus

Abstract:Objective To prepare the infective transcript in vitro of cDNA from Giardia canis virus(GCV).Methods According to the published nucleotide sequence of GCV,three pairs of specific overlapping primers were designed,including T7 RNA promotor in 5′-F1,and the infective cDNA cloning of GCV was constructed by RT-PCR,then the transcription reactions were performed using T7 RiboMAXTM Express Large Scale RNA Production System.The transcript of GCV was electroporated into the GCV-free strain of Giardia trophozoites.Results The GCV cDNA was prepared,whose nucleotide sequence was the same with the published of GCV,and GCV could be detected by extraction of total nucleic acid and observation of ultrastructure.Conclusion The successful transfection suggested that the transcript of GCV is infective,which provides the foundation for study on the character of GCV.
