Effect of Continuous Ultra-filtration in Series on Antioxidant Ability of Hydrolysates Derived from Big Head Carp Protein

Abstract:Hydrolysates derived from bighead carp protein(BHCP) were purified by continuous membrane ultra-filtration with molecular weight cut-off (MWCO) 10 kDa and 5 kDa and the scavenging ability of each fraction on free radical DPPH· were evaluated. The purified fraction was used to protect red blood cell (RBC), which was attacked by ·OH, and the microcosmic structures of RBC were observed by scanning electron microscopy (SEM). The result showed that about 56.7% of the hydrolysates was most composed of short peptide with molecular weight <5 kDa, which showed stronger antioxidant ability. After the purification by membrane ultrafiltration with MWCO 5 kDa, EC50 of hydrolysates on DPPH· was changed from 2.739 mg/mL to 1.556 mg/mL. The microcosmic structures showed that the aberration rate of RBC, which was protected by hydrolysates purified by ultrafiltration with MWCO 5 kDa (HU5kDa) was lower than that of RBC without any protection. It indicated that BHCP had the antioxidant ability. The chelating Fe2+ ability of all fractions of hydrolysates were evaluated and the antioxidant activities were related with the chelating ability.
