Abstract:Objective:To isolate and purify 10-deacetyl baccatinⅢ(10-DABⅢ)from leaves and branches of Taxus mairei.Methods:10-DABⅢwas extracted and purified by liquid-liquid and back ex- traction,column chromatography and recrystallization,and elucidated by UV and HPLC.Results:The total yield of 10-DABⅢ,a precursor of anti-cancer drug taxol and docetaxel was 7.9% and the purity of 10-DASⅢwas 91%.Conclusion:The novel isolation process for 10-DABⅢwas warranted for the pilot manufacture.

Abstract:One new oleanolic acid triterpenoid saponin, 3-O- β - d -glucopyranosyl olean-11, 13(18)-diene-23,28-dioic acid, (hereafter referred to as DS-1) was isolated from the traditional Chinese medicinal plant Dianthus superbus ( D. superbus ). DS-1 plays an important role in the bioactivity of D . superbus . Thus, a sensitive, reliable and accurate reversed-phased liquid chromatography with tandem mass spectrometry (LC–MS/MS) in negative ion mode was developed and validated for the quantification and pharmacokinetic study of DS-1 in rats plasma. The pharmacokinetic profile showed that DS-1 was rapidly absorbed and eliminated in plasma, indicating that significant accumulation of the compound in biological specimen is unlikely. In addition, poor absorption into systemic circulation was observed after oral administration of DS-1, resulting in low absolute bioavailability (0.92 %).

Abstract:To optimze the conditions for the extraction of ursolic acid from Hedyotis diffusa.Willd,extraction was studied by single factor and orthogonal experiment guided by the recovery of ursolic acid.Results showed that the successive order of factors affecting extraction was extraction time> extract times> alcohol concentration> solid-liquid ration.The optima conditions for extraction of ursolic acid were determined to be 95% alcohol at 85 ℃ in 1.5h for twice and solid to liquid ratio 1:8.

Abstract:OBJECTIVE The separation and purification of ursolic acid from Ilex kudingcha C.J.Tseng was studied in order to exploit its resource.METHODS The effect of different factors on leaching rate was studied by orthogonal experiment and variance analysis. Ursolic acid was obtained by extraction, precipitation and macro-porous resin adsorption. The samples were analyzed by HPLC. The mobile phase consisted of alcohol and water, and the eluate was collected, in which, ursolic acid was detected at 220 nm. RESULTS The optimal conditions were obtained.The leaching rate of ursolic acid was up to 1.44% by reflux extraction using 90% alcohol for 2.5 h twice at 1:14 of Ilex kudingcha C. J. Tseng mass to solvent volume. The optimal conditions of separation and purification of D8 resin included: the ursolic acid concentration was 1-2 mg·mL-1 and the solution was pH 6.0-7.0, eluate was pH 11.0. After adsorpted for an hour, resin was eluted successively with water, four times volume 30% alcohol,four times volume 90% alcohol in the flow rate of 2-3 mL·min-1. Ursolic acid product was acquired from 90% alcohol eluate.The purity of product was over 95% and the recovery was up to 97.82% . CONCLUSION The product was obtained with high purity and high recovery. It can be utilized in industrial production.
Purification of Eremophilane‐Type Sesquiterpenes from Ligularia Atroviolacea by High Speed Countercurrent Chromatography

Abstract:Following an initial cleanup step on the C-18 open column chromatography, a preparative high speed countercurrent chromatography (HSCCC) method for isolation and purification of 8 beta-H-eremophil-3,7(11)-dien-12,8 alpha; 15,6 alpha-diolide, and furanoeremophil-3-en-15,6 alpha-olide from the Chinese medicinal plant Ligularia atroviolacea was successfully established by a one-step separation, using n-hexane-ethyl acetate-ethanol-water (4: 1: 4: 1, v/v/v/v) as the two phase solvent system. The upper phase was used as the mobile phase in the head to tail elution mode. HPLC analysis of the fractions collected on the preparative HSCCC of 600 mg of crude extracts showed that the purity of 8 beta-H-eremophil-3,7(11)-dien-12,8 alpha; 15,6 alpha-diolide (54.7 mg) was 98.1% and that of furanoeremophil-3-en-15,6 alpha-olide (41.8 mg) was 98.4%. The chemical structures of the two eremophilane-type sesquiterpenes were identified by ESI-MS, H-1-NMR, and C-13-NMR analysis. To the best of our knowledge, 8 beta H-eremophil-3,7(11)-dien-12,8 alpha; 15,6 alpha-diolide was obtained from L. atroviolacea for the first time, while furanoeremophil-3-en-15,6 alpha-olide was first isolated as a natural product.

Abstract:Isolation and purification of triterpene acid from fruit of Gardenia Jasminodes Eills were studied,the optimum processing conditions are: the granularity of powder 0.4 mm, ratio of liquid to solid 8∶1, temperature 85 ℃, 3 times of extraction of 2.5h each, and concentration of alcohol 95 %.Extraction rate of ursolic lixiviate is 92.6 %. Structural identifications by DSC,IR, EI-MS ,~1H and ~ 13 C NMR,HPLC show that the triterpene acid is ursolic acid,the average content of ursolic acid is 0.444 5 %。

Abstract:Sodium hydroxide solution and sulfuric acid solution as precipitation solvents, the process of separation and purification of ursolic acid form Ilex kudingcha was studied. The optimal conditions were obtained as follows: firstly, the alcohol leaching solution was treated to pH 10.011.0 by sodium hydroxide solution with mass fraction 10%, then the polyphenols were removed by filtering, the filtrate was adjusted pH value to 4.0 by sulfuric acid solution with volume fraction 10%, and was diluted by acidic water solution (pH= 4.0), the crude product of ursolic acid was obtained; secondly, it was dissolved by methanol, and adsorbed in macro-porous adsorption resin column, then the resin was eluted fractionally by eluent of alcohol-water and the eluate was collected when pH value of the sample is 6.0(7.0) and pH value of the eluent is 11.0; finally, the product is obtained from the concentrate of 90% alcohol eluent. The products were determined by high performance liquid chromatography (HPLC). The results show that the purity of the product from precipitation separation and adsorption separation is 23.12% and 90.23%, respectively, the recovery is 92.90% and 98.02%, respectively. the total recovery is 89.09%.

Abstract:Imperatorin and isoimperatorin are important components of Angelica dahurica with many beneficial functions to human health. In this study, an ultrasound-assisted extraction technique has been developed for the fast extraction of imperatorin and isoimperatorin from Angelica dahurica root. The high extraction yield of imperatorin and isoimperatorin was obtained under optimum extraction conditions: ultrasonication time 20 min, extraction temperature 30°C, ultrasound power 240w, ethanol concentration 80%, and ratio of solvent volume to sample weight 12:1 (ml/g), particle size 60 meshes. Compared to heat-reflux extraction, ultrasound-assisted extraction reduced extraction time and obtained higher percentage extracted of imperatorin and isoimperatorin. Scanning electron micrographs revealed that ultrasound could result in the destruction of cell walls, which increased the mass transfer rate of the solvents into the root materials

Abstract:High performance liquid chromatography (HPLC) method was applied to determine the content of oleanolic acid (OA) and ursolic acid (UA) in different tissues of 'Ruantiaobaisha' and 'Dayeyangdun' loquat (Eriobotrya japonica Lindl.) fruits in this research. The results demonstrated that peel contained higher OA and UA contents, while flesh and kernel contained very lower amounts of OA and UA. On the basis of above results, the OA and UA contents in the peel of different developmental stages and different cultivars of loquat fruits were analyzed and compared. It is found that OA and UA contents in the peel varied with different developmental stages and cultivars. The OA and UA content in the peel of ripe fruit of different cultivars were in the range of 0.59-1.68 and 2.82-8.20 mg/g DW, respectively. These results can provide a theoretical basis for the comprehensive utilization of loquat fruit in the future.

Abstract:Bupleuri Radix (BR), as a well-known plant medicine of relieving exterior syndrome, has a long history of usage in China. Bupleuri Radix Polysaccharide (BRP), as the main component and an important bioactive substance of BR, has a variety of pharmacological activities, including immunoregulation, antioxidant, antitumor, anti-diabetic and anti-aging, etc. In this review, the advancements on extraction, purification, structure characteristics, biological activities and pharmaceutical application of BRP from different sources (Bupleurum chinense DC., Bupleurum scorzonerifolium Willd., Bupleurum falcatum L. and Bupleurum smithii Woiff. var. Parvifolium Shan et Y. Li.) are summarized. Meanwhile, this review makes an in-depth discussion on the shortcomings of the research on BRP, and new valuable insights for the future researches of BRP are proposed.

Abstract:Maslinic acid and corosolic acid with high purity were successfully separated from Eriobotrya japonica (Thunb.) leaves by two-step countercurrent chromatographic separation. Two biphasic solvent systems composed of petroleum ether-ethyl acetate-ethanol-water (6:4:5:5, v/v) and petroleum ether-ethyl acetate-ethanol-0.10 mol/L of hydroxypropyl-β-cyclodextrin with pH 7.0 (8:2:3.5:6.5, v/v) were selected according to the partition performance of the main structural isomeric pentacyclic triterpenes. The influences of pH value and concentration of hydroxypropyl-β-cyclodextrin in separation of two isomers were investigated. In first step countercurrent chromatographic separation, a mixture of two target structural isomers (14.12 mg of sample I) was separated from 40.00 mg of a partially purified sample. In second step countercurrent chromatographic separation, maslinic acid and corosolic acid were completely isolated from 12.00 mg of sample I with hydroxypropyl-β-cyclodextrin as aqueous phase additive. The recoveries of the two isomers were over 90%, yielding 5.18 mg of maslinic acid and 5.47 mg of corosolic acid, respectively.

Abstract:An efﬁcient method for the isolation and puriﬁcation of 12,13-dihydroxyeuparin from Radix Eupatorii Chinensis by high speed counter-current chromatography (HSCCC) was established in this paper. The ether extracts of Radix Eupatorii Chinensis were puriﬁed by HSCCC with a solvent system of hexyl hydride–ethyl acetate–methanol–water (1:2:1:2, v/v/v/v). The upper phase was used as the stationary phase and the lower phase as the mobile phase. About 8.4mg of 12,13-dihydroxyeuparin was obtained from 200mg of ether extracts from Radix Eupatorii Chinensis in one-step HSCCC separation, with the purity of 96.71%, as determined by HPLC. After methanol–water recrystallization, the purity of 12,13-dihydroxyeuparin reached 99.83%. Such a simple and effective method was fairly useful to prepare pure compound as reference substances for related study on Radix Eupatorii Chinensis.

Abstract:Barks and leaves of Eucommia ulmoides Oliver (EU) were successively fractionated by a multi-step treatment process. The extractum and water-soluble polysaccharides, alkali-extractable polysaccharides, and guttapercha, were sequentially obtained by hot-water extraction, alkaline treatment, and extended enzymatic hydrolysis, respectively. Results showed that the primary bioactive components of extractum from leaves were mainly consisted of chlorogenic acid, quercetin, geniposide and aucubin, and those from barks were consisted of geniposidic acid, geniposide and aucubin. In addition, the water-soluble polysaccharides from barks and leaves were mainly composed with glucose, whereas, the alkali-extractable polysaccharides from the barks and leaves were primarily the arabinose. Emphatically, the guttapercha fractions obtained from barks exhibited higher molecular weights with narrow polydispersity as compared to those from leaves. The data presented indicated that the multi-step process is an environmentally benign and advantageous scheme for value-added and potential industrial application of EU resources.

Abstract:A preparative counter-current chromatography (CCC) method for isolation and purification of oridonin, a new cancer chemoprevention agent, from the Chinese medicinal plant Rabdosia rubescens was successfully established. The crude oridonin was obtained by elution with a light petroleum/acetone solvent mixture from ethanol extracts of R. rubescens using column chromatography on silica gel. With a two-phase solvent system composed of n-hexane/ethyl acetate/methanol/water (1:2:1:2, v/v), 120 mg of oridonin at the purity of 97.8% was obtained from 200 mg of the crude sample in a single-step CCC separation. The structure of oridonin was identified by ESI-MS, 1H NMR, and 13C NMR.

Abstract:Peperomia pellucida is a member of the Piperaceae family. Extraction of plant material was carried out by Soxhlet extraction method using hexane and ethylacetate as solvents respectively. The extract was concentrated using a rotary evaporator, followed by isolation and purification using column and thin layer chromatographic techniques. Fraction C20 showed a clearly defined single spot with Rf value of 0.51. Using 1HNMR, 13C-DEPT, COSY, HSQC and HMBC and by comparison with literature values, the structure of the compound was established as an Ursane-type triterpenoid. The use of P. pellucida in ethnomedicine for the treatment of various ailments could be attributed to the Ursane-type triterpenoid and other bioactive chemical compounds present in the plant.
