Abstract:PURPOSE: Owing to the pivotal role that the calcineurin signalling pathway plays in the regulation of important cardiac and skeletal muscle phenotypes, e.g. fast-to-slow fibre transition, we hypothesized that polymorphisms in calcineurin genes are associated with elite endurance athletic status. METHODS: We compared genotypic/allelic frequencies in 55 polymorphisms of the main five genes (PPP3CA, PPP3CB, PPP3CC, PPP3R1 and PPP3R) encoding the calcineurin protein subunits, between 123 elite runners, and 125 healthy controls. The SNPs were genotyped by a matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.Analysis was corrected for multiple comparisons. RESULTS: There were no gender differences in genotypic/allelic frequencies within each group for any of the studied polymorphisms. The minor allele of the rs3804358 polymorphism in the PPP3CA gene was overrepresented in athletes (17%) compared with controls (8%) (P=0.003), whereas the minor allele of the rs3763679 polymorphism in the PPP3CB gene was underrepresented in athletes (23%) compared with controls (37%) (P=0.001). CONCLUSIONS: Though more research is needed, e.g. in other ethnic groups, our findings suggest that some variants in calcineurin genes might be among the numerous potential genetic variant candidates whcih can help explaining, at least partly, human variations in endurance exercise performance status.
