Abstract:Objective To investigate the existence of side population cells,with the potency of stem cells,in human gallbladder carcinoma cell line GBC-SD.Methods SP cells were sorted from GBC-SD cells by fluorescence-activated cell sorting(FACS) and detected by FACS again few weeks later,to assay the differentiation of SP cells.The expression of ABCG2 in SP cells was detected by flow cytometry(FCM).Results SP cells were isolated from GBC-SD cells with a proportion of(0.64±0.08)% and could generate both SP and non-SP cells after culture for 3-5 weeks.However,the cultures initiated with non-SP cells contained only non-SP cells.The expression of ABCG2 in SP cells was(89.56±3.86)%,on the contrary,there was nearly no expression in non-SP cells [(1.32±0.49)%].Conclusion SP cells,cancer-stem like cells,exist in human gallbladder carcinoma GBC-SD cells with overexpression of ABCG2.

Abstract:The cancer stem cell (CSC) hypothesis proposes that CSCs, which can renew themselves proliferate infinitely, and escape chemotherapy, become the root of recurrence and metastasis. Previous studies have verified that side population (SP) cells, characterized by their ability to efflux lipophilic substrate Hoechst 33342, to share many characteristics of CSCs in multiplying solid tumors. The purpose of this study was to sort SP cells from a human gallbladder carcinoma cell line, SGC-996 and to preliminarily identify the biological characteristics of SP cells from the cell line. Using flow cytometry we effectively sorted SP cells from the cell line SGC-996. SP cells not only displayed higher proliferative, stronger clonal-generating, more migratory and more invasive capacities, but showed stronger resistance. Furthermore, our experiments demonstrated that SP cells were more tumorigenic than non-SP counterparts in vivo. Real-time PCR analysis and immunocytochemistry showed that the expression of ATP-binding cassette subfamily G member 2 (ABCG2) was significantly higher in SP cells. Hence, these results collectively suggest that SP cells are progenitor/stem-like cells and ABCG2 might be a candidate marker for SP cells in human gallbladder cancer.

Abstract:【Objective】To study the biological characteristics of side population(SP) cells sorted from hepatocellular carcinoma Bel-7402 cell line.【Methods】Fluorescence-activated cell sorter(FACS) was used to sort SP cells and non-SP(NSP) cells from Bel-7402 cell line.Groups were: SP group and NSP group.The colony formation and proliferation ability were compared between SP cells and NSP cells by plate colony and growth curve assay.ATP-binding cassette superfamily G member 2(ABCG2) and CD133 mRNA levels in SP and NSP cells were examined by RT-PCR.The apoptosis was analyzed by flow cytometry.The sphere rate was counted for SP cells and NSP cells in serum-free medium.The oncogenicity of the SP and NSP cells were analyzed by tumor formation in nonobese diabeti-c/severe combined immune-deficient(NOD/SCID) mice.【Results】(2.4 ± 0.0) % SP cells were sorted from the Bel-7402 cells by FACS.Growth curve assay showed that the proliferation ability of SP cells was higher than NSP cells(P 0.05).Colony formation efficiency was significantly different(P 0.01) between SP(20.5 ± 2.6) % and NSP(5.1 ± 0.9) %.The level of mRNA expression of ABCG2 and CD133 in SP cells were 3.03(P 0.01) and 1.98(P 0.01) times of that in NSP cells,respectively.The apoptotic rate of SP cells were(0.89 ± 0.23)%,which were lower than those of NSP cells(P 0.05).The sphere rate in SP cells was(14.1 ± 1.9)%,higher than that in NSP cells in serum-free medium(P 0.05).With cell numbers of 1 × 103,104,105,the SP cells can form subcutaneous transplantation in NOD/SCID mice,but the NSP cells can not form it(P 0.05,P 0.05,and P 0.05).【Conclusion】The SP cells sorted from Bel-7402 cell line may enrich tumor stem cells.

Abstract:OBJECTIVE:To isolate and characterize the side population cells (SP cells) in the lung adenocarcinomas cell line A549.METHODS:The protein expression of ABCG2 in human lung adenocarcinoma cell line A549 was detected by immunohistochemistry. SP and NSP cells in the cell line A549 were isolated by FACS, and their differentiation was analysed. ABCG2 expression in the two cell subsets was detected by RT-PCR. The cell growth curves, cell division indexes, cell cycles, plate clone formation tests, migration and invasion assays, chemotherapeutic susceptibility tests, tests of the intracellular drug levels, and the tumor cell implantation experiments on nude mice were applied to study the biological properties of the two cell subsets. The expression of ABCG2 in the transplanted tumor in nude mice was detected by immunohistochemistry and RT-PCR.RESULTS:The positive rate of ABCG2 expression in the A549 cells by immunohistochemistry was 2.13%. SP and NSP cells were isolated by FACS. The SP cells could produce both SP and NSP cells, while NSP cells only produced NSP cells. SP cells expressed ABCG2, but NSP cells did not. The proliferation and migration abilities of the two cell subsets were similar, but the invasion and tumorigenic ability of SP cells was significantly higher than that of NSP cells. The susceptibilities to DDP and its intracellular levels of the two cell subsets were similar, but the susceptibilities to 5-FU, VP16, NVB and GEM and their intracellular levels of NSP cells were significantly higher than those of the SP cells.CONCLUSIONS:SP cells in the human lung adenocarcinomas cell line A549 is enriched with tumor stem cells. An effective way to get lung adenocarcinomas stem cells is to isolate SP cells by FACS.
Side Population Cells in Human Gallbladder Cancer Cell Line GBC-SD Regulated by TGF-β-induced Epithelial-Mesenchymal Transition.

Abstract:Mounting evidence has shown that side population (SP) cells are enriched for cancer stem cells (CSCs) responsible for cancer malignancy. In this study, SP technology was used to isolate a small subpopulation of SP cells in human gallbladder cancer cell line GBC-SD, and SP cells which had superior potential for proliferation in vitro and tumorigenesis in vivo were identified. Importantly, the abundance of GBC-SD SP cells was increased by a transforming growth factor-β (TGF-β)-induced epithelial-mesenchymal transition (EMT), and this effect was accompanied with a strong up-regulation of ABCG2 mRNA expression, and a decreased sensitivity to mitoxantrone. SP cells were restored upon the removal of TGF-β and the reversion of the cells to an epithelial phenotype, and smad3-specific siRNA reduced SP abundance in response to TGF-β. In conclusion, TGF-β-induced EMT by smad-dependent signaling pathway promotes cancer development and anti-cancer drug resistant phenotype by augmenting the abundance of GBC-SD SP cells, and a better understanding of mechanisms involved in TGF-β-induced EMT may provide a novel strategy for preventing cancer progression.
Isolation of side population cells in human carcinoma of gallbladder SGC996 cells and culture in vitro

Abstract:Objective To isolate side population cells and non-side population cells from human carcinoma of gallbladder SGC996 cell line,and cultured the two subpopulations in vitro,and prehminarily verified its characteristics of stem cells.Methods Flow cytometry was used to isolate side population cells and main population cells and cultured the two subpopulations in serum-free culture solution with epidermal growth factor (EGF),basic fibroblast growth factor (bFGF) and LIF and in serum culture solution respectively.Multiplication capacity in vitro of the two subpopulations was observed in serum culture solution by methyl thiazol tetrazolium (MTT).Results The proportion of side population cells was 1.0％ by flow cytometry,by addition of verapamil,it was 0.1％,which was greatly decreased.Side population cells exhibited higher multiplication capacity and were able to form “tumor spheres” in serum-free culture solution;while main population cells showed lower multiplication capacity and even died in the same condition.The result by MTT showed that,in serum culture solution the spectrophotometry values of side population cells on the 5th day,6th day and 7th day were 1.26 ±0.15,1.73 ±0.18 and 2.21 ±0.17,while the main population cells 0.95 ±0.06,1.31 ±0.10 and 1.86 ±0.11.Each group was significantly different (t =3.30,3.54,3.03,P ＜ 0.05 ).Conclusion Human carcinoma of gallbladder SGC996 cell line contained a small subpopulation of cells that could form “tumor spheres” and exhibited higher multiplication capacity in vitro,which provided further theoretical foundation for gallbladder cancer stem cells.

Abstract:Objective To study the tumorigenetic characteristics of side population (SP) cells and their exsistence in gastric cancer cell lines and tissues.Methods Flow cytometry and the DNA-binding dye Hoechst 33342 staining were used to analyze sorted SP cells in 3 gastric cancer cell lines(SGC-7901,BCIC-823 and MKN28).SP cells and non-SP cells(main population cells)isolated from SGC-7901 cell line weresubcutaneously injected into 36 nude mice with 500,5000 or 50 000 cells per mouse,respectively.Thetumorigenity was observed 8 weeks after injection.The expression of ATP-binding cassette sub-family Gmember 2(ABcG2)mRNA in three gastric cell lines and its level in gastric cancer tissues were detected by real-time PCR and immunostaining.respectively.Results The proportion of SP cells was accounted for 1.0%in SGC-7901 cells and 1.3%in BGc-823 cells.No SP cell was found in MKN28 cells.As low as 500 SP cells or 50 000 non-SP cells could initiate tumors in mice.The expression of ABcG2 mRNA was higher in SGC-7901(O.162)and BGC-823(O.096)cell lines than that in MKN28 cell line(0.005).The value of AtCG2 mRNA/beta-aetin mRNA and protein in gastric cancer tissues was different from those in gastritis tissues.Condasimm SP cells have strong ability of tumorigenesis compared with non-SP cells.The expression of ABCG2 is found in gastric cancer tissues and part gastritis tissues.The more the SP cells in gastric cell lines,the higher the expression of ABCG2.

Abstract:PURPOSE:To identify the side population (SP) cells from four hepatocellular carcinoma (HCC) cell lines with stepwise metastatic potentials.METHODS:SP cells were sorted from HCCLM3, MHCC97-H, MHCC97-L and Hep3B by flow cytometry, and then analyzed by differentiation study, clonogenic assay, chemoresistance study and tumorigenicity assay in vivo. The expression of ABCG(2) in SP cells was detected by immunocytochemistry, western blotting and real-time quantitative PCR, respectively.RESULTS:There was significant difference in SP proportion among HCCLM3, MHCC97-H, MHCC97-L and Hep3B (28.7 +/- 1.6%, 14.5 +/- 0.6%, 4.2 +/- 0.4%, 0.9 +/- 0.1%, respectively, P < 0.01). All the SP cells showed similar characteristics of self-renewal, high clonogenicity, remarkable chemo-resistance and high expression of ABCG(2). As low as 2,000 SP cells could initiate tumors in non-obese diabetic/severe combined immunodeficiency mice successfully.CONCLUSIONS:SP cells purified from HCC cell lines harbors cancer stem cell-like properties, and may be related to the metastatic potentials and therapeutic-resistance of HCC.

Abstract:AIM:To identify the side population(SP)of cancer stem-like cells in gastric cancer cell line MKN-28. METHODS:Immunofluorescence assay was used to determine the expression of ABCG2, an indicator of stem cells,in gastric cancer cell line MKN-28.MKN-28 tumor cells suspension was stained with Hoechst33342 and propidi- umiodide(PI)with the absence or presence of verapamil.Then the SP was analyzed by fluores- cence-activated cell sorter. RESULTS:A little amount of ABCG2 expression was found in MKN-28 cells,and the ABCG2- positive cells covered a percentage of 1.7%.SP cells accounted for 0.25% of the MKN-28 cells, which was decreased to 0.05% after the treat- ment of verapamil. CONCLUSION:SP cancer stem-like cells exist in gastric cancer cell line MKN-28,and they can be decreased by verapamil.

Abstract:CSCs (cancer stem cells) are a small subset of cells within a tumour that possesses the characteristics of stem cells and are considered to be responsible for resistance to chemoradiation. Identification of CSCs through stem cell characteristics might have relevant clinical implications. In this study, SP (side population) cells were sorted from a human ovarian cancer cell line by FACS to determine whether cancer stem cell-like SP cells were present. A very small fraction of SP cells (2.6%) was detected in A2780 cells. SP cells possessed the following characteristics: highly proliferative activity, marked ability for self-renewal in soft agar and culture medium, high expression of ABCG2, drug resistance to vinblastine in vitro, and strong tumourigenic potential in Balb/c nude mice. It is concluded that there exists in the A2780 cell line a small number of SP cells with high expression of ABCG2. The cells have the characteristics of cancer stem-like cells, and identification and cloning of such human SP cells can help in improving therapeutic approaches to ovarian cancer in patients.

Abstract:SummaryBackground: Recent studies have reported that the presence of cancer stem cells (CSCs) has major implications in the treatment of cancer and is responsible for tumor relapse. In the current study, we identified and characterized colon CSC-like side population (SP) cells from colon cancer tissue. Materials and Methods: The colon cancer samples were subjected to fluorescence-activated cell sorting (FACS) based on Hoechst 33342 dye exclusion for the purification of SP cells. The sorted SP cells were subjected to further characterization by immunofluorescence, real-time polymerase chain reaction (RT-PCR), multidrug resistance, and sphere formation assays. Results: We identified a fraction of 3.3% of SP cells in the colon cancer cell samples, which was reduced to 0.6% upon treatment with verapamil. The sorted SP cells showed high positivity with regard to CD133, CD44, CD147, and EpCAM, by fluorescence microscopic analysis. Further, these SP cells were highly resistant to multidrug treatment due to overexpression of the multidrug resistance 1 (MDR1) transporter protein ABCG2. Conclusion: Our findings suggest that the overexpression of ABCG2 and the expression of stem cell surface markers are collectively responsible for chemotherapy failure, tumor recurrence, and invasion in colon cancer.

Abstract:Objective Our aim is to sort side population cells from human hepatocellular carcinoma cell line 97H and to analysis the characteristic of morphology,phenotype and function of SP.Methods Microstructure and uhrastructure of SP cells were assayed by phase microscope and transmission electron microscopy,respectively,followed by sorting from MHCC97H using Flow Cytometry.The expression of ABCG2 in SP cells was assayed by Western Blotting.CD133 positive cells and CD117 positive cells in SP cells were investigated by Flow Cytometry.Colony-forming assay was used to measure the ability of SP cell growth and Non-obese diabetic/severe combined immunodeficiency (NOD/SCID)xenograft transplant experiments was performed to assay the ability of initiating tumor for SP cell.Results SP cells had round or short shuttle-like shape and were smaller than nonSP in volume.The numbers of organelle in SP cells were less than that in nonSP.Western blotting demonstrated that the expression of ABCG2 in SP cells was similar to that in nonSP cells.Flow Cytometer analysis showed that CD133 and CD117 positive cells were present in SP cells.Colony-forming assay showed that SP cells had higher clonogenicity.NOD/SCID xenograft transplant experiments demon strated that 2000 SP cells were sufficient to form tumor.Conclusion SP cells sorted from MHCC97H have characteristic of cancer stem cell-like phenotype and function.ABCG2 was not a molecular determinant of the SP phenotype in HCC.

Abstract:Active drug efflux by the adenosine triphosphate-binding cassette (ABC) transporter ABCG2 is one of the common mechanisms causing multiple drug resistance in various human cancers. In the intrinsic drug resistance of hepatocellular carcinoma (HCC), the role of ABCG2 is closely associated with 'side population (SP)', a minor subset of cancer stem-like cells with unique capacity to extrude lipophilic dye Hoechst 33342 and many chemotherapeutic agents. In this study, we showed that ABCG2 was intrinsically expressed in a subgroup of HCC tissues and its expression pattern significantly influenced the levels of drug efflux from HCC cell lines. In MHCC-97L HCC cell line with intrinsic ABCG2 expression, we confirmed the importance of SP cells to the drug efflux-related chemotherapy resistance and found that the SP analysis provided an efficient method to evaluate the functional activity of ABCG2 transporter. In this cell line, we discovered that the SP proportion was modulated by the treatments of Akt signaling inhibitors and serum supplement, which led to the finding that Akt signaling was able to regulate the SP cells' efflux activity via altering the subcellular localization of ABCG2 transporter. We further demonstrated that the Akt signaling inhibition attenuated the doxorubicin efflux from MHCC-97L cells and increased the drug efficacy. Our results indicate the protective role of intrinsic ABCG2 expression in HCC cells and suggest that suppressing Akt signaling could help overcome the drug efflux by ABCG2 transporter.
Identification of Side Population Cells Like Cancer Stem Cell in Human Pancreatic Cancer Cell Line SW1990

Abstract:Objective To isolate and identify side population (SP) cells like cancer stem cell from human pancreatic cancer cell line SW1990, for the purpose of further evaluation of their biological characteristics. Methods Cell suspension was stained with Hoechst 33342 and PI. Then SP cells were analyzed in the fluorescence activated cell sorter. Cell growth viability was measured by MTT. Stem cell marker CD133 was determined by flow cytometry. Cloning forming efficiency was determined by cloning plating. Expression of ABCG2 protein was detected by Western blot analysis. Results The proportion of SP cells was 2.7%, however it could be completely blocked by verapamil. 9 days later, the value of A492 of SP cells was 2.1, the cloning forming efficiency was (38.7 ± 6.8) % , the positive rate of CD133 was 69.63%, which were significantly higher than cells 0. 5, ( 15.5 ± 2.8)%, 16.71% of corresponding non-SP( P <0.05). The expression of ABCG2 in SP cells was significantly higher than that in non-SP cells. Conclusions SP cells existed in human pancreatic cancer cells SW1990.

Abstract:Objective To establish the method of purification of CD133 + gallbladder carcinoma cells and to investigate the different biological characteristics of CD133 + cells and CD133-cells in GBC-SD cell lines.Methods Flow cytometry was used to determine the proportion of CD133 + in two gallbladder carcinoma cell lines therein the GBC-SD was sorted by magnetic activated cell sorting (MACS) method.The CD133 mRNA and protein expression was evaluated by using semi-quantitative polymerase chain reaction and immunofluorescence.The differences of cell proliferation and drug resistance to 5-fluoropyrimidinedione (5-Fu) between CD133+ group and CD133-group were evaluated by cell counting Kit 8-assay (CCK-8).The growth of single cells in suspension culture was observed by colony formation assay.Results Two gallbladder carcinoma cell lines including GBC-SD and SGC996 were found to contain (68.5 ±2.9) ％ and (0.3 ± 0.2) ％ of CD133 + subsets cells respectively.The relative mRNA expression levels of CD133 in GBC-SD (0.6466 ±0.0259) was higher than in SGC996 (0.2181 ±0.0108,P ＜0.01).GD133 protein was localized on the cell surface,and showed strong expression in GBC-SD cells.After sorting,CD133 protein expression in CD133 + group was significantly stronger than that in CD133-group.The purity of CD133 + subsets cells isolated from GBC-SD by MACS was (90.4 ± 0.9) ％.The relative mRNA expression levels of CD133 in CD133+ group (0.7734 ±0.0217) was higher than that in CD133-group (0.2146 ± 0.0174,P ＜ 0.01).The proliferation ability of CD 133 + cells was stronger than that of CD 133-cells (P ＜0.01).The survival rate in CD133 + group was higher than that in CD133-group after treated with 5-Fu (P ＜0.05).The colony formation showed single CD133 + cells could form the new big sphere,and the formation rate of colony sphere in CD133 + group [(36.25 ±2.99)％] was higher than that in CD133-group [(4.50 ± 1.29)％,P ＜ 0.01].The nude mouse transplantation tumor experiment showed a remarkable difference in tumor formation rate among CD133 + group (100％),non-sorted group (60％),and CD133-group (0).Conclusion The high purity of CD133 + gallbladder carcinoma cells can be successfully isolated by MACS.Furthermore,the CD133 + subsets of gallbladder cancer possess some properties including the proliferation,chemoresistance,self-renewal and tumorigentic ability in vitro and in vivo.

Abstract:OBJECTIVE:To investigate an approach enriching cancer stem cells (CSCs) more effectively from laryngeal cancer cell line.METHODS:CD133(+)SP and CD133(-)SP subpopulation was detected and isolated from Hep-2 cell line using Hoechst33342 dye and phycoerythrin (PE)-conjugated CD133 monoclonal antibody assisted by fluorescence activated cell sorting technology. Sorted CD133(+)SP and CD133(-)SP cells were compared in CSCs-related assays including proliferation, differentiation, spheroid formation and drug sensitivity.RESULTS:CD133(+)SP cells accounted for a very small fraction of (0.30 ± 0.12)% in Hep-2 cell line, far less than the proportion of CD133(+) subgroup and side population subgroup, which were (3.15 ± 0.83)% and (17.1 ± 2.0)% respectively. Intriguingly, CD133(+)SP cells proliferated much faster than CD133(-)SP cells in RPMI1640 and gave rise to CD133(-)SP cells and other heterogeneous cells that formed the bulk of the tumor. In contrast, CD133(-)SP cells were not able to differentiate into CD133(+)SP cells. In serum-free medium CD133(+)SP cells grew as spherical clusters and remained floating. In addition, CD133(+)SP cells manifested the marked resistance to chemotherapy than CD133(-)SP cells.CONCLUSIONS:Compared with CD133(-)SP cells, CD133(+)SP subpopulation exhibited extraordinary cancer stem-like properties, were enriched for cancer stem cells more effectively and might serve as an ideal putative candidate for CSCs research in laryngeal cancer.

Abstract:Objectives Existence of side population (SP) in human ovarian cancer cell line OVCAR3 and characteristics of both stem cells and cancer cells were detected. Relation between SP phenotype and ABCG2 protein( ATP-binding cassette superfamily G member 2/breast cancer resistance protein, BCRP) was examined,and the potential to use OVCAR3 as tools for investigating the characteristics of cancer stem cells (CSCs)was estimated as well. Methods Hoechst 33342 efflux SP cells and dye-uptaking non-SP cells were separated by flow cytometry from human serous ovarian adenocarcinoma cell line OVCAR3. Cell colonyforming efficiency (CFE) of both subgroups ware evaluated. Soft agar assay was performed to compare the growth capacity of SP cells and non-SP cells. ABCG2 expression was detected by Western blotting and immunofluoreacence using an anti-ABCG2 monoclonal antibody. Results Flow cytometric analysis confirmed the presence of small subpopulations of cells excluding Hoechst dye in OVCAP,3 cells (2. 0% ) in a verapamil-sensitive manner. Hoechst dye uptake in OVCAR3 cells was enhanced by addition of verapamil, consistent with activity of ABCG2 protein. CFE of both SP and non-SP was 46.17%±23.27% and 6.17%±3.06%, respectively(P<0. 05). The Hoechst dye excluding SP cells exhibited immunoreactivity to stem cell markers ABCG2, demonstrated by Western blotting. They could grow in soft agar plate and formed cell sphere. Conclusions Human epithelial ovarian carcinoma cell line OVCAR3 contains a small subpoptdation of cells that exhibits a cancer stem cell-like phenotype. Special significance is the expression of ABCG2 in human tumor cells, a calcium-sensitive cell surface protein that acts to exclude Hoechst dye. These cells play a principal role in maintaining cancer cell line. It can be seen that cancer cell lines are potentially useful tools for investigating the characteristics of CSCs under controlled experimental conditions.

Abstract:The present study aimed to isolate and identify the properties of the cluster of differentiation (CD) 133(+) subset in human gallbladder cancer cells. The CD133(+) and CD133(-) subpopulations of the GBC-SD cell line were separated using immunomagnetic separation, and the biological features of the two subpopulations were analyzed in vitro and in vivo. In particular, the present study aimed to determine whether the two subpopulations were resistant to anti-tumor reagents and to identify the underlying molecular mechanisms involved. Following cell sorting of GBC-SD cells using immunomagnetic beads, 90.2 +/- 2% of cells were identified as CD133(+). Immunofluorescence confirmed that CD133 was expressed at higher levels in the Cd133(+) group compared with the CD133(-) group. The proliferation of the CD133(+) group was significantly increased compared with the CD133(-) group in vitro and in vivo. Following treatment with fluorouracil or gemcitabine, cells in the CD133(+) group exhibited a decreased sensitivity to these drugs. The number of transmembrane cells was significantly increased in the CD133(+) group compared with the CD133(-) group. In addition, the expression levels of ATP binding cassette subfamily G member 2, CD44, C-X-C motif chemokine receptor 4 (CXCR4), phosphorylated-protein kinase B (Akt) and CD133 in the CD133(+) group were significantly increased, compared with those in the CD133(-) group. In CD133(+) GBC-SD cells, stromal cell-derived factor 1 alpha (SDF-1 alpha) or treatment with AMD3100, an inhibitor of CXCR4, promotes or suppresses the SDF-1 alpha/CXCR4 axis, respectively, resulting in increased or decreased CD133 expression through the Akt signaling pathway. Inhibition of the Akt signaling pathway resulted in decreased CD133 expression in GBC-SD cells. Immunomagnetic beads were successfully used for isolation of the CD133(+) subset from GBC-SD cells. Furthermore, the CD133(+) subset revealed an increased potential for tumor formation, cell proliferation, invasion and resistance to chemotherapeutic agents with expression of stem cell-associated genes. Therefore, in GBC-SD cells, the CXCR4/Akt/CD133 signaling pathways may be activated.

Abstract:Objective To sort side population cells from hepatocellular carcinoma cell line HepG2 and analze the biological characteristics.Methods Fluorescence-activated cell sorting (FACS) was used to sort side population (SP) cells from HepG2 cells.SP cells served as experimental group and the same generation of HepG2 cells as control group.Non-obese diabetic/severe combined immunodeficiency(NOD/SCID)xenograft transplant experiments,cell-matrix adhesion experiment,Matrigel invasion assay,migration assay and suspension sphere formation assay were used to test the differences in biological characteristics between SP cells and original HepG2 cells.Results NOD/SCID xenograft transplant experiments demonstrated that 2 × 104 SP cells were sufficient to form tumor.There was no significantly difference in adhesion ability [(16.19 ± 3.68) ％ vs.(25.01 ± 13.97) ％] and metastatic potential [(20 ± 4) vs.(16 ± 3)] between the two groups.There was significant difference between experimental group and control group in migrating cells [(87 ± 10) vs.(47 ± 8),(P ＜ 0.05)] and spheres [(54 ± 9) vs.(18 ± 3),(P ＜ 0.05)].Conclusion SP cells sorted from HepG2 cells have cancer stem cell-like property and function,indicating that they may richen in hepatocarcinoma stem cells.

Abstract:Gallbladder cancer is the most common malignant neoplasm of the biliary tract, responsible for 80–95% of cases. Appropriate models are required for investigating the molecular pathogenesis of gallbladder cancer. In this study, we aimed to establish a gallbladder cancer cell line from primary tumour. Single cell RNA sequencing, Light and electron microscopy, DNA content analysis, cytogenetic analysis, short tandem repeat (STR) DNA fingerprint analysis, immunophenotypic characterization, and xeno-transplantation were utilized to characterize the novel ZJU-0430 cell line in vitro and in vivo. The cell line showed multiple cell shapes and characteristic epithelial morphologies under the microscope, but no too much heterogeneity by scRNA-Seq, with a population doubling time (PDT) of 19.81 h, which was shorter than that for GBC-SD cells. An immunophenotypic analysis revealed that ZJU-0430 cells were positive for CD24, CD44, CD29 and CD133 expression, and partially positive for CD184, and CD326 expression, and negative for CD34, CD90, CD117, and CD338 expression, similar to the primary cancer cells. A pathological analysis confirmed the origination of cell line from gallbladder tumour. ZJU-0430 cells had higher migration, invasion and proliferation properties than GBC-SD cells in vitro, and showed in vivo tumorigenicity in nude mouse xenograft settings. The results confirm the potential utility of ZJU-0430 cell line as a representative model of gallbladder cancer and suggest that it could be used in the in vitro and in vivo studies of gallbladder cancer pathogenesis and to develop new therapeutics.
