Abstract:Objective To construct the small interfere RNA(siRNA)plasmids with different tar- gets of mitogen-activated protein kinase(MEK) and observe its interfere effects on,cell cycle and DNA methylation in human colon cancer cells SW1116.Methods Different specific siRNA oligonucleotides were designed according to MEK sequences in GenBank,and cloned into pGC silencer eukaryotic expres- sion vector,then sequence analysis was performed.The recombinant plasmids were transfected into SW1116 cell by Lipofectamine,then the cells were cultured in G418 selective medium.The fluorescence microscopy was used to evaluate the efficacy of transfection,Western blot was used to determined the effect of MEK siRNA on silencing MEK or ERK signaling pathway activation.The methylation status of the promoter of the p16~(INK4A)gene was assayed by methylation-specific PCR and bisulfite DNA sequencing.Cell via- bility was analyzed by MTT,and cell cycle distribution was studied by flow cytometry.Results The two recombinant siRNA expression vectors of MEK mRNA were successfully constructed,transfected with SW1116 individually or simultaneously.The mean efficacy of transfeciton was 72.1%,MEK protein was silenced by siRNA,the inhibition rates were 74.2%,69.1%and 90.2%after transfection and selective cultured for 2 weeks.The protein expression of ERK,the downstream molecule of MEK was down-reg- ulated,the cell viability was decreased 2-4 folds,and cell cycle was blocked in G1 phase,the methylation status of the promoter of p16~(INK4A) gene was demethylation.Conclusions The recombinant siRNA vectors of MEK can effectively suppress the expression of MEK in SW1116.down-regulate the MEK signaling and decrease the methylation of p16~(INK4A)gene,along with the inhibition of cell cycle progrsssion.
