Effect of propofol on spatial performance and level of hippocampal NMDA receptor NR2A subunit in rats

Abstract:Objective To investigate the effect of propofol on the spatial performance in Morris water maze(MWM)and the changes in hippocampal NMDA receptor NR2A subunit by immunofluorescence in rats.Methods Thirty Sprague-Dawley rats were randomly divided into propofol group and control group.An intraperitoneal injection of propofol(50 mg/kg)was performed in the propofol group.After righting reflex of rats were lost,propofol(50 mg/kg)was administered intraperitoneally again in order to maintain anesthesia.The control group was blank.Twelve hours later,rats were subjected to MWM so that their spatial learning and memory could be evaluated.Meanwhile the changes in hippocampal NMDA receptor NR2A subunit expression were measured by immunofluorescence.Results The escape training latencyin of the propofol group was shorter than that of the control group(P<0.05),the time spent on target quadrant in probe test was longer in the propofol group than in the control group(P<0.05).Hippocampal NMDA receptor NR2A subunit expression was increased in the propofol group.Conclusion Spatial learning and memory is improved and expression of hippocampal NMDA receptor NR2A subunit obviously is increased in rats after single anesthesia by propofol.
