Effects of total alkaloids from Radix Linderae on adjuvant-induced arthritis in rats

Abstract:Objective: To investigate the preventive and therapeutic effects of total alkaloids from Radix Linderae(TARL) on adjuvant-induced arthritis(AA) in rats and underlying mechanisms.Methods: AA was induced by a single intradermal injection of Freund's complete adjuvant into the right hindpaws of rats.Effects of TARL on the primary and secondary paw swelling as well as body weights of rats were observed.In vitro,effect of TARL on splenocyte proliferation was determined by MTT assay.Effects of TARL on production of nitric oxide and secretion of interleukin 1 from mouse peritoneal macrophages were detected by Griess reagent assay and thymocyte proliferation method,respectively.Results: Similarly to glucosides from roots of Tripterygium wilfordii Hook.f.(雷公藤多苷),TARL dramatically attenuated the secondary paw swelling,and increased the body weights in AA rats.But, it only showed an inhibitory tendency to the primary paw swelling.TARL concentration-dependently reduced Con A-induced proliferation of spleen lymphocytes and LPS-induced release of nitric oxide and interleukin 1 from peritoneal macrophages of mice.Conclusion: TARL shows preventive and therapeutic effects on adjuvant-induced arthritis,which is probably mediated by downregulating the functions of T lymphocytes and macrophages.
