Abstract:The hydrogen peroxide (H2O2) and cytochrome c-dependent oxidation of o-phenylenediamine (o-PD) was investigated by spectrophotometry and electrochemistry. The results indicated that o-PD underwent facile catalytic oxidation in the presence of cytochrome c, and that the degradation of cytochrome c by hydrogen peroxide can also be partly prevented in the presence of o-PD. The hydroxyl radical scavengers (mannitol and sodium benzoate) and oxo-heme species scavenger (uric acid) do not inhibit the oxidation, which implies that the hydroxylation of o-PD may not be involved in its oxidation. Combining with the results of the mass spectrum, elemental analysis, nuclear magnetic resonance and Fourier transform infrared spectrum of the isolated product, a conceivable structure of the product was suggested.
Electrochemical Probe of Hydroxylation of 4-Nitrophenol by Cytochrome C with Hydrogen Peroxide

Abstract:The reaction of hydrogen peroxide with cytochrome c makes them coupled to lead to the hydroxylation of 4-nitrophenol. In situ electrochemical probe was used to detect the hydroxylation of 4-nitrophenol, which can avoid the tedious extraction procedure, the loss of the active species and the interference of some colored substances in the detection of 4-nitrocatechol by spectroscopic method. The hydroxyl radical scavengers mannitol and sodium benzoate did not eliminate hydroxylation, but the inhibitory effect of uric acid on the hydroxylation lead to the formation of the ferryl species of the protein during the reaction. These studies suggest that the electrochemical probe might efficiently detect the trace 4-nitrocatechol from the onset of the hydroxylation reaction and thus provides a more sensitive tool.

Abstract:An amperometric biosensor for hydrogen peroxide (H 2 O 2 ) has been constructed by immobilizing cytochrome c on an indium/tin oxide (ITO) electrode modified with a macroporous material. Cyclic voltammetry showed that the direct and quasi-reversible electron transfer of cytochrome c proceeds without the need for an electron mediator. A surface-controlled electron transfer process can be observed with an apparent heterogeneous electron-transfer rate constant (k s ) of 29.2 s −1 . The biosensor displays excellent electrocatalytic responses to the reduction of H 2 O 2 to give amperometric responses that increase steadily with the concentration of H 2 O 2 in the range from 5 μM to 2 mM. The detection limit is 0.61 μM at pH 7.4. The apparent Michaelis-Menten constant (K m ) of the biosensor is 1.06 mM. This investigation not only provided a method for the direct electron transfer of cytochrome c on macroporous materials, but also established a feasible approach for durable and reliable detection of H 2 O 2 . Figure Biosensor for hydrogen peroxide was developed by immobilizing cytochrome c in the macroporous ordered silica foam (MOSF) through the electrostatic interaction. The achievement of the direct electron transfer between cytochrome c and electrode surface indicated that the MOSF modified electrode displayed good affinity and biocompatibility for cytochrome c .

Abstract:We report on an amperometric biosensor for hydrogen peroxide. It is obtained via layer-by-layer assembly of ordered mesoporous carbon nanospheres and poly(diallyldimethylammonium) on the surface of an indium tin oxide (ITO) glass electrode and subsequent adsorption of cytochrome c. UV–vis absorption spectroscopy was applied to characterize the process of forming the assembled layers. Cyclic voltammetry revealed a direct and quasi-reversible electron transfer between cytochrome c and the surface of the modified ITO electrode. The surface-controlled electron transfer has an apparent heterogeneous electron-transfer rate constant (k s ) of 5.9 ± 0.2 s−1 in case of the 5-layer electrode. The biosensor displays good electrocatalytic response to the reduction of H2O2, and the amperometric signal increase steadily with the concentration of H2O2 in the range from 5 μM to 1.5 mM. The detection limit is 1 μM at pH 7.4. The apparent Michaelis-Menten constant (K m ) of the sensor is 0.53 mM. We assume that the observation of a direct electron transfer of cytochrome c on mesoporous carbon nanospheres may form the basis for a feasible approach for durable and reliable detection of H2O2.
Probing trace of intracellularly-originated hydrogen peroxide based Pt-Cd bimetallic nanozyme on an enzyme-free electrochemical sensor

Abstract:Background: Measurement of endogenous cellular hydrogen peroxide (H2O2) can provide information on cellular status, and help to understand cellular metabolism and signaling processes, thus contributing to elucidation of disease mechanisms and new diagnostics/therapeutic approaches. Results: In this work, Pt-Cd bimetallic nanozyme was successfully prepared via the solvothermal synthetic method for sensitive detection of H2O2. The synthesized Pt-Cd bimetallic nanozyme could exhibited good electrochemical activity. Then, the materials were analyzed for the electrochemical properties and catalytic properties of H2O2 by cyclic voltammetry and chronoamperometry, respectively. Results indicated that the synthesized nanozyme had superior sensitivity (295 μA⸳mM-1⸳cm-2) and selectivity toward H2O2 with a detection limit of 0.21 μM. Further, the Pt-Cd bimetallic nanozyme displayed good electrochemical properties compared to platinum catalysts alone. The application was extended to determine the produced H2O2 from human hepatocellular carcinoma cells (HepG2) and normal hepatocyte (LO2) samples after ascorbic acid stimulation, thus enabling the early warning of cellular carcinogenesis. Significance: This strategy promises simple, rapid, inexpensive and effective electrochemical sensing and provides a new pathway for the synthesis of bimetallic nanozymes to construct an electrochemical sensor for the sensitive detection of H2O2.

Abstract:Colloidal gold modified carbon paste electrodes were prepared by mixing 24-nm-diameter colloidal Au particles with carbon paste. The modified electrodes displayed a low charging current and a favorable electrochemical response of hexacyanoferrate (111). The direct electrochemical behavior of a horse-heart cytochrome c (cyt.c) adsorbed on this electrode surface is described. It showed a surface-controlled electrode process with the electron transfer rate constant of (1.21 +/- 0.08) s(-1) and alpha of 0.67. Cyclic voltammograms showed small peak-to-peak separations at low scan rates. The adsorbed cyt.c maintained its activity and could also electrocatalyze the reduction of hydrogen peroxide. Since this behavior was quite pronounced the electrode was used for H2O2 detection. The Km,PP value for this sensor was found to be 2.28 +/- 0.17 mM, allowing measurements down to 0.01 mM H2O2.

Abstract:In this paper,a luminol analog,L012,with a high-electrochemiluminescence(ECL)illumination is covalently assembled at indium tin oxide(ITO)surface that exhibits the ECL response to hydrogen peroxide in the solution.The ITO slide is firstly functioned with amine group that react with glutaraldehyde to introduce aldehyde group at the surface.Upon the exposure to L012 with amine group,the reaction between the aldehyde group and the amine group results in the linkage of L012 at the electrode surface.In the presence of hydrogen peroxide,enhanced ECL from L012 at the electrode surface is observed that is linearly related with the concentration of hydrogen peroxide.The detection limit is determined to be 4.3 μM(S/N=3).The successful establishment of ECL response to hydrogen peroxide using L012 modified electrode will provide a new functioned ECL surface for the future ECL imaging of hydrogen peroxide release from single cells.

Abstract:Efficient catalytic decomposition of H 2 O 2 is accompanied by electron transfer through Fe-N x active sites of hemin in the human body. Inspired by this reaction process, the Fe SAs/Co CNs were successfully synthesized by combined Co atomic sites with nitrogen-carbon doped Fe single-atom active sites (SAs). The synergy between transition metals not only reduces agglomeration during synthesis but also improves its own electrical conductivity due to the interaction between Fe and Co that promotes the formation of graphite surface. Crucially, the synergistic effect of the Co site significantly enhanced the peroxidase activity of the Fe SAs and the reaction rate of the Fenton-like reaction, resulting in an efficient detection of H 2 O 2 . Catalytic kinetic calculations and enzymatic kinetic calculations were used to verify the electron transfer rate and catalytic performance of their constructed electrochemical sensing interfaces. The results showed that Fe SAs/Co CNs@GCE showed better detection performance than Fe SAs CNs@GCE. It was applied successfully to detect H 2 O 2 released from cells in real-time as well. The linear detection range of Fe SAs/Co CNs@GCE for H 2 O 2 was 1–16664 μM, and the detection limit was as low as 0.25 μM. Furthermore, an electrochemical sensing chip was constructed using Fe SAs/Co CNs@SPE and the prepared microfluidic channel. The constructed portable FeSAs/Co CNs@SPE had a linear range of 1–400 μM and a detection limit of 0.36 μM and achieved the recovery detection of H 2 O 2 in serum. The electrochemical sensing interfaces constructed based on Fe SAs/Co CNs all have efficient catalytic performance and excellent real-time hydrogen peroxide detection performance, which have practical application potential for human oxidative stress level detection. And it provides a novel approach to the trace detection of bioactive small molecules.

Abstract:A carboxyl ion implantation-modified indium tin oxide (COOH/ITO) electrode was prepared, and the existence of a carboxyl group on the electrode was verified by X-ray photoelectron spectroscopy (XPS). Hemoglobin (Hb) was immobilized on the electrode, and cyclic voltammetric techniques were employed to evaluate the electrochemical behavior of Hb. Based on the activity of Hb on the COOH/ITO electrode towards the reduction of hydrogen peroxide, a mediator-free H2O2 biosensor was constructed, which showed a linear range from 1.0 μM to 200.0 μM with a detection limit of 0.3 μM (S/N = 3).
Electrochemical Approach of the Reductive Activation of O 2 by a Nonheme Fe II Complex. Some clues for the Development of Catalytic Oxidations.

Abstract:We report an in-depth study of the reductive activation of O 2 by the non heme [Fe II (L 5 2 )(MeCN)] 2+ complex carried out by cyclic voltammetry. Experimental evidence is obtained for the slow coordination of dioxygen to the ferrous center yielding an Fe II /O 2 adduct with a strong Fe II -O 2 character rather than an Fe III -superoxo one. Electron injection in the Fe II -O 2 species occurs at a potential of ca. -700 mV vs SCE, i.e. 200 mV above the O 2 to O 2 •- reduction, leading to the formation of a Fe III -peroxo intermediate and then Fe III -hydroperoxo upon protonation by residual water. The experimental CVs recorded at variable scan rate or variable Fe II concentration are well simulated taking into account a detailed mechanism initiated by the competitive reduction of O 2 and the Fe II -O 2 adduct. Analysis of the concentration of the reaction intermediates generated as a function of the applied potential indicates that the Fe III -peroxo intermediate significantly accumulates at a potential of -650 mV. Oxidative bromination of anisole is assayed under electrolytic conditions at this potential to yield bromoanisole products. The low faradic yields observed reveal that deleterious reactions such as direct reduction of reaction intermediates likely occur. Based on the detailed mechanism elucidated, a number of improvements to achieve more efficient catalytic reactions can be proposed.

Abstract:The direct electrocatalytic reduction of hydrogen peroxide in alkaline medium at a carbon ionic liquid electrode modified with copper oxide nanoparticles was investigated. The electrode was prepared by mixing graphite particles, ionic liquid (n-octylpyridium hexafluorophosphate) and copper oxide nanoparticles. Unlike the film-modified electrode, the fabrication of this electrode is simple and highly reproducible. The combination of the good conductivity of the ionic liquid and the high catalytic activity of the nanoparticles resulted in an electrode with attractive properties for the determination of hydrogen peroxide. The concentration of NaOH and the loading of copper oxide nanoparticles were optimized. The linear range for the determination of hydrogen peroxide is from 1.0 μM to 2.5 mM, the detection limit is 0.5 μM. High stability, sensitivity, selectivity and reproducibility, fast response, the ease of preparation, and surface renewal made the electrode well suitable for the determination of hydrogen peroxide in real samples.
Fabrication of Hierarchically Porous Carbon Networks for the Electrochemical Determination of Superoxide Anion Released from Living Cells

Abstract:Realizing the quantitative detection of superoxide anions (O-2(center dot-)) is of great significance to the pathological research based on the correlation between O-2(center dot-) and many diseases. Currently, the main challenge for O-2(center dot-) electrochemical detection in the actual sample is the development of effective nano-mimetic materials to replace natural enzymes to achieve high-selectivity detection of O-2(center dot-). In this work, hierarchically porous carbon networks (HPCN) were successfully prepared through 1,3,5-trimethylbenzene (TMB)-induced self-assembly and carbonization strategy. Owing to the introduction of TMB, HPCN obtains the changed morphology, enriched pore structure, and improved defect degree, which promotes mass transfer and leads to faster electron transfer efficiency. Therefore, the non-enzymatic O-2(center dot-) electrochemical sensor fabricated with HPCN (denoted as HPCN/SPCE) exhibits high sensitivity (607.4 mu A cm(-2) mM(-1)) and excellent selectivity for detecting O-2(center dot-). Importantly, the sensor successfully achieved the dynamic monitor of O-2(center dot-) released from living cells and has the potential to distinguish between cancer and normal cells.
A Novel Non-Enzymatic Electrochemical Hydrogen Peroxide Sensor Based on a Metal-Organic Framework/Carbon Nanofiber Composite

Abstract:A co-based porous metal-organic framework (MOF) of zeolitic imidazolate framework-67 (ZIF-67) and carbon nanofibers (CNFs) was utilized to prepare a ZIF-67/CNFs composite via a one-pot synthesis method. Scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), and X-ray diffraction (XRD) were employed to investigate the morphology, structure, and composition of the resulting composite. A novel high-performance non-enzymatic electrochemical sensor was constructed based on the ZIF-67/CNFs composite. The ZIF-67/CNFs based sensor exhibited enhanced electrocatalytic activity towards H2O2 compared to a pure ZIF-67-based sensor, due to the synergistic effects of ZIF-67 and CNFs. Meanwhile, chronoamperometry was utilized to explore the detection performance of the sensor. Results showed the sensor displayed high-efficiency electrocatalysis towards H2O2 with a detection limit of 0.62 μM (S/N = 3), a sensitivity of 323 μA mM−1 cm−2, a linear range from 0.0025 to 0.19 mM, as well as satisfactory selectivity and long-term stability. Furthermore, the sensor demonstrated its application potential in the detection of H2O2 in food.

Abstract:Hydrogen peroxide (H2O2) biosensors are some of the most promising sensors being developed, because they are highly selective and respond quickly to a specific substrate. Hemoglobin (Hb) has been used as an ideal biomolecule for the construction of hydrogen peroxide biosensors because of its high sensitivity to H2O2. Direct electron transfer from Hb has been widely investigated for application in the detection of H2O2 because of the reaction’s simplicity, high selectivity and intrinsic sensitivity. A method for the electrochemical detection of hydrogen peroxide based on the immobilization of hemoglobin on a DNA/Fe(pyterpy)2+-modified gold electrode was investigated. The pyterpy monolayers were primary electron depositions on the gold electrode surface of a quartz crystal microbalance (QCM). This layer offers a template to which negatively charged DNA attaches. The process of fabricating the electrode was verified by a quartz crystal analyzer (QCA). The experimental parameters, such as pH, applied potential and amperometric response, were evaluated and optimized. Under the optimized conditions, this sensor shows a linear response between 3.0×10−6 and 9.0×10−4M concentrations of H2O2. The detection limit was determined to be 7×10−7M (based on S/N=3).

Abstract:Using chitosan as an effective linker between CMK-3 and glassy carbon electrode surface, {Hb/CMK-3}n multilayer film-modified electrodes were constructed through layer-by-layer assembly. The morphology of thus-formed {Hb/CMK-3}n film was characterized by scanning electron micrographs, and the interaction of hemoglobin (Hb) with CMK-3 was studied by UV–vis spectroscopy and electrochemical methods. Under optimal conditions, {Hb/CMK-3}6 film showed a couple of stable and well-defined redox peaks at about −377 and −296mV in pH 7.0 buffers. Furthermore, the {Hb/CMK-3}6 film displayed excellent electrocatalysis to the reduction of both H2O2 and O2. Based on thus-formed film and its direct electron transfer behavior, a novel biosensor was presented for the determination of H2O2 ranging from 1.2 to 57μM with the detection limit of 0.6μM at S/N=3. CMK-3 provided a desirable matrix for protein immobilization and biosensor preparation.

Abstract:The electrochemical identification of the urea denaturation of horse heart cytochrome c in bulk solution at the 4,4′-dithiodipyridine-modified gold electrode is reported. The results are similar to the three-step transitions of equilibrium studies (Myer et al., Biochemistry, 19 (1980) 199) of urea denaturation of cytochrome c in bulk solution. This method permits a clear resolution of which of the three steps of urea denaturation is electrochemically related. In addition, by analysing the effects of urea on the structural forms of cytochrome c and on the solution properties, as well as the cyclic voltammetric responses of the protein, the individual forms of the urea denaturation of cytochrome c can be understood. The results reflect the superposition of protein denaturation on the electrode surface and in solution.

Abstract:A cytochrome c oxidase model that consists of a cobalt(II) porphyrin with a copper(I) triazacyclononane macrocycle fastened on the distal face and an imidazole covalently attached to the proximal face has been synthesized and characterized. Redox titrations with molecular oxygen (O-2) and cobaltocene were carried out, and O-2 was found to bind irreversibly in a 1:1 ratio to the model compound. This O-2 adduct (a bridged peroxide) can be fully reduced to the deoxygenated form with four equivalents of cobaltocene. The model compound was adsorbed on an edge-plane graphite electrode, and rotating ring-disk voltammetry was used to monitor the electrocatalytic reduction of O-2. Four-electron reduction of O-2 was observed at physiological pH.

Abstract:This paper describes the construction of a mixed monolayer of ferrocenylalkanethiol and encapsulated horseradish peroxidase (HRP) at a gold electrode for amperometric detection of H2O2 at trace levels. By tuning the alkanethiol chain lengths that tether the HRP enzyme and the ferrocenylalkanethiol (FcC(11)SH) mediator, facile electron transfer between FcC(11)SH and HRP can be achieved. Unlike most HRP-based electrochemical sensors, which rely on HRP-facilitated H2O2 reduction (to H2O), the electrocatalytic current is resulted from an HRP-catalyzed oxidation reaction of H2O2 (to O-2). Upon optimizing other experimental conditions (surface coverage ratio, pH, and flow rate), the electrocatalytic reaction proceeding at the electrode was used to attain a low amperometric detection level (0.64 nM) and a dynamic range spanning over 3 orders of magnitude. Not only does the thin hydrophilic porous HRP capsule allow facile electron transfer, it also enables H2O2 to permeate. More significantly, the enzymatic activity of the encapsulated HRP is retained for a considerably longer period (>3 weeks) than naked HRP molecules attached to an electrode or those wired to a redox polymer thin film. By comparing to electrodes modified with denatured HRP that are subsequently encapsulated or embedded in a poly-L-lysine matrix, it is concluded that the encapsulation has significantly preserved the native structure of HRP and therefore its enzymatic activity. The electrode covered with FcC(11)SH and encapsulated HRP is shown to be capable of rapidly and reproducibly detecting H2O2 present in complex sample media.
Direct Detection of a Single [4Fe-4S] Cluster in a Tungsten-Containing Enzyme: Electrochemical Conversion of CO2 into Formate by Formate Dehydrogenase

Abstract:The conversion of CO2 into fuels and valuable chemicals is one of the central topics to combat climate change and meet the growing demand for renewable energy. Herein, we show that the formate dehydrogenase from Clostridium ljungdahlii (ClFDH) adsorbed on electrodes displays clear characteristic voltammetric signals that can be assigned to the reduction and oxidation potential of the [4Fe-4S](2+/+) cluster under nonturnover conditions. Upon adding substrates, the signals transform into a specific redox center that engages in catalytic electron transport. ClFDH catalyzes rapid and efficient reversible interconversion between CO2 and formate in the presence of substrates. The turnover frequency of electrochemical CO2 reduction is determined as 1210 s(-1) at 25 degrees C and pH 7.0, which can be further enhanced up to 1786 s(-1) at 50 degrees C. The Faradaic efficiency at -0.6 V (vs. standard hydrogen electrode) is recorded as 99.3% in a 2-h reaction. Inhibition experiments and theoretical modeling disclose interesting pathways for CO2 entry, formate exit, and OCN- competition, suggesting an oxidation-state-dependent binding mechanism of catalysis. Our results provide a different perspective for understanding the catalytic mechanism of FDH and original insights into the design of synthetic catalysts.
