Expression and Their Clinical Significance of Mir-222 and MBD2 in Tissues of Colon Carcinoma

Abstract:Background and purpose:MicroRNAs are 19 to 25-nucleotide noncoding RNA molecules that regulate gene expression at the level of transcription and translation.This study investigated the expression of the miR-222 and methyl-CpG binding domain protein 2(MBD2) in tissues of colon carcinoma and their clinical significance.Methods:Tissue specimens and adjacent tissues of 76 cases as control from Jan.2008 to Jun.2010 were collected,and hybridization in situ and immunohistochemistry were used to detect the miR-222 and MBD2 expression in those specimens respectively.Results:The positive expression rate of the miR-222 was 92.1% in colon carcinoma and 28.9% in adjacent comparison.The up-regulation of miR-222 expression was associated with advanced clinical stage,and the expression of miR-222 was higher with lymph node metastasis than without lymph node metastasis in colon carcinoma.The positive expression rate of the MBD2 was 19.7% in colon carcinoma and 81.6% in adjacent comparison.The down-regulation of MBD2 expression was associated with advanced clinical stage,and the expression of MBD2 was lower with lymph node metastasis than without lymph node metastasis in colon carcinoma.The expression of miR-222 had a close negative correlation to that of MBD2 in colon carcinoma(P0.01).Conclusion:Overexpression of miR-222 might be associated with the tumorigenesis,development and metastasis by targeting MBD2,and they could be the indicators in the diagnosis and prognosis of colon carcinoma.
