Abstract:Objective: This study aimed to investigate the expression of EphB4 receptor and its ligand EphrinB2 in esophageal squamous cell carcinoma and their relationship with angiogenesis.Methods: The expression of EphB4 receptor and its ligand EphrinB2 and VEGF was detected by immunohistochemistry in tumor specimens from 60 esophageal squamous cell carcinoma cases and paratumor normal specimens from 10 cases.The expression of EphB4,EphrinB2,and VEGF mRNA in 60 patients with esophageal cancer and 10 patients with paratumor normal esophageal mucosa was detected by reverse-transcription polymerase chain reaction.The correlation among the expression levels of EphB4,EphrinB2,and VEGF mRNA was analyzed.Results: The expression levels of EphB4,EphrinB2,and VEGF mRNA,as well as of the expression levels protein,were significantly higher than that in paratumor normal esophageal mucosa(P<0.05).The relative amount of EphB4 mRNA was positively correlated with EphrinB2 mRNA in patients with esophageal cancer(r=0.541,P<0.05).The relative amounts of EphB4 mRNA and EphrinB2 mRNA were positively correlated with VEGF mRNA in patients with esophageal cancer(r=0.642 and 0.582,respectively;P<0.05).Conclusion: EphB4 and EphrinB2 are highly expressed in esophageal cancer tissue,which may be closely associated with the angiogenesis of esophageal cancer.
Correlation of EphA2 Expression with Invasion and Angiogenesis of Colorectal Carcinoma

Abstract:Objective:To investigate the expression and significance of erythropoietin-producing hepatocellularA2(EphA2) protein in colorectal carcinoma and the correlation between EphA2 expression and tumor angiogenesis.Methods: Immunohistochemistry was used to examine the EphA2 expression in 70 surgically resected colorectal carcinoma and the paired normal colorectal mucosa tissues.Microvessels in colorectal carcinoma were counted after immunostaining for CD34,and the correlation between EphA2 expression and microvessel counts(MVCs)in colorectal carcinoma were assessed.Results: Expressions of EphA2 protein in carcinoma tissues were obviously higher than that in normal colorectal mucosa tissues(P 0.01).The overexpression of EphA2 protein correlated with differentiation grades,invasion depth and gross type of carcinomas(P 0.05),but didn't with age,sex of patients,lymph node,and hematogenous metastasis of adenocareinomas(P 0.05).We observed specific EphA2 staining was present in both cancer cells and tumor vascular endothelial cells.Microvessel density(MVD)after immunostaining for CD34 was significantly correlated with overexpression of EphA2,high microvessel density presented in EphA2-strong positive tumor areas.Conclusion: There is a correlation between EphA2 protein overexpression,and tumorigenesis infiltration of colorectal carcinoma.
Over-Expression of EphA2 and EphrinA-1 in Human Gastric Adenocarcinoma and Its Prognostic Value for Postoperative Patients

Abstract:This study aims to investigate the expression and significance of EphA2 and EphrinA-1 in human gastric adenocarcinoma progression and prognosis. The expression of EphA2 and EphrinA-1 was detected in the cell lines and tissues of gastric adenocarcinoma. Different expression levels of EphA2 and EphrinA-1 were found in two cell lines. The expression of EphA2 and EphrinA-1 was significantly higher in gastric adenocarcinoma tissues than in normal tissues. Statistical analysis showed a significant correlation of EphA2 expression with the depth of tumor invasion, tumor-node-metastasis (TNM) stages, and lymph node metastasis. EphrinA-1 over-expression was significantly correlated with TNM stages and lymph node metastasis, while EphA2 expression was found to be an independent prognostic factor of postoperative gastric adenocarcinoma. In conclusion, the increased expression of EphA2 and EphrinA-1 plays an important role in the progression of human gastric adenocarcinoma, in which elevated EphA2 expression is an independent factor that indicates poor prognosis in postoperative gastric adenocarcinoma.
High Expression of EphA1 in Esophageal Squamous Cell Carcinoma is Associated with Lymph Node Metastasis and Advanced Disease

Abstract:Eph receptors play important roles in the development of cancer. The present study aimed to investigate the expression of EphA1 and its clinicopathologic significance in esophageal squamous cell carcinoma (ESCC). The expression levels of the EphA1 transcript and protein were compared between ESCC and matched normal mucosa in the same patient. High expression levels of the EphA1 transcript and protein were detected in 25.6% (21/82) and 23.2% (19/82) of tumors compared with matched normal mucosa. The up‐regulation of EphA1 transcript in tumors was positively associated with differentiation (p = 0.002), disease stage (p = 0.034), and lymph node metastasis (p = 0.042). Increased expression of EphA1 protein in tumors was more often observed in patients with well‐differentiated tumors (p = 0.004), lymph node metastasis (p = 0.028), and advanced tumor stage (p = 0.008). EphA1 protein expression was detected in intraepithelial neoplasias as well. Decreased expression of EphA1 protein was detected in 65.2% (15/23) of samples with low‐grade neoplasia and in 85.3% (8/15) of samples with high‐grade intraepithelial neoplasia. Increased staining of EphA1 protein was not detected in low‐grade intraepithelial neoplasia samples, but was detected in 21.3% (2/15) of high‐grade intraepithelial neoplasia samples. Taken together, our results show that EphA1 appears to be a differentiation marker for esophageal squamous cells, and its increased expression is positively associated with lymph node metastasis and advanced disease stage.

Abstract:Objective To investigate the expression of EphA2 and its correlation with the clinicopathological features in hepatocellular carcinoma(HCC).Methods We collected 52 samples of surgically resected paired HCC and normal tumor-adjacent tissues.EphA2 expression was detected in the samples using immunohistochemical staining.The correlation between EphA2 protein expression and clinicopathological feature and tumor recurrence was analyzed.Results The protein expression of EphA2 in HCC tissues was significantly increased compared to those of the normal tumor-adjacent tissues(P<0.05).Clinicopathological analysis suggested that EphA2 protein high-expression was associated with more tumor number,tumor capsule absence,invasion of portal vein(PV) and advanced TNM stage(P<0.05).Multivariate analyses using Cox′s proportional hazards model suggested that EphA2 expression was significantly associated with HCC postoperative recurrence.Conclusion The expression of EphA2 is up-regulated in HCC tissues,and the high-expression of EphA2 is correlated with poor clinicopathological characteristics and recurrence of HCC,suggesting that EphA2 may play an important role in the progression and metastasis of HCC.
High Expression of EphA4 is Associated with Invasion and Lymph Node Metastasis in Colorectal Carcinomas.

Abstract:Eph receptors and ephrin ligands have been implicated in development of embryo, especially in vascular and nervous system. Increasing evidence suggests that Eph/ephrin signaling plays important roles in carcinogenesis. EphA4 is a member of Eph receptors and its role has been found in development of nerve and in tumorigenesis. Till now, EphA4 expression and its clinical significance in colorectal carcinoma have not been well investigated. In the present study, a set of 102 colorectal carcinoma tissue specimens were subjected to immunohistochemistry by using a specific polycolonal antibody for EphA4. Expression of EphA4 protein was more often detected in colorectal carcinoma than in normal mucosa (P<0.001). High expression of EphA4 protein in colorectal carcinomas was positively associated with tumor size (P=0.008, rs =0.260), depth of invasion (P=0.004, rs =0.286), lymph node metastasis (P=0.013, rs =0.244), and TNM stage (P=0.005, rs =0.279), and was negatively associated with age of patients (P=0.027, rs =-0.219). No significant relationship between expression of EphA4 and sex and tumor location was found. The survival analyses showed that patients whose carcinoma exhibited high expression of EphA4 had a poor outcome (P=0.006). Our results suggest that EphA4 play a role in the invasion and lymph node metastasis of colorectal carcinoma. The EphA4 protein expression level may be used as a potential prognostic marker in colorectal carcinoma. Furthermore, the EphA4 gene may provide a novel target for therapy of colorectal carcinoma.

Abstract:Objective To investigate the relationship between the expression of EphB2,EphrinB2,EphrinA1 and the pathogenesis of laryngeal carcinoma,and to explore the mechanism of pathogenesis of laryngeal carcinoma.Methods The expression of EphB2,EphrinB2 and EphrinA1 protein with the flow cytometere Fluorescence index was defined as the quantitative expression index of EphB2,EphrinB2 and EphrinA1 protein.The FI value over 1.0 would be considered as positive expression,otherwise,as negative expression.Results The qualitative expressions of EphB2,EphrinB2 and EphrinA1 in laryngeal carcinoma tissues were obviously higher than those in para-carcinoma and in normal laryngeal mucosa tissues respectively(P0.05 or 0.01),in laryngeal carcinoma tissues,the qualitative expressions of EphB2,EphrinB2 and EphrinA1 with positive metastasis were remarkably higher than those without metastasis respectively(P0.05 or 0.01),which in clinical stage Ⅲ,Ⅳ were significantly higher than those in clinical stageⅠ,Ⅱ respectively(P0.05 or 0.01).There was a multiple linear regression correlation in the expression of EphB2,EphrinB2 and EphrinA1 in laryngeal carcinoma tissues,and which was a positive correlation.Conclusion The high expression of EphB2,EphrinB2 and EphrinA1 may be related with the pathogenesis and development of laryngeal carcinoma.
Expression of EphA2 and E-cadherin in Gastric Cancer: Correlated with Tumor Progression and Lymphogenous Metastasis

Abstract:In this study, gastric cancer progression was correlated with the over-expression of erythropoietin-producing hepatocellular (Eph)A2 receptor and down-expression of epithelial cadherin (E-cadherin). Immunohistochemistry of EphA2 and E-cadherin were performed on these tumor samples from 165 primary lesions of gastric cancer. The results showed that expression of EphA2 was obviously increased in gastric cancer tissues ( P < 0.01), which was positively correlated with the depth of cancer invasion, tumor-node-metastasis (TNM) stage and lymph node metastasis ( P < 0.05). Meanwhile, the expression of E-cadherin was significantly reduced ( P < 0.01), which was negatively correlated with the depth of cancer invasion, grade of tumor differentiation, TNM stage and lymph node metastasis ( P < 0.05). The correlation between EphA2 and E-cadherin expression was negative ( r = −0.198, P = 0.011). In conclusion, either the over-expression of EphA2 or the down-expression of E-cadherin is correlated with cancer progression and lymphogenous metastasis in gastric cancer, suggesting that both of them may play an important role in tumor progression and metastasis.
Expression of EphB2 in gastric carcinoma and the correlation with metastasis

Abstract:Objective:The receptor tyrosine kinase EphB2 has recently been shown to be a direct transcriptional target of TCF/beta-catenin.To detect the expression of EphB2 in the tumor tissues of primary gastric carcinoma as well as lymphatic and visceral metastases and to investigate their roles in tumorigenesis and metastasis,and gastric cancer clinical pathology parameters of the relationship.Methods:The protein expression of EphB2 in normal mucosatissues(n=71),primary tumors(n=71),lymp haticmetastases(n=71),and distant visceral metastases(n=26)were examined by immunohistochemical staining and tissue microarray.The difference between primary tumor and normal tissue as well as primary tumor and matched metastases were compared.Results:The positive rate of EphB2 decreased gradually accompanied with the tumor development.Immunohistochemical staining revealed down-regulated expression of EphB2 in lymph node and visceral metastases.Positive rates of EphB2 expression in normal tissues,primary tumor tissues,lymph node and visceral metastases were 83.1%(59/71),60.6(43/71),43.7%(31/71)and 26.9%(7/26).EphB2 expression was significantly higher in normal tissues than that in primary tumor tissues(P<0.01).It was significantly lower in lymph node and visceral metastases than that in primary tumor tissues(P=0.003).Primary tumors and metastases in EphB2 expression exist in the case and gender(male)and highly relevant to the depth of invasion.(P<0.05).Conclusion:Tumor metastasis results from gradual accumulation of abnormal genetic alterations.Down-regulation of EphB2 play a critical role in metastasis of gastric carcinoma.
The relationship between the expression of EphA1 in gastric carcinoma, metastasis and survival

Abstract:Background and purpose:Receptor tyrosine kinases of the Eph subfamily are involved in the carcinogenesis.In this study,EphA1 expression as well as associated EphA1 expressions within clinicopathologic parameters were analyzed to investigate the role of EphA1 in gastric carcinoma.Methods:The expression of EphA1 transcription in gastric carcinoma tissues and corresponding normal tissues were determined by the quantitative real time reverse transcriptase polymerase chain reaction(real time RT-PCR).The status of CpG island associated with the promoter of EphA1 were checked by methylation specific PCR(MSP) whereas the protein of EphA1 was examined through immunohistochemical staining(IHC).Results:Down-regulation of the EphA1 transcription was detected in 19(34%) cases,up-regulation in 14(25%) cases,and no differences in expression were detected in 23(41%) cases. The expression level of EphA1 transcription was associated with tumor size(P=0.05),stage(P=0.001) and lymph node metastasis(P=0.011).Methylated EphA1 DNA was detected in most examined carcinomas(12/15) with EphA1 down-regulation but rarely in those with up-regulaton(1/5) or those with no difference in expression(3/18)(P=0.001). IHC in 145 tissue specimens showed that the clinicopathological significance of EphA1 protein was consistent with that of mRNA.The survival analysis in 89 cases showed that patients with a down-regulation of EphA1 had a better prognosis than those with up-regulation(P=0.005).Conclusion:EphA1 may play an important role in the invasion and metastasis of gastric carcinoma.
Predictive Value of Epha2 and Ephrina-1 Expression in Oesophageal Squamous Cell Carcinoma

Abstract:The aim of this study was to analyse the protein and mRNA expressions of EphA2 and EphrinA-1 in oesophageal squamous cell carcinomas and to explore their clinicopathological associations and predictive values in oesophageal squamous cell carcinoma. Tissue array and immunohistochemistry were used to assess the protein expressions of EphA2 and EphrinA-1 in tumours from 173 patients with oesophageal squamous cell carcinoma. Paraffin sections from 20 cases in which the tumours showed variable EphA2 and EphrinA-1protein expressions were used for laser capture microdissection and processed for RT-PCR detection of EphA2 and EphrinA-1 mRNA. Among the 173 oesophageal squamous cell carcinomas, 33 (19.1%) were negative, 44 (25.4%) weakly-positive, 58 (33.5%) moderately-positive and 38 (22.0%) strongly-positive for EphA2 immunostaining. For EphrinA-1 protein expression, 27 tumours (15.6 %) were negative, 41 (23.7 %) weakly-positive, 80 (46.2 %) moderately-positive and 25 (14.5%) strongly-positive. EphA2 and EphrinA-1 were often co-localized in the same tumour areas and vascular endothelial cells. Variable amounts of EphA2 and EphrinA-1 mRNAs were observed in the 20 tumours analysed. No significant association was observed between EphA2 and EphrinA-1 protein expressions and age, tumour location, tumour size, histological differentiation or clinical stage. However, there was a significant correlation between EphA2 expression and lymph node metastases (p < 0.001). In univariate analysis, high levels of EphA2/EphrinA-1 protein expression, higher number of lymph node metastasis, higher histological grade and clinical stage were significantly associated with shorter overall survival. In Cox multivariate analysis, only EphA2, number of lymph node metastasis and clinical stage were of independent significance. We conclude that EphA2 protein expression is confirmed to be of predictive value for unfavourable survival for oesophageal cancer patients and may be a good target for oesophageal cancer therapy.
Expression and Correlation of Hypoxia-Inducible Factor-La and EphA2 in Esophageal Squamous Cell Carcinoma

Abstract:Objective To investigate the expressions and the correlation of hypoxia-inducible factor(HIF)-1α and EphA2 in esophageal squamous cell carcinoma.Methods The subcellular localizations of HIF-1α and EphA2 in Eca109 cell line were characterized by immunofluorescence staining.The expressions of HIF-1α and EphA2 in human esophageal squamous cell carcinoma tissues were detected by immunohistochemistry.Western blot was used to detect the expression of EphA2 in Eca109 cell line with knock-down HIF-1α gene and in TEl3 cell line under normoxia and hypoxia in vitro.Results ① HIF-1α and EphA2 could be detected in cytoplasm in Eca109.② The positive rates of HIF-1α and EphA2 were significantly higher in esophageal squamous cell carcinoma tissues than those in normal esophagus tissues (70.73% vs 0%,P<0.05;78.04% vs 28%,P<0.05).The correlation analysis indicated that the expression of EphA2 had a positive correlation with that of HIF-1α in esophageal squamous eell carcinoma.③ Western blot showed that the expression of EphA2 in Eca109 and TE13 was inhibited with silencing HIF-1α.Conclusions HIF-1α and EphA2 are overexpressed in esophageal squamous cell carcinoma tissues.The expression of EphA2 protein positively correlates with expression of HIF-1α protein.EphA2 may be a potential target of HIF-1α.

Ephrin-A3 Promotes Hepatocellular Carcinoma Cell Proliferation and Metastasis by Interacting with EphA2

Abstract:Background: Emerging evidence suggests that Eph/ephrin-system involves different facets of tumorigenesis and cancer progression. Whether ephrin-A3, a cognate ligand to several Eph receptors, plays a role in HCC cell proliferation and metastasis remains unclear. Methods: The expression of ephrin-A3 was detected by immunohistochemistry and qRT-PCR in human HCC tissues. The clinical significance of ephrin-A3 was analyzed using multiple databases. The biological functions of ephrin-A3 were investigated in vitro and in vivo. Immunofluorescence and co-immunoprecipitation were used to detect the interaction between ephrin-A3 and EphA2. Findings: Elevated expression of ephrin-A3 was positively correlated with the histological grade, metastasis and poor prognosis in human HCC. Knockdown of ephrin-A3 led to significantly decreased proliferative activity and metastatic potential, and re-introduction of ephrin-A3 rescued the decreased proliferation, migration, and invasion induced by ephrin-A3 knockdown in HCC cells. Ephrin-A3 interacted with EphA2 and positively regulated EphA2 and AKT phosphorylation. Knockdown of EphA2 attenuated HCC cell proliferation, migration, and invasion induced by ephrin-A3 overexpression. Furthermore, ephrin-A3-induced EphA2 phosphorylation is both a substrate and a positive regulator of AKT. In HCC tissues, ephrin-A3 expression was positively correlated with EphA2, and patients with high co-expression of ephrin-A3 and EphA2 exhibited poorer prognosis. Interpretation: Ephrin-A3 promotes HCC metastasis by interacting with EphA2 and activating EphA2 and AKT. Thus, our study implicates ephrin-A3 and EphA2 as potential prognostic biomarkers, and provides valuable information for HCC prognosis and treatment. The status of the CpG island associated with the promoter region of EphA1 was assessed by methylation-specific PCR (MSP), and EphA1 protein expression of was examined by immunohistochemical staining (IHC). Down-regulation of EphA1 transcripts was detected in 34% of the cases, up-regulation in 25% of the cases, and no difference in expression in 41% of the cases. The EphA1 transcript expression level was associated with tumor size (P=0.05), stage (P=0.001), and lymph node metastasis (P=0.011). Methylated EphA1 DNA was detected in most of the carcinomas with EphA1 down-regulation that were examined. EphA1 protein expression was associated with depth of wall invasion (P=0.069), stage (P<0.001), and lymph node metastasis (P=0.018). The survival analysis showed that patients whose tumor exhibited EphA1 up-regulation had a poorer outcome than those whose tumor exhibited down-regulation (P=0.005) or no difference in expression (P=0.003). EphA1 may have roles in invasion and metastasis by gastric carcinoma. The EphA1 expression level is a potential prognostic marker in gastric carcinoma, and the EphA1 gene may provide a novel target of therapy for gastric carcinoma.

Abstract:Objective:Eph is the largest member of the family of receptor tyrosine kinases (RTK). Hypermethylation of CpG island is an important mechanism leading to down-regulation of EphA7 in gastric cancer. The objective of this study is to investigate the expression level of EphA7 in gastric carcinoma and its roles in the development,progression and prognosis of gastric carcinoma. Methods:The expression level of EphA7 transcript in gastric carcinoma specimens and normal mucosa tissues was detected using real-time RT-PCR. Results:The expression level of EphA7 in gastric carcinoma was divided into three groups according to the expression rate in gastric cancer tissue and matched normal mucosa. Statistics analysis showed that the expression of EphA7 is significantly related to age and stage. Overexpression of EphA7 has a tendency to relate to metastasis. Conclusion:EphA7 is differentially expressed in gastric carcinoma specimens. Overexpression of EphA7 may play a role in the development and progress of gastric carcinoma.
The Expressions of EphB4 and Ephrinb2 in Lung Adenocarcinomas: a High Level of the EphB4 Protein is Associated with Lymph Node Metastasis.

Abstract:Eph tyrosine kinase receptors and their ephrin ligands are involved in normal development and tumorigenesis. Eph receptors are frequently over expressed in a wide variety of human malignant tumors, being associated with tumor growth, invasion, angiogenesis, and metastasis. This study aimed to evaluate the clinical significance of EphB4 and its ligand of ephrinB2 protein expressions in human lung adenocarcinomas. EphB4 and ephrinB2 protein expressions were assessed immunohistochemically on paraffin embedded tissues obtained from 93 patients with lung adenocarcinoma. Fifty-one out of 93 (54.8%) specimens were negative for EphB4 expression, and 42 out of 93 (45.2%) were positive for EphB4 expression. EphrinB2 expression was consistently negative in all tissues. EphB4 expression was significantly associated with tumor differentiation (P = 0.001), lymph node metastasis (P = 0.021), and Ki67 (P = 0.012). No significant relationship was found between EphB4 expression and gender, age, or ALK mutation. Our data show that EphB4 is differentially expressed in lung adenocarcinoma tissues. A high level of EphB4 protein expression was associated with lymph node metastasis in lung adenocarcinoma.
Expression of EphA2 mRNA and VEGF in human hepatocellular carcinoma and their relationship

Abstract:Objective To clarify the expression of EphA2mRNA and VEGF in hepatocellular carcinoma (HCC) and explore their relationship. Methods The expression of EphA2mRNA was determined with in situ hybridization and that of VEGF with immunohistochemistry in cancerous and adjacent tissues from 53 cases of HCC. Results The rate of expression of EphA2mRNA was significantly higher in HCC tissue than in the adjacent tissue (86. 79% vs. 69. 81%, P<0. 001). The rate of expression of VEGF in HCC tissue was 75. 4% (40/53) and there was no expression of VEGF in the adjacent tissue. The expression of EphA2mRNA and VEGF in HCC was markedly correlated with tumor cell grade (P<0. 01 and 0. 05). EphA2mRNA level was positively correlated with VEGF level (P<0. 01). Conclusions EphA2mRNA and VEGF are over-expressed in HCC and the over-expression is closely related to the malignant behaviors of the tumor. Furthermore, there is a significant relationship between the expression of EphA2mRNA and that of VEGF in HCC.

Abstract:The prognostic role of EphA2 in human carcinomas remains controversial. We systematically reviewed the evidence of assessment of EphA2 expression in cancers to help clarify this issue. PubMed, Embase and Web of Science databases were searched to identify eligible studies to evaluate the association of EphA2 expression and overall survival (OS) of cancers. Hazard ratios (HRs) were pooled to estimate the effect. EphA2 overexpression was significantly correlated with poor OS of patients with cancer (HR: 1.94, 95% confidence interval [CI]: 1.65-2.28). Subgroup analysis also indicated a significant relation between EphA2 overexpression and OS in gastric cancer (HR: 1.95, 95% CI: 1.48-2.59). However, there was no significant relation between EphA2 overexpression and OS in lung cancer (HR: 1.30, 95% CI: 0.93-1.83). Our analyses demonstrate that EphA2 overexpression was effectively predictive of worse prognosis in various human carcinomas. For certain cancers, EphA2 might be a marker of poor prognosis in patients with cancer, except for lung cancer.

Abstract:Objective:Eph receptors constitute the largest sub-family of receptor tyrosine kinases(RTKs) and have an important role in oncogenic transformation and tumor progression.This study aimed to analyze the aberrant expression of EphA1 gene in gastric cancer and its correlation with clinical parameters,so as to explore the significance of EphA1 in the etiology and progression in gastric cancer.Methods: Expression of EphA1 protein was determined using immunohistochemical staining in 60 gastric cancer patients.Correlation between EphA1 protein expression and clinical parameters was evaluated by statistics.Results: Over-expression was more commonly observed in more advanced patients(P=0.015) and in those with lymph nodes metastases(P=0.032).There was no correlation between EphA1 expression and other clinical parameters.Conclusion: EphA1 gene could promote the progression of gastric cancer.
