Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:The antifungal activity and efficacy of a novel quinoline compound ZJ5337 synthesized by Zhejiang Chemical Industry Institute Co. Ltd. was evaluated. In vitro, ZJ5337 at 10 -20 mg/L showed high inhibition activity against the appressorium formation of Magnaporthe grisea. Greenhouse experiments showed that ZJ5337 at 50 -100 mg/L exhibited high activity against the diseases caused by M. grisea and Sphaerotheca fuliginea with the efficacy greater than 80%. In addition, the duration of ZJ5337 was up to 15 days. ZJ5337 was unable to be transferred effectively between root and leaf, and among leaves of cucumber. The rain-wash test showed that rain-wash had significant impact on the efficacy of ZJ5337 within three hours, but not after 4 hours of the compound application. Field trials showed that 300-500 mg/L 2. 5%ZJ5337 EC efficacy of 66% -91% against M. grisea and did not show any phytotoxic to the rice tested.

Abstract:To discover new strobilurin analogs with high pesticidal activity, 11 novel methoxy acrylates bearing biaryl methanone oxime ether moieties were synthesized from aroyl pyridines, and characterized by H-1 and C-13 NMR and HRMS. In vitro fungicidal evaluation indicated that all of the target compounds exhibited excellent activities against tested seven phytopathogenic fungi, and compounds 5d and 5g displayed comparable overall activities to azoxystrobin and trifloxystrobin. Some of the target compounds also exhibited excellent in vivo control effects at 400 mg/L, for example, compound 5h could 100% control wheat powdery mildew and cucumber anthracnose. Furthermore, at 25 mg/L, these compounds, including control agents, completely inhibited spores germination of rice blast, but no effect at all for cucumber gray mold. These results suggested that this class of compounds might deserve to be developed as new potential agricultural fungicides.

Abstract:Sulfonyl-containing compounds, which exhibit a broad spectrum of biological activities, comprise a substantial proportion of and play a vital role, not only in medicines but also in agrochemicals. As a result increasing attention has been paid to the research and development of sulfonyl derivatives. A series of thirty-eight 2-substituted phenyl-2-oxo- III, 2-hydroxy- IV and 2-acyloxyethylsulfonamides V were obtained and their structures confirmed by IR, ¹H-NMR, and elemental analysis. The in vitro and in vivo bioactivities against two Botrytis cinerea strains, DL-11 and HLD-15, which differ in their sensitivity to procymidone, were evaluated. The in vitro activity results showed that the EC50 values of compounds V-1 and V-9 were 0.10, 0.01 mg L-1 against the sensitive strain DL-11 and 3.32, 7.72 mg L-1 against the resistant strain HLD-15, respectively. For in vivo activity against B. cinerea, compound V-13 and V-14 showed better control effect than the commercial fungicides procymidone and pyrimethanil. The further in vitro bioassay showed that compounds III, IV and V had broad fungicidal spectra against different phytopathogenic fungi. Most of the title compounds showed high fungicidal activities, which could be used as lead compounds for further developing novel fungicidal compounds against Botrytis cinerea.

Abstract:Two series of N-pyridylpyrazole derivatives containing 1,2,3-triazole and quinazolin-4(3H)-one substructures were designed and synthesized. In total, 18 novel compounds were prepared, and all compounds were characterized by H-1 NMR, C-13 NMR and elemental analysis (EA). Preliminary bioassay results revealed that a few of new compounds with quinazolin-4(3H)-one moiety exhibited good insecticidal activity against the oriental armyworm (Mythimna separata). In addition, the compounds Ia-h with 1, 2, 3-triazole moiety showed broad-spectrum antifungal activities against Fusarium oxysporum f.sp.cucumerinum, Cercospora arachidicola Hori, Botryosphaeria dothidea, Alternaria solani, Gibberella zeae and Phytophthora capsici at 50 [mu g/mL concentration. The EC50 values of Ib and If against Botryosphaeria dothidea were 13.9 and 11.0 mu g/mL, respectively, which were comparable to Chlorothalonil. These results indicated the potential application of N-pyridylpyrazole derivatives as fungicide in further study.

Abstract:Twenty-one hexahydropyrrolidoindole alkaloids were designed and synthesised via acylation reaction at the 3-N position from the commercially available indole-3-acetonitrile as the starting material in excellent yields. The effects of all target compounds against Verticillium dahlia , Fusarium oxysperium sp. , Cytospora juglandis , Aspergillu sflavu , Aspergillus niger and Fusarium oxysporum were determined. The results of bioassays indicated that the majority of tested compounds displayed comparable or better in vitro bioactivity than the positive control. Notably, compounds 8 and 17 revealed potent activity against C. juglandis and A. sflavu , both with the same minimum inhibitory concentration value of 1.9 μg mL −1 , which has fungicidal activity far exceeded that of amphotericin B and chlorothalonil .

Abstract:Diseases caused by plant viruses and pathogens pose a serious threat to crop yield and quality. Traditional pesticides have gradually developed drug resistance and brought certain environmental safety issues during long-term overuse. There is an urgent need to discover new candidate compounds to address these issues. In this study, we achieved the efficient synthesis of iheyamine A and its derivatives, and discovered their excellent antiviral activities against tobacco mosaic virus (TMV). Most compounds displayed higher antiviral activities against TMV than commercial ribavirin at 500 μg/mL, with compounds 3a (Inactive effect IC50: 162 µg/mL), 3d (Inactive effect IC50: 249 µg/mL), 6p (Inactive effect IC50: 254 µg/mL), and 7a (Inactive effect IC50: 234 µg/mL) exhibiting better antiviral activities than ningnanmycin at 500 μg/mL (Inactive effect IC50: 269 µg/mL). Meanwhile, the structure-activity relationships of this type of compounds were systematically studied. We chose 3a for further antiviral mechanism research and found that it can directly act on viral coat protein (CP). The interaction of 3a and CP was further verified via molecular docking. These compounds also showed broad-spectrum fungicidal activities against 8 plant pathogenic fungi, especially for P. piricola. This study provides a reference for the role of iheyamine alkaloids in combating plant pathogenic diseases.

Abstract:Background: Fungal diseases present a significant threat to global agriculture, necessitating the development of new, safe, and effective fungicides. Existing fungicides face resistance and health risks, prompting the synthesis of novel compounds. Researchers have synthesized aldehyde-based thiourea and thiazolyl hydrazine derivatives, evaluating their antifungal activities to identify impactful pesticide molecules. Results: The results showed that most of the compounds had broad-spectrum antifungal activity against six plant pathogenic fungi and four post-harvest fungi. Notably, compound LN18 showed the best antifungal activity against Monilinia fructicola with a half-maximal effective concentration (EC50) of 0.17 μg mL-1, which was better than the commercial fungicide natamycin. A structure-activity relationship (SAR) study showed that the presence of unsaturated double bonds in the structure and the length of the carbon chain were the main factors affecting antifungal activity. The presence of unsaturated double bonds and an increase in the length of the carbon chain greatly improved inhibitory activity against the tested pathogens. The preliminary mechanism study showed that LN18 could damage the integrity of the mycelial plasma membrane, leading to leakage of intracellular nucleic acid and protein. LN18 also induced an increase in the intracellular reactive oxygen species level to exert its antifungal effects. In addition, compound LN18 had a stronger antifungal effect in vivo, and better phytotoxicity than natamycin, indicating broad application prospects in agriculture. Conclusion: Aldehydes-thiourea and thiazolyl hydrazine derivatives demonstrate remarkable antifungal efficacy against plant pathogenic and post-harvest fungi, offering a promising avenue for commercialization as highly efficacious, cost-effective and safe antifungal agents. © 2024 Society of Chemical Industry.

Abstract:To develop new fungicides with high efficiency, 46 novel sulfonamide derivatives were designed and synthesized by introducing pinacolone fragment into chesulfamide which was used as lead compound. All compounds were characterized by 1H NMR, 13C NMR, and MS spectra, and the structure of compound P-27 was also confirmed by X-ray single crystal diffraction. It was found that a variety of compounds present excellent inhibitory effect against Botrytis cinerea. The inhibition rates of P-29 on tomato and strawberry were 90.24% (200 mg/L) and 100% (400 mg/L) in vivo respectively, which were better than the lead compound chesulfamide (59.23% on tomato seedlings and 29.63% on strawberries).

Abstract:(E)-3-(2-chlorophenyl)-1-(2,4-dichlorophenyl) prop-2-en-1-one was prepared from 2-chlorobenzaldehyde followed by cyclization with hydrazine monohydrate. Eight new 3-(2,4-Di-chlorophenyl)-5-(2-chlorophenyl)-4,5-dihydro-N-acylpyrazole derivatives were synthesized and characterized by elemental analysis, IR and 1H NMR spectroscopy. The experimental results show that the inhibition ratio of compounds 3f towards H. Oryzae and P. oryzae at 50 mg·L−1 is 55.2% and 57.1%, respectively. The inhibition ratio of compounds 3g towards H. Oryzae, P. oryza, S. Sclerotiorum at 50 mg·L−1 is 53.3%, 60.0%, 50.4% respectively.

Abstract:In an attempt to search for potent fungicide, a series of novel N-aryl-1H-pyrazole-5-carboxylate derivatives was designed and synthesized. Their chemical structures were characterized by 1H NMR spectra and high resolution mass spectrometry(HRMS). The preliminary bioassay results indicated that some target compounds displayed better fungicidal activities against certain fungi at 50 µg/mL or favorable antitumor activities at 5 µg/mL compared with chlorothalonil and 5-fluorouracil, respectively. The structure-activity relationship demonstrated that the introduction of ester group and amide bond was favorable to the improvement of activities against Physalospora piricola and Phytophthora capsici.

Abstract:Unrestricted reproduction and spread of pest had caused great damage to the quality and yield of crops in recent years. Besides the use of traditional chemical pesticides, natural products also make a huge contribution against pests. Chasmanthinine, a diterpenoid alkaloid isolated from Aconitum franchetii var. villosulum , shown extremely antifeedant activity against Spodoptera exigua . Therefore, a series of novel Chasmanthinine derivatives were synthesized and their biological activity was studied in this work. Compound 33 showed the strongest antifeedant activity (EC 50 = 0.10 mg/cm 2 ) among all the test compounds. The mechanism research of 33 revealed that its antifeedant effect was related to the inhibition of carboxylesterase (CES), and proved the thiophene acyl group could form a strong binding effect with CES by molecular docking. Moreover, compound 10 exhibited the strongest cytotoxicity (IC 50 = 12.87 μM) against Sf9 cell line and moderate contact toxicity. The mechanism research indicated that compound 10 could induce Sf9 cells apoptosis. In summary, the results lay a foundation for the application of diterpene alkaloids in plant protection.

Abstract:A series of new 5-(2-ethylcyclohexylidene)-2-aminoimidazolin-4-one derivatives 3a-3f were synthesized via Knoevenagel condensation,methylation and substitution reaction using 2-ethylcyclohexanone and 2-thiohydantoin as starting material.Their structures were confirmed by 1H NMR,IR and MS spectral data.The preliminary bioassay showed that some of the compounds have good fungicidal activity against Sclerotinia scleotiorum at the concentration of 50 μg/mL.The EC50 values of compound 3a,3c,3e and 3f against S.scleotiorum were 12.10,8.73,10.11 and 7.67 μg/mL,respectively.

Abstract:Olefin-imidacloprid is a highly active metabolite of imidacloprid. To solve the problem of high bee toxicity of olefin-imidacloprid, according to the bioisosterism, a series of novel olefin-imidacloprid mimics containing 1,3-disubstituted1,2,4-triazoline were designed and synthesized. The insecticidal activity of these compounds against A. craccivora, Nilaparvata lugens and F. occidentalis and in vitro antifungal activity were determined. The results showed that these compounds showed certain insecticidal activity at 400 mu g/mL and 100% lethal rate was obtained from compound II-7 at 100 mu g/mL against F. occidentalis. The toxicity of some compounds to bees was predicted by admetSAR, and they showed low level of toxicity. On the other hand, the target compounds showed different degrees of antifungal activity against 9 plant pathogenic fungi at the concentration of 50 mu g/mL. Compound I-15 exhibited the highest activity with the inhibition rate higher or equal to hymexazol against five pathogens. The structure-activity relationship analysis showed that the compounds bearing alkyl substituent at site 3 in triazoline unit displayed higher insecticidal activity than that of an aryl substitution, meanwhile, the more lipophilic groups, the higher fungicidal activity. The substitution of pyridine ring or thiazole ring at site 1 in triazoline unit had little effect on their antifungal activity. The molecular docking results showed that the binding pattern of compound II-7 to acetylcholine binding protein (AChBP) was similar to that of imidacloprid (IMI), indicating that these compounds also had the potential to bind to AChBP, which provided a new reference for further molecular optimization.

Abstract:A series of 2-(3-(trifluoromethyl)-5-(alkoxy)-1H-pyrazol-1-yl)-4-aryloxypyrimidine derivatives were designed and synthesized. The structures of all the title compounds were confirmed by H-1 NMR and elementary analysis. These compounds were screened for herbicidal activity against rape and barnyard grass. Compound B13 exhibited moderate herbicidal activity.

Abstract:Imidazole alkaloids, a common class of five-membered aromatic heterocyclic compounds, exist widely in plants, animals and marine organisms. Because of imidazole's extensive and excellent biological and pharmacological activities, it has always been a topic of major interest for researchers and has been widely used as an active moiety in search of bioactive molecules. To find more efficient antibacterial compounds, a series of novel imidazole-fragment-decorated 2-(pyrazol-4-yl)-1,3,4-oxadiazoles were designed and synthesized based on our previous works via the active substructure splicing principle, and their bioactivities were systematically evaluated both in vitro and in vivo. The bioassays showed that some of the target compounds displayed excellent in vitro antibacterial activity toward three virulent phytopathogenic bacteria, including Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas axonopodis pv. citri (Xac) and Pseudomonas syringae pv. actinidiae (Psa), affording the lowest EC50 values of 7.40 (7c), 5.44 (9a) and 12.85 (9a) μg/mL, respectively. Meanwhile, compound 7c possessed good in vivo protective and curative activities to manage rice bacterial leaf blight at 200 μg/mL, with control efficacies of 47.34% and 41.18%, respectively. Furthermore, compound 9a showed commendable in vivo protective and curative activities to manage kiwifruit bacterial canker at 200 μg/mL, with control efficacies of 46.05% and 32.89%, respectively, which were much better than those of the commercial bactericide TC (31.58% and 17.11%, respectively). In addition, the antibacterial mechanism suggested that these new types of title compounds could negatively impact the cell membranes of phytopathogenic bacteria cells and cause the leakage of the intracellular component, thereby leading to the killing of bacteria. All these findings confirm that novel 2-(pyrazol-4-yl)-1,3,4-oxadiazoles containing an imidazole fragment are promising lead compounds for discovering new bactericidal agents.

Abstract:Ten novel benzotriazinone-3-anime derivatives were designed and synthesized,and their structures were confirmed by 1H NMR and MS spectra.The preliminary bioassay results showed that the title compounds possessed insecticidal activity.At the concentration of 500 mg / L,compands 4c,4d and 7c showed more than 80% insecticidal activities against Mythimna separata;at 500 mg / L,compand 7d showed more than 80 % insecticidal activity against Aphis fabae and more than 70% insecticidal activity against Tetranchus urticae.
