Abstract:AIM: To study the protective effect of the ginkgo biloba (EGB) extract on liver from experimental type 2 diabetic rats and to explore its possible mechanism. METHODS: Thirty-nine male Sprague-Dawley rats were divided randomly into four groups: normal control group, high-fat group, diabetic group and EGB-treated group. After fed with high-fat diet for 4 weeks, the later two groups were injected with streptozotocin intraperitoneally to induce type 2 diabetes mellitus. EGB-treated group was injected intraperitoneally with EGB at a dose of 8 mg·kg-1·d-1, and the other three groups were treated with normal saline of the same volume. After 8 weeks, the morphologic change of hepatic tissue was observed under transmission electron microscope (TEM) and light microscope (LM), respectively. In addition, the activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX), total nitric oxide synthase (TNOS), inducable nitric oxide synthase (iNOS) and the content of malondialdehyde (MDA), nitric oxide (NO) in liver homogenate were detected biochemically. RESULTS: Obvious liver fatty degeneration, apparent decrease of glycogen granules in cytoplasm of hepatocytes under light microscope and hepatocytes pyknosis, lots of lipid deposits in cytoplasm of hepatocytes, proliferation of hepatic stellate cells and collagen under TEM were observed in diabetic group. The activity of SOD, CAT, GSH-PX decreased but the activity of tNOS, iNOS and the content of MDA, NO-2/NO-3 increased in diabetic group compared with normal control group. The pathological change was relieved in EGB-treated group. The activity of SOD, CAT, GSH-PX increased, the activity of tNOS, iNOS and the content of MDA, NO-2/NO-3 decreased in the liver of rats in EGB-treated group compared with diabetic group. CONCLUSION: EGB exerts a beneficial effect on liver in experimental type 2 diabetic rats. Anti-lipid peroxidation and suppression of NO production may be involved in this process.

Abstract:The prevalence of degenerative disorders in public health has promoted in-depth investigations of the underlying pathogenesis and the development of new treatment drugs. Ginkgo biloba leaves extract (EGb) is obtained from Ginkgo biloba leaves and has been used for thousands of years. In recent decades, both basic and clinical studies have established the effects of EGb. It is widely used in various degenerative diseases such as cerebrovascular disease, Alzheimer’s disease, macroangiopathy and more. Here, we reviewed several pharmacological mechanisms of EGb, including its antioxidant properties, prevention of mitochondrial dysfunctions, and effect on apoptosis. We also described some clinical applications of EGb, such as its effect on neuro and cardiovascular protection, and anticancer properties. The above biological functions of EGb are mainly focused on aging-related disorders, but its effect on other diseases remains unclear. Thus, through this review, we aim to encourage further studies on EGb and discover more potential applications

Abstract:The current study aimed to evaluate the neuroprotective effect of Ginkgo biloba extract (GbE) on the progression of acute cerebral ischemia-reperfusion injury in diabetic rats, and to determine the molecular mechanism associated with this effect. Streptozotocin (STZ) induced diabetic rats were pretreated with GbE (50, 100 and 200 mg/kg/day; intragastric) for 3 weeks. During this period, body weight changes and fasting blood glucose levels were assessed each week. Following pretreatment, rats were subjected to suture occlusion of the middle cerebral artery for 30 min, which was followed by 24 h of reperfusion. Neurological deficits were subsequently evaluated at 2 and 24 h following reperfusion. Rats were sacrificed after 24 h reperfusion, and infarct volume and S100B content were measured to evaluate the neuroprotective effect of GbE. The results of the present study demonstrated that GbE pretreatment improved neurological scores, and reduced cerebral infarct volume and S100B content. Oxidative stress markers, including glutathione (GSH) and superoxide dismutase (SOD) were increased, and malondialdehyde (MDA) contents were reduced following GbE treatment. The levels of p-Akt, p-mTOR and glutamate transporter 1 (GLT1) were observed to be increased in GbE-pretreated rats. These results indicated that GbE pretreatment may serve a protective role against cerebral ischemia-reperfusion injury in diabetic rats by inhibiting oxidative stress reaction, upregulating the expression of Akt/mTOR and promoting GLT1 expression. In conclusion, the current study revealed the protective role and molecular mechanisms of GbE in diabetic rats with cerebral ischemia-reperfusion injury, and may provide novel insight into the future clinical treatment of this condition.

Abstract:Aim To evaluate the effect of ginkgolide B treatment on vascular endothelial function in diabetic rats. Methods The study included four groups with 15 male Sprague-Dawley rats: control group; control group treated with ginkgolide B; diabetic group; and diabetic treated with ginkgolide B. The activity of superoxide dismutase (SOD), malondialdehyde content, and nicotinamide adenine dinucleotide phosphate (NADPH) oxidase subunits, and glutathione peroxidase 1 (GPX1) protein expression were determined in aortic tissues. Vasoconstriction to phenylephrine (PHE) and vasorelaxation to acetylcholine (Ach) and sodium nitroprusside (SNP) were assessed in aortic rings. Nitric oxide (NO) and hydrogen sulfide (H2S) were measured, as well as cystathionine gamma lyase (CSE) and cystathionine beta synthetase (CBS) protein expression, and endothelial nitric oxide synthase (eNOS) activity. Results Diabetes significantly impaired PHE-induced vasoconstriction and Ach-induced vasorelaxation (P < 0.001), reduced NO bioavailability and H2S production (P < 0.001), SOD activity, and GPX1 protein expression (P < 0.001), and increased malondialdehyde content and NADPH oxidase subunits, and CSE and CBS protein expression (P < 0.001). Ginkgolide B treatment improved PHE vasoconstriction and Ach vasorelaxation (P < 0.001), restored SOD (P = 0.005) and eNOS (P < 0.001) activities, H2S production (P = 0.044) and decreased malondialdehyde content (P = 0.014). Vasorelaxation to SNP was not significantly different in control and diabetic rats with or without ginkgolide B treatment. Besides, ginkgolide B increased GPX1 protein expression and reduced NADPH oxidase subunits, CBS and CSE protein expression. Conclusion Ginkgolide B alleviates endothelial dysfunction by reducing oxidative stress and elevating NO bioavailability and H2S production in diabetic rats.
Therapeutic Effect of Gink Gobioba Extract on Diabetic Cardiomyopathy Induced by SZT and ISO in Rats

Abstract:Objective To investigate the therapeutic effect of gink gobioba extract(GBE) on diabetic cardiomyopathy(DCM) induced by streptozotocin(SZT) and isoprenaline(ISO) in rats.Methods 20 male Wistar rats were randomly divided into normal control group(n=5),model group(n=5),positive control group(n=5),and treatment group(n=5).The rats in normal control group were given nothing and the other rats were given STZ and ISO to construct DCM rat models.The rats in model group were given saline and the rats in positive control group and treatment group were given benazepril and GBE,respectively.The body weight,blood sugar level,serum AST and LDH levels,expression level of TGF-β1 protein,and morphology of myocardium of rats in various groups were observed.Results 1 and 2 weeks after treatment,the body weight and blood sugar level of the rats in model group were lower than those in normal control group(P0.05).The body weights and blood sugar levels of the rats in positive control and treatment group were lower than those in normal control group(P0.05).The levels of AST,LDH,and TGF-β1 in model group were higher than those in normal group(P0.05);and the levels of AST,LDH,and TGF-β1 in positive control and treatment group were higher than those in model group(P0.05).The myocardial cells of the rats in normal control group had no swelling.The cells in model group showed clear boundaries and more scattered necrosis,and there were lots of fibroblasts and some collagen fibers in focal necrosis.The myocardial cells of the rats in treatment group had smaller focal necrosis and lower fibrosis.Conclusion GBE can alleviate myocardial injury induced by STZ plus ISO of rats.
Antihyperglycemic Effect Ofginkgo Bilobaextract in Streptozotocin-Induced Diabetes in Rats

Abstract:TheGinkgo bilobaextract (GBE) has been reported to have a wide range of health benefits in traditional Chinese medicine. The aim of this study was to evaluate the antihyperglycemic effects of GBE on streptozotocin- (STZ-) induced diabetes in rats. Diabetes was induced in maleWistarrats by the administration of STZ (60 mg/kg b.w.) intraperitoneally. GBE (100, 200, and 300 mg/kg b.w.) was administered orally once a day for a period of 30 days. Body weight and blood glucose levels were determined in different experimental days. Serum lipid profile and antioxidant enzymes in hepatic and pancreatic tissue were measured at the end of the experimental period. Significant decreases in body weight and antioxidant ability and increases in blood glucose, lipid profile, and lipid peroxidation were observed in STZ-induced diabetic rats. The administration of GBE and glibenclamide daily for 30 days in STZ-induced diabetic rats reversed the above parameters significantly. GBE possesses antihyperglycemic, antioxidant, and antihyperlipidemia activities in STZ-induced chronic diabetic rats, which promisingly support the use of GBE as a food supplement or an adjunct treatment for diabetics.
The Different Effect of Ginkgo Biloba Extract, Ginkgolides B, Atorvastatin on Endothelial Cells

Abstract:Cultured human umbilical vein endothelial cells(HUVECs) were exposured to 10% diabetic serum(DS),and then treated with Ginkgo biloba extract(GBE),ginkgolides B(GB),atorvastatin(AT) for 30 minutes,3 hours,and 3 days respectively.The survival rate of the cells and the level of Lactate dehydrogenase(LDH),maleic dialdehyde(MDA),nitric oxide(NO) in the supernatant were detected.The survival rate of HUVECs cells decreased significantly after 3 days' treatment with 10% DS(P0.05).Drugs and the time of interfering showed crossover effect on the levels of MDA and LDH.The levels of NO in AT,GBE and GB groups were higher than those in DS group(P0.05),especially in GBE group.HUVECs were vulnerable to DS.AT,GB and GBE protected HUVECs against the damage induced by DS.GBE demonstrated the most obvious protection effects.

Abstract:BACKGROUND:Diabetic gastrointestinal dysfunction (DGD) is a common complication in diabetic patients, and enteric glial cells (EGCs) found in the gastrointestinal tract have been shown to play an essential role in gastrointestinal dysfunction. Thus, targeting EGCs may be helpful for the control of DGD. This study aimed to evaluate the protective effect of Ginkgo biloba extract (GBE) from G. biloba dropping pills against hyperglycaemic stress-induced EGCs injury and its underlying mechanism.METHODS:In vitro, the protective effect of GBE on CRL-2690 cells was evaluated by MTT assay and TUNEL assay. The expression of related markers was evaluated by RNA sequencing and validated by using western blotting. In vivo, STZ-induced C57BL/6J WT mice were used as models to evaluate the effects of GBE on blood glucose, body weight, and EGCs' activity and relevant signalling pathways were validated by immunofluorescence.RESULTS:The results showed that GBE (25 μg/ml) treatment significantly attenuated hyperglycaemic stress-induced cytotoxicity and cell apoptosis in CRL-2690 cells, which was verified in an STZ-induced (100 mg/kg, 3 days) diabetic mouse model with continuous GBE administration (25/100 mg/kg/day, 6/12 weeks). Further mechanistic study based on transcriptomic data revealed that GBE exerted its beneficial effect by regulating immune-related pathways, and TLR2/BTK/NF-κB/IL-1α/IL-10 comprised the main targets of this drug.CONCLUSIONS:This study demonstrates the protective effect of GBE against hyperglycaemic stress-induced EGCs injury using both in vitro and in vivo models and further reveals that the effect was achieved by targeting TLR2 and its downstream molecules BTK/NF-κB/IL-1α/IL-10. This study may be helpful for expanding the clinical application of GBE in treating DGD.
Improvement Of Ginkgolide B On Diabetic Nephropathy In Streptozotocin (Stz)-Induced Rats

Abstract:Aim: The goal of this study was to explore ginkgolide B as a therapy for amelioration of diabetic nephropathy. Methods: Healthy male Sprague-Dawley rats were randomly assigned to four groups: the control group, control treatment group, diabetes group, and the diabetes treatment group. Rats from the diabetes group and the diabetes treatment group were intraperitoneally injected with STZ (70 mg/kg) to induce diabetes. After 3 days, rats from the control and diabetes treatment groups received ginkgolide B, and animals of the control and diabetes groups received control diet. After administration of ginkgolide B for 8 weeks, the kidneys were collected for histological examination and analysis of Western blot. Blood samples were used to determine renal function. Results: Compared with the control group, levels of blood urea nitrogen (BUN), creatinine, and malondialdehyde (MDA) in the serum were significantly increased in the diabetes group, and activity of superoxide dismutase (SOD) was decreased. Expression of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-alpha, interleukin (IL)-6 was increased in the diabetes group. Treatment with ginkgolide B significantly reduced the levels of blood urea nitrogen (BUN), creatinine, and MDA in serum, and 24 hour urea protein. Ginkgolide B significantly decreased expression of TNF-alpha, IL-6, transforming growth factor (TGF)-beta, and cyclooxygenase (COX)-2 in the kidney from rats in the diabetes treatment group compared with the diabetes group. Levels of heme oxygenase (HO)-1 and Nrf-2 were increased in kidney in the diabetes treatment group compared with the diabetes group. Conclusion: The results suggest that ginkgolide B ameliorates diabetic nephropathy via improving antioxidant and anti-inflammatory functions, and easing kidney fibrosis.

Abstract:Objective:To study the effect of Ginkgo biloba extract on serum insulin and glucose metabolism in the aged with impaired glucose tolerance(IGT) and discuss the role and mechanism of Ginkgo biloba extract in ameliorating insulin resistance.Method:Ginkgo biloba extract——Ginaton was given via vein one time per day for 28 days.TC,TG,VLDL,serum glucose and insulin of individual spots in 75g oral glucose tolerance test(OGTT)were measured before and after treatment respectively.Result:The level of serum glucose and insulin of the individual spots such as fasting,postprandial 1 hour and postprandial 2 hours all descended significantly in 68 treated cases(P0.05).The level of TG and VLDL also descended significantly in those cases(P0.05).Multivariate regressive analysis showed that TG and VLDL were significantly relative with serum glucose and insulin of the individual spots in OGTT.Conclusion:Ginkgo biloba extract can ameliorate glucose metabolism and hyperinsulinism of the aged with IGT by adjusting the level serum lipid(especial TG and VLDL).Debasing the level of lipid(especial free fatty acids) is a way for ameliorating IGT of the aged.

Abstract:BACKGROUND: <i>Ginkgo biloba</i> extract 50 (GBE50) has a variety of pharmacological functions such as anti-inflammatory, antioxidant and maintenance of glucose and lipid metabolism homeostasis. However, the therapeutic effects and mechanisms of GBE50 on non-alcoholic fatty liver disease (NAFLD) remain unknown. Therefore, in this study, we evaluated the therapeutic effects of GBE50 in NAFLD by using a high-fat diet (HFD) mice model.METHODS: C57BL/6J mice were fed a HFD diet for 15 weeks and were given respectively 25, 50, and 100 mg/kg GBE50 daily by gavage from 3 to 15 weeks. After the administration, blood samples and liver tissues were collected for biochemical detection, histological measurement, immunohistochemistry and Western blot, respectively.RESULTS: We found that GBE50 treatment could ameliorate insulin resistance (IR), glucose intolerance, lipid accumulation, hepatic steatosis and liver injury in HFD-fed mice. Further mechanism exploration discovered that the hepatoprotective effects of GBE50 on NAFLD may be related to the strengthening of IRS-1 signal activation and the weakening of NF-κB, Akt and endoplasmic reticulum stress signals activation.CONCLUSION: GBE50 is a potentially powerful therapeutic agent for the treatment of NAFLD.

Abstract:Objective:This study is to investigate the ef fect of Ginkgo b iloba extract(GbE)on atherosclerotic plaque formation in cholesterol-fed rabbit s and low density lipoprotein(LDL)oxidization in vitro.Method:Cholesterol-fed rabbits were applied to study the effect of Ginkgo bi loba extract on atherosclerotic plaque formation;Low density lipoprotein oxidiz a tion by cupric anion was tested to investigate effects of Ginkgo biloba extract on LDL oxidation.Result:Atherosclerotic plaque fo rmation(plaque area percentage 19.0%±8.2% vs 44.9%±17.6%，P<0.01)was significantly inhibited by GbE(0.5 g/kg/d)；MDA content increase and vitamin E decrease can be prevented by GbE in a dose and time depen dent manner. Conclusion:Ginkgo biloba extract cou ld attenuate the development of atherosclerotic plaque in cholesterol-fed rabbi ts and oxidation of LDL can be inhibited in vitro by Ginkgo biloba extract.
Cardiac Dysfunction is Attenuated by Ginkgolide B Via Reducing Oxidative Stress and Fibrosis in Diabetic Rats

Abstract:Objective(s): Diabetic cardiomyopathy is a leading factor of high morbidity and mortality in diabetic patients. Our previous results revealed that ginkgolide B alleviates endothelial dysfunction in diabetic rats. This study aimed to investigate the effect of ginkgolide B on cardiac dysfunction and its mechanism in diabetic rats.Materials and Methods: Diabetes was induced in rats through the intraperitoneal injection of streptozotocin (STZ). Hemodynamics was monitored to assess cardiac function. Oxidative stress was examined by detecting levels of malondialdehyde (MDA) and superoxide dismutase (SOD) in serum, and expression of sirtuin (SIRT)1, heme oxygenase (HO)-1, and phosphorylated AMPK in the heart. Masson’s trichrome staining and expression of transforming growth factor (TGF)-β1, smooth muscle actin (α-SMA), and phosphorylated (p-) Smad2 and Smad3 were used to evaluate cardiac fibrosis. Inflammatory cytokine in serum and levels of p-PI3K, p-Akt, p-p38, and p-JNK in the heart were determined.Results: Ginkgolide B significantly improved hemodynamics in diabetic rats. Compared with diabetic rats, treatment with ginkgolide B significantly decreased levels of inflammatory cytokines, improved oxidative stress via reducing MDA concentration, and elevating SOD activity in serum and increasing expression of SIRT1, HO-1, and p-AMPK. Further, ginkgolide B alleviated cardiac fibrosis by decreasing expression of TGF-β1, α-SMA, and p-Smad2 and p-Smad3. Meanwhile, ginkgolide B reduced Levels of p-P38, and p-JNK, and increased levels of p-PI3K and p-Akt.Conclusion: The results suggested that ginkgolide B alleviated cardiac dysfunction by reducing oxidative stress and cardiac fibrosis.
Application of GC/MS-based metabonomic profiling in studying the lipid-regulating effects of Ginkgo biloba extract on diet-induced hyperlipidemia in rats

Abstract:Aim: To evaluate the lipid-regulating effects of extract from Ginkgo biloba leaves (EGB) using pharmacological methods and metabonomic profiling in a rat model of diet-induced hyperlipidemia. Methods: EGB was orally administered at a dose level of 40 mg/kg in both the EGB-prevention and -treatment groups. All rat samples obtained were examined for known and potential biomarkers and enzyme activity using commercial assay kits and GC/MS-based metabonomic profiling coupled with principal component analysis (PCA). Results: The data obtained from the assay kits indicated that EGB reduced total cholesterol and low density lipoprotein cholesterol levels and increased high density lipoprotein cholesterol levels in rat plasma obtained from both the EGB-prevention and -treatment groups compared with those of the diet-induced hyperlipidemia group. EGB also increased the activities of lipoprotein lipase and hepatic lipase and excretion of fecal bile acid in rats from the EGB-prevention and-treatment groups. Using GC/MS-based metabonomic analysis, more than 40 endogenous metabolites were identified in rat plasma. PCA of rat plasma samples obtained using GC/MS produced a distinctive separation of the four treatment groups and sampling points within each group. Metabolic changes during hyperlipidemia formation and improvement resulting from EGB treatment were definitively monitored with PCA score plots. Furthermore, elevated levels of sorbitol, tyrosine, glutamine and glucose, and decreased levels of citric acid, galactose, palmitic acid, arachidonic acid, acetic acid, cholesterol, butyrate, creatinine, linoleate, ornithine and proline, were observed in the plasma of rats treated with EGB. Conclusion: EGB exerts multi-directional lipid-lowering effects on the rat metabonome, including limitation of the absorption of cholesterol, inactivation of HMGCoA and favorable regulation of profiles of essential polyunsaturated fatty acid (EFA). Further experiments are warranted to explore the mechanisms of action underlying the lipid-regulating effects of EGB against hyperlipidemia.

Abstract:Grape seed proanthocyanidin extracts (GSPEs) have been reported to be effective in treating arteriosclerosis, while little is known about therapeutic agents against diabetic macrovascular complications. We used streptozocin to induce diabetic rats. GSPEs (250 mg/kg of body weight) were administrated to diabetic rats for 24 weeks. Aortic blood pressure and pulse wave velocity (PWV) were determined in anesthetized rats. Serum glycated hemoglobin and advanced glycation end products (AGEs) were determined. An electronic microscope was used to observe the changes in aortic ultrastructure. Immunohistochemistry was used to evaluate the receptor of advanced glycation end product (RAGE) protein expression in aortic tissue. GSPEs significantly decreased aortic PWV, blood pressure, and aortic medial thickness (P<0.05), and inhibited the migration of vascular smooth muscle cells. GSPEs significantly reduced the AGEs (P<0.05) and the expression of RAGE in aortas of diabetic rats. GSPEs play an important role against diabetic macrovascular complications. This study may provide a new recognition of natural medicine for the treatment of diabetic macrovascular complications.
Intervention Study of Ginkgo Biloba Extract in Rat Model of Lipid-Induced Insulin Resistance

Abstract:Our research focused on the correlation between IR and glucolipid metabolism, vascular endothelial dysfunction and steatohepatitis. Then to provide evidence of Ginkgo on early-stage prevention of IR related diseases in a rat model. Forty male SD rats were equally randomized to four groups: normal group, model group, Ginkgo group and rosiglitazone group. Blood samples and serum biochemical indicators were collected and detected. Insulin sensitive index (ISI) was selected to evaluate the model. Compared with normal group, ISI, NO, T-AOC and SOD significantly decreased, while serum glucose, lipid, ET-1, MDA, and other indicators significantly increased in model group (P < 0.05 or P < 0.01). But compared with model group, it was opposite for both Ginkgo and rosiglitazone groups (P < 0.05 or P < 0.01). Fatty streaks and typical athesclerotic plaques exhibited early-stage injuries of arterial atherosclerosis emerged in the fourth month, while they were improved remarkably in Ginkgo group. The hepatocytes exhibited diffused steatosis, hepatic lobular inflammation and abnormal mitochondria in model group, but were smaller and only tiny lipid droplets were observed in both Ginkgo and rosiglitazone groups. We can make the conclusion IR may lead to endothelial dysfunction and steatohepatitis. (2) Ginkgo biloba extract can improve glucolipid metabolism and may have protective effects on hepatocytes and endothelial function of blood vessels and partly or remarkably reverse endothelial dysfunction and steatohepatitis due to IR, and may have related intervention on IR.
Ginkgo Biloba Extract Suppresses Hypertrophy and Extracellular Matrix Accumulation in Rat Mesangial Cells

Abstract:Aim: To observe the effects of Ginkgo biloba extract (EGb) on the hypertrophy of mesangial cells and the accumulation of extracellular matrix (ECM) in mesangial cells. Methods: Cultured mesangial cells were allotted into 7 groups: normal group, solvent control group, high glucose group, low dose of EGb group, moderate dose of EGb group, high dose of EGb group, and captopril group. Activities of cell antioxidases, S phase percentage and G(0)/G(1) phase percentage, collagen IV and laminin, Smad2/3 and Smad7, TGF-beta(1) mRNA were measured by different methods. Results: For EGb-treated groups, when compared with high glucose group, the cell percentage of S phase was raised and the percentage of G(0)/G(1) was lowered. The intensity of oxidative stress was weakened. The expression of Smad2/3 was greatly decreased and Smad7 was increased. Collagen IV, laminin and TGF-beta(1) mRNA were also reduced. Conclusion: EGb can suppress cell hypertrophy and the accumulation of ECM in rat mesangial cells, which means it could play a vital role in the delay of glomerulosclerosis in diabetic nephropathy.

Abstract:Diabetic cataract is one of the earliest secondary complications of diabetes, and it is characterized by opacification of the eye lens. In this study, we examined the protective effects of Ginkgo biloba extract (EGb761) on rat lenses cultured in high-glucose conditions. The cultured rat lenses were divided into six groups: normal group, high-glucose group, high glucose plus low, medium, and high concentrations of EGb761 groups, and a high glucose plus bendazac lysine group. The activities of antioxidases, aldose reductase, advanced glycosylation end products, transforming growth factor-β2, Smad2/3, E-cadherin, and α-smooth muscle actin were assessed by different methods. Compared with the levels in the high glucose group, EGb761 decreased the intensity of oxidative stress, decreased aldose reductase activation and the level of advanced glycosylation end products, and suppress the transforming growth factor-β2 or Smad pathway activation, further increase the expression of E-cadherin and decrease α-smooth muscle actin, and therefore, prevents the pathological changes of high glucose-induced lens epithelial cells and ameliorated lens opacity. These results suggest that EGb761 has protective effects on several pharmacological targets in the progress of diabetic cataract and is a potential drug for the prevention of diabetic cataract.
Protective effects of extracts of ginkgo biloba leaf on cerebral ischemia/reperfusioninduced oxidative injury in rats

Abstract:Objective We tried to observe the protective effects of extracts of Ginkgo biloba leaf(EGB) on cerebral ischemia/reperfusion induced oxidative injury in rats.Methods Sixty male Wistar rats were randomly divided into following six groups(n=10for each):(a)normal control,(b) injury model,(c)sham operation,(d)low dose of EGB,(e)middle dose of EGB,(f) high dose of EGB.EGB dissolved in distilled in water at dosages of 25,50 and 100mg/kg was given to group d,e,f by gastric tube,continued for 14d;but group a,b,c was only given the equal volume of distilled water at dose of 10ml/kg.At 1 h after the last drug administration,the rats in group b,d,e,f were submitted to incomplete cerebral ischemia(1h)/reperfusion(30min) injury;but group c was only operated and not submitted to cerebral ischemia/reperfusion injury.The cerebral cortex tissue was taken and homogenized for following index assays:SOD and NOS activities,GSH,MDA content and total antioxidative catacity(T-AOC).Results Compared with the normal control,the SOD activity,GSH content and T-AOC in rat brain significantly decreased,but NOS activity and MDA content significantly increased in injury model group.Compared with the injury model,SOD activity,GSH content and T-AOC in rat brain significantly increased,but NOS activity and MDA content were significantly decreased after administration of EGB,especially at dosage of 50 mg/kg.Conclusions EGB has protective effects on cerebral ischemia/reperfusion induced oxidative injury in rats and shared the characteristic of dose-related effect.

Abstract:AIM:To study whether extract of Ginkgo biloba (EGb) can protect against atherosclerosis.METHODS:Confluent monolayers of bovine coronary endothelial cells (BCECs), bovine coronary smooth muscle cells (BCSMCs), and cocultures of the two were incubated with medium containing VEGF and/or EGb, and flux of 125I-labeled oxidized low density lipoprotein (ox-LDL) across the monolayers was measured.RESULTS:Incubation with VEGF significantly increased the permeability of BCEC monolayers to 125I-ox-LDL in a time- and concentration-dependent manner, but had no effect on permeability of BCSMCs or endothelial cells-smooth muscle cells cocultures. EGb significantly inhibited the VEGF-induced hyperpermeability of BCECs.CONCLUSION:VEGF was important in the formation and development of atherosclerosis. The inhibition of VEGF-induced permeability by EGb suggests that extracts of Ginkgo biloba leaves may have important clinical applications in the treatment of cardiovascular diseases.
