Abstract:Objective To investigate the effects of Glossy ganoderma spore oil(GSSO) on the proliferation,apoptosis and migration of human gastric adenocarcinoma cell BGC823.Methods After BGC823 cells were treated with GSSO,the inhibition growth efficacy was determined using cell count kit(CCK-8),and cell apoptosis was analyzed by flow cytometry,cell migrating ability was determined by cell migration assay.Results Compared with control(DMSO solvent),GSSO concentration-dependently inhibited the BGC823 cell proliferation(P<0.05);from the result of flow cytometry,GSSO could induce the apoptosis of BGC823 cell when its concentration was in excess of 0.005 %(P<0.001).GSSO inhibited the BGC823 cell migration significantly when the GSSO's concentration attained to 0.005%(P<0.001).Conclusion GSSO could inhibit the proliferation and cell migration obviously and induced apoptosis of BGC823 cell.
Glossy Ganoderma Spore Oil Induced Apoptosis of Human Lung Adenocarcinoma LTEP-a2 Cells Through Up-regulating Mir-16 Expression

Abstract:Objective To investigate the effects of glossy gnoderma sore oil on the proliferation,apoptosis,miR-16 expression of human lung adenocarcinoma LTEP-a2 cell line,and explore its anti-cancer mechanism. Methods The LTEP-a2 cells were treated with glossy ganoderma spore oil for 24 and 48 hours.The inhibition on cell growth was determined by using cell count kit(CCK-8),cell morphological changes were observed by light microscopy,cell apoptosis was analyzed by flow cytometry,and the expression of miR-16,Bcl2 and PDCD4 were determined by real-time PCR. Results Glossy ganoderma spore oil time-and dose-dependently inhibited the LTEP-a2 cells proliferation;changed the cell morphology obviously after the glossy ganoderma spore oil reached to 2 μL·mL-1.Glossy ganoderma spore oil induced the LTEP-a2 cells apoptosis even at a low concentration by Annexin-V/PI double stained detection;and it up-regulated the expression of miR-16 and down-regulated the bcl2 and VEGF expression significantly. Conclusion Glossy ganoderma spore oil could inhibit the cell proliferation obviously and change the cell morphology,the mechanisms of which should be associated with up-regulating the miR-16 expression and down-regulating the bcl2 and VEGF expression.
Effect of Glossy Ganoderma Spore Oil on Human Hepatoma Cell Line HepG2

Abstract:Objective To investigate the effect of glossy ganoderma spore oil on human hepatoma cell line HepG2 and its mechanism.Methods Cell proliferation was measured by cell count kit(CCK-8).The mobility was evaluated by migration assay.Cell apoptotic rate was examined by flow cytometry(FCM).RT-PCR analysis was used to detect mRNA expression of apoptosis related factors Bcl-2,Bax,p53 and Toll like receptors;the cytoskeleton was observed by the immunofluorescent staining.Results Glossy ganoderma spore oil inhibited the proliferation of HepG2 cells in a dose and time dependent manner.The migrating speed of HepG2 cells was reduced.The apoptotic rate of HepG2 cells increased with the elevated concentrations of glossy ganoderma spore oil.The mRNA expressions of VEGF,Bcl-2 and Bax were down-regulated while p53 expression up-regulated.The mRNA expression of Toll-like receptors was also changed by glossy ganoderma spore oil.The cytoskeleton of HepG2 cells was changed,and the pseudopod was decreased,representing the typical apoptosis appearance.Conclusion Glossy ganoderma spore oil can inhibit the proliferation and the mobility,induce apoptosis of human hepatoma cell line HepG2,change the expression of Toll-like receptors,which may be related to its anti-tumor effects.

Abstract:OBJECTIVE:To investigate the effects of glossy ganoderma spore oil on the proliferation, apoptosis, expression of miR-21 and its target genes of human lung adenocarcinoma SPC-A1 cell line, and to explore its possible mechanism.METHOD:The SPC-A1 cells were treated with glossy ganoderma spore oil for 24 and 48 hours. The inhibition growth efficacy was determined using cell count kit (CCK-8). Cell morphological changes were observed by light microscopy. Cell apoptosis was analyzed by flow cytometry. The expression of miR-21, PTEN and PDCD4 were determined by Real-time PCR.RESULT:Glossy ganoderma spore oil concentration-dependently inhibited the SPC-A1 cell's proliferation. When the concentration of glossy ganoderma spore oil attained to 0.2%, the cells' morphology changed obviously. Glossy ganoderma spore oil could induce the apoptosis of SPC-A1 cells at low concentration. Glossy ganoderma spore oil down-regulated the expression of miR-21 and up-regulated the expression of PTEN and PDCD4 significantly.CONCLUSION:glossy ganoderma spore oil could inhibit the proliferation obviously and cause the changes of cell morphology. Furthermore, glossy ganoderma spore oil induced apoptosis of SPC-A1 cell through down-regulating the expression of miR-21 and up-regulating tumor suppressors.
Glabridin Inhibits Osteosarcoma Migration and Invasion Via Blocking the P38‐ and JNK‐mediated CREB–AP1 Complexes Formation

Abstract:Osteosarcoma is the most common bone malignancy, and it seriously affects the quality of life of affected children and adolescents. Glabridin (GLA), a major component of licorice root extract, has been reported to exert antitumor effects against a variety of tumor types; however, its effects on osteosarcoma have not been elucidated. In the current study, we investigate the effects and potential antimetastatic mechanisms of GLA on osteosarcoma in vitro and in vivo. Flow cytometry showed that GLA induced G2/M cell cycle phase arrest and promoted cell apoptosis. Transwell and wound-healing assays showed that GLA significantly decreased the migration and invasion of osteosarcoma cells. Further western blotting and quantitative real-time polymerase chain reaction showed that the expression of matrix metalloproteinase (MMP)-2 and MMP-9 in MG63 and HOS cells were reduced after GLA treatment. Moreover, western blotting demonstrated that GLA downregulated the phosphorylation of p38 mitogen-activated protein kinases and c-Jun N-terminal kinase. A coimmunoprecipitation assay illustrated that formation of cAMP response element-binding protein (CREB)-activating protein 1 (AP1) complexes and the DNA binding activities of CREB and AP1 in MG63 and HOS cells were impaired following treatment with GLA. Finally, GLA inhibited tumor growth and suppressed osteosarcoma cell metastasis in vivo. Overall, our findings highlight the potential of GLA as a therapeutic agent for the prevention and treatment of tumor metastasis.
Effects of Ganoderma Lucidum Spore Oil and Ganoderma Lucidum Extraction Spore Oil on Angiogenesis Regulatory Factors

Abstract:Objective To study the role of ganoderma lucidum spore oil and ganoderma lucidum extraction spore oil in neovascularization of human high malignant breast cancer .Methods Human high malignant breast cancer cell MDA-MB-231 and tumor-bearing nude mice established with MDA-MB-231 were treated with different doses of ganoderma lucidum spore oil and ganoderma lucidum extraction spore oil .Epidermal growth factor receptor ( EGFR) expression level was examined by Western blotting and the RNA expression levels of neovascularization related molecules such as EGFR , vascular endothelial growth factor (VEGF), metalloproteinases(MMPs), thrombospondin(TSP-1), platelet derived growth factor( PDGF) , fibroblast growth factor ( FGF) were detected by Real-time PCR.Results Both ganoderma lucidum spore oil and ganoderma lucidum extraction spore oil inhibited the expression of EGFR in vitro and in vivo in a dose-dependent way.Both compounds induced down-regulation of VEGF and up-regulation of TSP-1 at RNA level.The effect of Ganoderma lucidum extraction spore oil was more significant than that of ganoderma lucidum spore oil .Conclusion Both ganoderma lucidum spore oil and ganoderma lucidum extraction spore oil inhibite the expression of neovascularization related molecules and increase the expression of molecules inhibiting neovascularization , whereas the effect of ganoderma lucidum extraction spore oil is more obvious .
Spore Oil enhances the effect of cyclophosphamide inhibiting programmed death-1 and prolongs the survival of H22 tumor-bearing mice

Abstract:Objective: To investigate the effect of Ganoderma Lucidum Spore Oil (GLSO) on the tumor growth and survival of H22 tumor-bearing mice treated with cyclophosphamide (CTX), and explore the underlying mechanism. Methods: Allograft H22 hepatocellular carcinoma mouse model was applied to investigate the effect of GLSO on the tumor growth and survival of animals, and Kaplan-Meier survival analysis was used to analyze the life span. Plasma biochemical examination was used to determine the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), urea (UREA) and creatinine (CRE). Western blot analysis was performed to detect Programmed Death-1 (PD-1), Programmed Death Ligand 1 (PD-L1), Janus Kinase 2 (JAK2), phosphorylated Signal Transducer and Activator of Transcription 3 (p-STAT3), and Signal Transducer and Activator of Transcription 3 (STAT3) expression. Results: GLSO increased the anti-tumor effect of CTX and prolonged the survival of H22 tumor-bearing mice treated with CTX. Meanwhile, GLSO increased the thymus index and showed no obvious toxicity to liver functions of animals. GLSO also decreased the level of UREA in H22 tumor-bearing mice treated with CTX. Furthermore, GLSO could inhibit the expression of PD-1 in spleen, which was independent of JAK2 expression and STAT3 phosphorylation. However, GLSO did not affect the expression of PD-L1, JAK2, and p-STAT3 in tumor tissue. Conclusion: GLSO could strengthen the anti-tumor effect of CTX and prolong the life span of H22 tumor-bearing mice, while the underlying mechanism might be relevant to the amelioration effect of thymus function and inhibition of PD-1 expression in spleen. Furthermore, these findings implied the promising role of GLSO in combination with CTX to extend the survival of patients in clinical chemotherapy of hepatocellular carcinoma.

Abstract:Purpose: Osteosarcoma is a malignant musculoskeletal tumor with early metastasis and a poor prognosis, especially in adolescents. Ganoderma lucidum (Leyss. Ex Fr.) Karst (G. lucidum), a traditional East Asian medicine, has been reported to play a critical role in antitumor and immunomodulatory activity. The aim of this study was to investigate the effects and molecular mechanisms of water extract of sporoderm-broken spores of G. lucidum (BSGWE) on osteosarcoma PD-L1 (programmed cell death-ligand 1) transcriptional regulation, efficacy enhancement, and side effect remission. Methods: The antitumor effects on cell proliferation of BSGWE in osteosarcoma cells were detected by apoptosis flow cytometry, and the migration ability of HOS and K7M2 cells were evaluated by cell scratch assay. Potential signaling regulation of PD-Ll was detected by western blotting. To confirm the signaling pathway of BSGWE-related PD-L1 downregulation, a pho-STAT3 turnover experiment was carried out. Colivelin was administered as a pho-STAT3 activator to rescue the BSGWE-induced PD-L1 inhibition. To further study in vivo signaling, in a Balb/c osteosarcoma allograft model, tumor volume was measured using an in vivo bioluminescence imaging system. The body weight curve and tumor volume curve were analyzed to reveal the remission effects of BSGWE on PD-L1 antibody-related body weight loss and its immunomodulatory effects on the osteosarcoma and spleen. The PD-L1 expression level and expression of related transcription-factor pho-STAT3 in tumor cells and spleens were assessed by IHC analysis. Results: BSGWE suppressed the proliferation and migration of osteosarcoma cells in vitro via induction of apoptosis. In addition, BSGWE downregulated PD-L1 expression and related STAT3 (signal transducers and activators of transcription) phosphorylation levels in a dose-dependent manner. Western blotting and qRT-PCR assay revealed that BSGWE downregulated PD-L1 expression by inhibiting STAT3 phosphorylation. A turnover experiment showed that colivelin administration could rescue PD-L1 inhibition via pho-STAT3 activation. BSGWE not only downregulated PD-L1 expression via the STAT3 pathway in an allograft Balb/c mouse model, but also relieved complications including weight loss and spleen atrophy in a mouse monoclonal antibody therapy model on the basis of its traditional advantages in immune enhancement. Conclusion: BSGWE downregulated PD-L1 expression via pho-STAT3 inhibition of protein and RNA levels. BSGWE enhanced PD-L1 antibody efficacy via phosphorylated STAT3 downregulation in vitro and in vivo. BSGWE also relieved complications of weight loss and spleen atrophy in a murine allograft osteosarcoma immune checkpoint blockade therapy model.
Anticancer Effect of Ganoderma Spores Oil on Primary Cancer Cells Isolated from Pleural Effusion

Abstract:Objective:To investigate the effect of Ganoderma spores oil on primary cancer cells isolated from the pleural effusion of patients with lung adenocarcinoma.Methods:Primary cancer cells were isolated from the pleural effusion(n=12) of patients,the response of these cancer cells to Ganoderma spores oil and chemotherapeutic agents commonly used in clinic were determined by CCK-8 assay,and compared with culture cell line A549.Flow cytometer was used to examine the apoptotic-induced effect of Ganoderma spores oil on primary cancer cells.RT-PCR was used to examine the role of Ganoderma spores oil in the expression of apoptotic-related genes including bax and bcl-2.Results:The primary cancer cells isolated from effusions showed sensitivity to Ganoderma spores oil with IC50 values of 0.28±0.04 μl/ml,similar to the IC50 values of 0.46±0.08 μl/ml in culture cell line A549(P0.05).The inhibition ratios of ganoderma spores oil,docetaxel,cisplatin and gemzar to primary cancer cells were 48.7%±3.3%,31.6%±14.1%,30.2%±11.6% and 51.4%±5.6%.There was no significant difference between Ganoderma spores oil and chemotherapeutic agents commonly used clinically(P0.05).Ganoderma spores oil had an important apoptotic-induced role in primary cancer cells.We also observed that Ganoderma spores oil could up-regulate bax mRNA and down-regulate of bcl-2 mRNA in primary cancer cells.Conclusion:Based on the response of cancer cells isolated from pleural effusion in this study,we believed that Ganoderma spores oil might have a potential anticancer effect.
Anti-inflammatory Effects of Ganoderma Lucidum Sterols Via Attenuation of the P38 MAPK and NF-κB Pathways in LPS-induced RAW 264.7 Macrophages

Abstract:Ganoderma lucidum exhibits pronounced anti-inflammatory effects, polysaccharides and triterpenoids are regarded as major constituents displaying the anti-inflammatory activities, whether sterols contribute to this activity remains unclear. Herein Ganoderma lucidum sterols (GLS) were innovatively isolated by a single-step procedure, the profile of GLS was characterized by HPLC-ELSD and shown similar to that of sterols separated by a traditional method, but much higher in content. Furthermore, GLS inhibited inflammation in macrophages by significantly attenuating LPS-induced cell polarization as well as releases and mRNA expressions of pro-inflammatory mediators NO, TNF-α, IL-1β and IL-6. Moreover, the anti-inflammatory activity of GLS was mediated by MAPK and NF-κB pathways, GLS suppressed MAPK pathways by blocking phosphorylation of p38 but not ERK and JNK, which is complementary with inhibitory effects of Ganoderma polysaccharides and triterpenes on JNK and ERK, indicating Ganoderma sterols may exert synergistic anti-inflammatory effect with polysaccharides and triterpenes. GLS also inhibited NF-κB pathways by restraining phosphorylation and degradation of IκB-α and blocking phosphorylation of NF-κB p65. Molecular docking confirmed that sterols of GLS were directly bound to active sites of p38 and p65 to suppress their activation. Therefore, our findings suggest GLS as natural and safe anti-inflammatory agents to prevent and treat inflammatory diseases.
Radiosensitizing effects of cyclooxygenase-2 selective inhibitor LM-1685 on A549 human lung adenocarcinoma cells

Abstract:Objective To investigate the radiosensitizing effects of cyclooxygenase-2 selective inhibitor LM-1685 on A549 cells in vitro.Methods A549 human lung adenocarcinoma cell line was used in this study.Cell growth kinetics Was determined using MTT assay.Cell survival was analyzed by clonogenic assay.The change of cell cycle Was measured by flow cytometry.Results LM-1685 inhibited the growth of A549 cells,showing a dose-dependent and time-dependent manner.LM-1685(50/μmol/L),either with or without IL-1β,showed the radiosensitizing effects on A549 cells,and the sensitizing enhancement ratio(SER)was 1.12 and 1.06,respectively.LM-1685(50 μmol/L)abrogated radiation-induced G2/M arrest of the tested A549 cells.Conclusions Cyclooxygenase-2 selective inhibitor can enhance the radiosensitivity of A549 cell line.Abrogation of radiation-induced G2/M arrest could be part of the mechanism.
The Effects of Ganoderma Lucidum Spore Oil on MCF-7 Cell Apoptosis and Migration

Abstract:Objective:To observe the effects of Ganoderma lucidum spore oil on proliferation,apoptosis and migration of human breast cancer cell MCF-7 in vitro,and explore its possible mechanisms.Methods: The proliferation inhibition rate of MCF-7 cells was determined by using the cell count kit 8(CCK-8).Wound healing assay was used to evaluate the cell migration.Cell apoptosis was detected by flow cytometry.Furthermore,to analyze the expression level of Bcl-2,Bax and VEGF mRNA,RT-PCR was performed.Results: Different concentrations of Ganoderma lucidum spore oil could inhibit MCF-7 cell proliferation as well as migration and took on dose and time dependently.It also induced cell apoptosis in a dose dependent way.The mRNA expressions of Bax related to apoptosis were increased,while Bcl-2 was decreased.On the other hand,the VEGF mRNA expression,which was important to tumor metastasis,was also declined.Conclusion: Ganoderma lucidum spore oil can inhibit MCF-7 cell proliferation and migration,on the other hand,it induces cell apoptosis.The mechanisms may be partly due to the decreased Bcl-2/Bax rate and VEGF mRNA expression.

Abstract:OBJECTIVE:To investigate the effects of vascular endothelial growth factor (VEGF) antisense oligodeoxynucleotide (ASODN) on the mRNA and protein expression of VEGF, Flt-1, and kinase insert domain containing receptor (KDR) and VEGF excretion in human gallbladder carcinoma cells.METHODS:Human gallbladder carcinoma cells of the line GBC-SD were cultured and transfected with VEGF ASODN and sense oligodeoxynucleotide (SODN) mediated by Oligofectamine. The toxicity of SODN and Oligofectamine to the GBC-SD cells was examined by MTT method. RT-PCR was used to detect the mRNA and expression of VEGF, Flt-1, and KDR, and ELISA was used to detect the protein expression of VEGF.RESULTS:MTT method showed that SODN and Oligofectamine were not toxic to the GBC-SD cells. The mRNA expression levels of VEGF, Flt-1, and KDR of the ASODN and ASODN + Oligofectamine groups were all significantly lower than those of the control group (all P < 0.05), and were the lowest 72 hours after transfection, and then gradually increased. ELISA showed that there were not significant differences in the VEGF protein concentration in the supernatant of the GBC-SD cells among the SODN, SODN + Oligofectamine, and control groups (all P < 0.05), however, the VEGF protein concentration in the supernatant of the GBC-SD cells of the ASDN and ASDN + Oligofectamine groups were significantly lower than that of the control group (both P < 0.05).CONCLUSION:VEGF ASODN inhibits the mRNA and protein expression of VEGF, Flt-1, and KDR and VEGF excretion in human gallbladder carcinoma cells. Oligofectamine strengthens the effect of ASODN.
Antitumor activity of enzymatically hydrolyzed Ganoderma lucidum polysaccharide on U14 cervical carcinoma–bearing mice

Abstract:Polysaccharides from Ganoderma lucidum have been demonstrated to possess diverse biological activities. Despite lots of studies on the biological activities of Ganoderma lucidum polysaccharide (GLP), little is known regarding the medicinal potential of low–molecular weight enzymatically hydrolyzed Ganoderma lucidum polysaccharide (EGLP). EGLP was prepared by enzymatic degradation and its potential effects in U14 cervical tumor–bearing mice were evaluated. Both GLP and EGLP delayed tumor growth of the tumor xenograft. The EGLP was superior to native polysaccharide. Moreover, EGLP treatment could effectively protect the immune organs of U14 cervical carcinoma–bearing mice. In addition, the EGLP treatment ameliorated oxidative stress as compared with cyclophosphamide (CTX). Compared with the MC group, the expression of Bcl-2 and COX-2 was obviously decreased by EGLP treatment, whereas the expression of Bax and cleaved caspase-3 was obviously increased. These results indicated that EGLP showed stronger antitumor activity with lower toxic effects and had the potential to be a novel antitumor agent.

Effects of water-soluble Ganoderma lucidum polysaccharides on the immune functions of patients with advanced lung cancer

Abstract:Preclinical studies have established that the polysaccharide fractions of Ganoderma lucidum have potential antitumor activity. Recent clinical studies have demonstrated that G. lucidum polysaccharides enhance host immune functions [e.g., enhanced natural killer (NK) cell activity] in patients with advanced solid tumors, although an objective response was not observed. This open-label study aimed to evaluate the effects of water-soluble G. lucidum polysaccharides (Ganopoly, Encore International Corp., Auckland, New Zealand) on immune functions in patients with advanced lung cancer. Thirty-six patients were enrolled and treated with 5.4 g/day Ganopoly for 12 weeks. In the 30 cancer patients who completed the trial, treatment with Ganopoly did not significantly alter the mean mitogenic reactivity to phytohemagglutinin, mean counts of CD3, CD4, CD8, and CD56, mean plasma concentrations of interleukin (IL)-2, IL-6, and interferon (IFN)-gamma, or NK activity in the patients, but the results were significantly variable. However, some cancer patients demonstrated markedly modulated immune functions. The changes in IL-1 were correlated with those for IL-6, IFN-gamma, CD3, CD8, and NK activity (P < .05), and IL-2 changes were correlated with those for IL-6, CD8, and NK activity. The results suggest that subgroups of cancer patients might be responsive to Ganopoly in combination with chemotherapy/radiotherapy. Further studies are needed to explore the efficacy and safety of Ganopoly used alone or in combination with chemotherapy/radiotherapy in lung cancer patients.

Abstract:Aim To determine the possible mechanism of the anti-tumor effect of GlPS.Methods MTT assay was adopted to measure the proliferation of human prostate carcinoma(PC-3M)cells.Human umbilical vein endothelial cells(HUVECs)were cultured in vitro and identified by morphology and immunofluorescence staining.Adhesion and migration ability of PC-3M and HUVECs monolayer were examined.Results GlPS showed no cytotoxic effect on PC-3M cells as well as the proliferation of HUVECs.GlPS could significantly inhibit PC-3M adhesion to and migration through HUVECs.Conclusion GlPS exerts the anti-tumor activity via inhibition of PC-3M adhesion to and migration through HUVECs.

Abstract:Ganoderma lucidum spore powder, valued for its nutritional and medicinal properties, contains polysaccharides crucial for its efficacy. However, the complex structural nature of these polysaccharides necessitates further investigation to fully realize their potential. This study aimed to investigate the effects of acid heat treatment on Ganoderma lucidum spore polysaccharides (GLSPs) to enhance their properties and application in antitumor activity. The GLSP was obtained via acid heat treatment, concentration, and centrifugal separation. This process led to a notable reduction in polysaccharide molecular weight, increasing water solubility and bioavailability. Analytical techniques including NMR spectroscopy and methylation analysis revealed a polysaccharide composition comprising four distinct monosaccharides, with molecular weights of 3291 Da (Mw) and 3216 Da (Mn). Six different linkage modes were identified, with a molar ratio of 1:5:2:3:4:3. In vivo experiments demonstrated the GLSP's significant inhibitory effect on the growth of four tumor models (sarcoma S180, Lewis lung cancer, liver cancer H22, and colon cancer C26) in mice, with no observed toxicity. These findings suggest the GLSP's potential as an antitumor therapeutic agent for clinical use.

Abstract:Objective:To explore the biological effect of arsenic trioxide (As2O3) in inhibition of proliferation of gastric adenocarcinoma cells SGC-7901 and.To investigate the effect of As2O3 to mitochondria and caspase-3 of the human gastric adenocarcinoma cells SGC-7901.Methods:The inhibition of proliferation of SGC-7901 cells treated with As2O3 was studied in growth curve and MTT assay.The nuclei were stained by Hoechst 33258, the nuclearmorphologic changes were observed by fluorescence microscope and apoptosis index was calculated.The apoptosis of SGC-7901 cells induced by As2O3 was investigated through method of detecting mitochondrial transmembrane potential, flow cytometry and detecting caspase-3.Results:As2O3 can inhibit significantly the growth of SGC-7901.SGC-7901 cells treated by As2O3 exhibited morphologic features of apoptosis, including nuclear shrinkage, nuclearcondensation, nuclear fragmentation and formation of apoptotic bodies.A typical apoptosis of SGC-7901 cells treated by As2O3 was detected by method of detecting mitochondrial transmembrane potential, flow cytometry and detecting caspase-3.Conclusion:As2O3 can induce significantly apoptosis of gastric cancer cells SGC-7901.The apoptosis induced by As2O3 was associated closely with the disruption of mitochondrial transmembrane potential and activation of caspase-3.
Experimental Study of the Inhibition of Triterpenoids Isolated from Ganoderma Lucidum on the Development of Oral Squamous Cell Carcinoma

Abstract:Objective:To discuss the inhibition mechanism of Ganoderma triterpenoids on the development and progression of oral cancer. Methods:60 hamster cheek pouch dynamic cancer animal models were established and divided into two groups. A group was treated with Ganoderma triterpenoids,B group was as control.The expression of VEGF were detected with immunohistochemistry method. The data was analyzed with CS10.34. Results:The pathological results showed that the normal epithelial cases of group A (Ganoderma triterpenoids group) were more than those of group B(the control group)and the epithelial dysplasia rate was significantly lower than group B(P0.01) on 6th week. On 9th week the rates of moderate and severe epithelial dysplasia and carcinoma were lower in group A than in group B(P0.05). On 12th week the cancer rate was significantly lower in group A than that in group B(P0.01). In the whole dynamic carcinogenesis observation period,the occurrence of two groups' histological difference was significant(P0.01). The severity in group A was still lower than that in group B and the grade of cancer induced was lower. Immunohistochemical study showed that the expression of VEGF gradually strengthened with the development of squamous cell carcinoma from normal mucosa. There were significant differences on the three grades,the simple hyperplasia,dysplasia and carcinoma information,of group A and B by rank sum test (P0.05). Conclusion:Ganoderma triterpenoids can inhibit carcinoma of the oral mucosa and showed significant inhibition of angiogenesis. It can be used for oral squamous cell carcinoma treatment.
Polyhydroxy Sterols Isolated From Ganoderma sinense Spores and Their Cytotoxic Activities

Abstract:Background: The fruiting body of Ganoderma sinense Zhao, Xu et Zhang iscertified as an authoritative medical material in the Chinese Pharmacopoeia 2020 edition and has been used as a crude drug for a long time. G sinense spores are crucial part of the fruiting body of G sinense and exhibited antitumor and immune-enhancing activities according to our previous reports. However, there were few studies about the chemical composition of G sinense spores. This study was aimed at the structure determination and antitumor effect of the sterols isolated from G sinense spores. Methods: Modern chromatographic methods were applied to isolate compounds from the ruptured spores of G sinense. Their chemical structures were elucidated by analyses of one-dimensional and two-dimensional 1 H nuclear magnetic resonance, and high-resolution electrospray ionization mass spectrometry data. The cytotoxicities of the isolated compounds against 3 tumor cell lines, human non-small cell lung cancer A549 and 95D cells, and human hepatocellular carcinoma HepG2 cells were measured by Cell Counting Kit-8. Results: A novel ergosterol derivative, (22 E,24 R)-3β,5β,6α,7α,14β-pentahydroxyergosta-8,22-dien-15-one (1), and 5 known sterols, (22 E,24 R)-3β,5α,9α,14β-tetrahydroxyergosta-7,22-dien-6-one (2), (22 E,24 R)-ergosta-7,9(11),22-triene-3β,5α,6β-triol (3), (22 E,24 R)-ergosta-7,9(11),22-triene-3β,5α,6β,14α-tetrol (4), β-daucosterine (5), and (22 E,24 R)-ergosta-7,22-diene-3β,5α,6β,9α-tetrol (6), were obtained from the ruptured spores of G sinense. Compound 1 exhibited cytotoxicity against A549 cells and HepG-2 cells with IC 50 values of 65.12 ± 4.76 and 97.34 ± 6.36 μM, respectively. Compounds 1, 2, and 4 were obtained from G sinense for the first time. Conclusion: This paper is a continuation of an investigation of the chemical ingredients from G sinense spores and their antitumor effect. Only compound 1 showed weak cytotoxicities against A549 cells and HepG-2 cells.
