Abstract:The best time for sterilization,the effects of different combinations of 6-BA and NAA on the inducement and proliferation of axillary bud,the effects of different concentrations of NAA on rooting,and the survival of transplanting to different substrates for explants ofDianthus caryophyllus was studied by tissue culture technique with the stem segments of the "Masters",a cultivar of Dianthus caryophyllus as materials,respectively.The results of experiments were:Aseptic seedlings would be obtained by the treatments of 0.1％ HgCl2 for 1 min or 10％ NaClO for 4 min,as it significantly reduced the contamination and improved the survival rate and rooting rate of explants.Combinatorial treatments with 3 mg/L of 6-BA and 0.1 mg/L of NAA,resulted in the best inducement and proliferation for the axillary bud growth.The germination rate (value of 83％) as well as coefficient of proliferation (value of 8.3) was better than those of treatment with growth regulator 6-BA only.The rooting rate ofexplants rose along with the increase of NAA concentration in the medium and it was up to 90％ when the 1 mg/L of NAA,while there was significantly difference between value for rooting number ratio and the root length (p＜0.05).The vermiculite or perlite mixed with vermiculite was the suitable medium for transplanting the tissue cultures of Dianthus caryophyllus.
