Abstract:Baculoviruses are arthropod specific viruses that have long been used as biological agents to control the insects attacking crop plants.They are also the key components of insect-baculovirus expression system(BEVS),which has been one of the four main expression systems in gene engineering.Additionally,baculovirus has been a new gene transfer vector used in gene therapy due to its advantages of the extremely high expression of foreign genes and being non-reproduce vector.The advances in application of Baculoviruses are reviewed in this paper.

Abstract:This paper reviews the effect of recombinant baculovirus insecticides on the predatory and parasitic natural enemies of insect pests，the recombinatio n between recombinant baculovirus and the genome of other organisms and the ecol ogical effect of recombinant baculovirus. So far studies have shown that compare d with wild\|type baculoviruses，the ecological fitness of recombinant baculovir u ses is reduced，thereby reducing the risks to ecosystems and non\|target organis m s such as predators and parasites. However，the use of the recombinant baculovir uses as biocontrol agents is not risk\|free. The long term biosafety of recombin ant baculoviruses needs to be profoundly and completely studied.

Abstract:As the molecular biology of insect baculoviruses advances, the insect baculoviruses are utilized as vectors in biotechnological researches and improved for their insecticidal effect.The baculovirus expression vector system (BEVS) was established by expressing a foreign gene incultured insect cells and in insect larvae or pupae with the baculovirus as a vector.The baculoviruses were also utilized as vectors for introduction of foreign genes into silkworm genome to develop transgenic silkworm.Furthermore,the baculovirus effects as biologicalinsecticides were promoted by insertion of genes of toxin,hormone or proteinase inhibitors into the baculovirus genome or by deletion of a gene from the genome.The host range of recombinant baculoviruses could be widened by hybridization of different baculoviruses.This paper reviews the advances in the baculovirus utilization in the above respects.

Abstract:Baculovirus has been indispensable in biological control,but their application as bioinsecticides was limited until recently because of their slow killing action and very narrow specificity. To improve the insecticidal efficacy of baculovirus by the technology and methods of genetic engineering,here we reviewed recent progresses in the development of integration of exogenous genes,modification of endogenous genes,bio-safety assessment of recombinant baculoviruses and the wider application of genetically modifed baculoviruses as biopesticides in future.

Abstract:As one of the four expression systems, baculovirus-insect expression system has been widely utilized in synthesis of recombinant proteins. This system has many advantages, such as high yield regulated by polyhedrin promoter, proper post-translation modification, ease of purification from serum-free culture supernatant, and lake of endotoxin contamination in these systems, but there are some shortcoming needing to improve. This paper presented current status and possible development of insect factories: High intensity early promoters' group of the baculovirus expression vector system (BEVS) has been successfully synthesized. Parent virus has been reconstructed in order to enlarge host range and improve the stability of recombination proteins; Instect cell culture media were optimized carefully to find new medium for special cell strain and serum free media was manufactured commercially for suspension cell mass production; Host cell engineering was being studied for complex glycosylation or providing stable cell environment; The vector with tag and effcient method for protein purification had made great progress. Also, the potential future uses of BEVS in research fields, medicine and agriculture are discussed in this paper.

Abstract:The baculovirus expression vector system is one of the most powerful and versatile eukaryotic expression systems available. However, as the recombinant baculovirus is usually generated by replacing the foreign gene into the polyhedrin locus, the resulting polyhedrin-negative virus is less infectious to the host larvae when administered via oral ingestion. This limits the large-scale production of the recombinant protein, as the host larvae can only be inoculated through dorsal injection, which is a laborious task. In this paper, we describe a new Bombyx mori nucleopolyhedrovirus polyhedrin-plus Bac-to-Bac baculovirus expression system for application in silkworm, B. mori. In this system, the foreign gene and the polyhedrin are co-expressed, and polyhedra are produced as in the wild-type virus, and thus the recombinant baculovirus can be used directly via oral infection. It effectively improves the efficiency of the baculovirus expression system and also widens the application of baculovirus in other fields, such as the development of new biological insecticides.

Abstract:The presently available expression tools and vectors (e.g., eukaryotic expression vectors and the adenovirus expression system) for studying the functional genes in Bombyx mori are insufficient. The baculovirus expression system is only used as a protein production tool; therefore, recombinant proteins expressed by B. mori using the baculovirus expression system equipped with a polyhedrin promoter cannot be used for in vivo research applications. In this work, we constructed and screened a eukaryotic expression vector for silkworm cells The EGFP and B. mori Argonaute2 proteins were found to be efficiently expressed using the screened pIEx-1 vector with the FuGENE 6 transfection reagent. Additionally, we constructed a novel nucleopolyhedrovirus ie1-Bacmid expression system for the production of recombinant protein; we then used the system to highly express the EGFP and B. mori Argonaute2 proteins. In this system, the protein of interest can be efficiently expressed 13 h after infection by controlling the B. mori nucleopolyhedrovirus immediate early ie1 promoter. The ie1-Bacmid system provides a powerful “adenovirus-like” expression tool; not only can the tool be used to study baculovirus molecular biology for the silkworm but it is also useful in other research applications as well, such as the study of gene functions involved in cellular physiological processes.

Abstract:Baculovirus expression system,based on the baculovirus as exogenous?gene expression vector and the insect cell(or live insect) as receptor,is one of eukaryotic?expression systems.Compared with other expression systems,the baculovirus expression system has special advantages,for example: as expression vector,the baculovirus genome can accommodate more exogenous genes;the very late promoter of baculovirus efficiently modulates exogenous gene expression;as receptor,insect cell maintains the ability to process and modify protein of interest;on general,the baculoviruses only infect arthropods and do not threaten human and animals.Therefore,the expression system is gaining increasing concerns and is applied into the research and production of subunit vaccines,specially for virus-like particle—the protein particle generated by self-assembly of one or more virus structural proteins but containing no genome—having incomparable?advantages: this article reviews the advantages in detail and discusses the development trend of virus-like particle vaccines.

Abstract:Baculoviruses are double-stranded DNA viruses which are highly selective for several insect groups. They are valuable natural control agents, but their utility in many agricultural applications has been limited by their slow speed of kill and narrow host specificity. Baculoviruses have been genetically modified to express foreign genes under powerful promoters in order to accelerate their speed of kill. In our and other laboratories, the expression of genes coding for insect juvenile hormone esterases and various peptide neurotoxins has resulted in recombinant baculoviruses with promise as biological insecticides. These viruses are efficacious in the laboratory, greenhouse and field and dramatically reduce damage caused by insect feeding. The recombinant viruses synergize and are synergized by classical pesticides such as pyrethroids. Since they are highly selective for pest insects, they can be used without disrupting biological control. Because the recombinant virus produces fewer progeny in infected larvae than the wild-type virus, they are rapidly out-competed in the ecosystem. The viruses can be used effectively with crops expressing endotoxins of Bacillus thuringiensis. They can be produced industrially but also by village industries, indicating that they have the potential to deliver sustainable pest control in developing countries. It remains to be seen, however, whether the current generation of recombinant baculoviruses will be competitive with the new generation of synthetic chemical pesticides. Current research clearly indicates, though, that the use of biological vectors of genes for insect control will find a place in agriculture. Baculoviruses will also prove valuable in testing the potential utility of proteins and peptides for insect control.

Abstract:Usually the recombinant baculovirus was generated through replacing the foreign gene into the polyhedrin locus resulting in a polyhedrin-negative virus. Such polh-negative baculovirus can infect insect cell lines well and insect also through subcutaneous injection, but is less infective to silkworm through per oral ingestion. In order to improve the inoculation efficiency, a polyhedrin-plus Bac-to-Bac expression system for application in silkworm was constructed in this paper. Using this new system, the foreign gene and polyhedrin were both expressed at higher level and large amount of polyhedra were formed. Thereby the recombinant baculovirus can infect silkworm larva by oral administration. This new system also widen the application of baculovirus in the field of biological insecticide, gene therapy and other aspects.

Abstract:Baculovirus has been widely used for the production of recombinant proteins in insect cells. The extremely high yield by baculovirus-infected insect cells or larvae makes it an attractive tool for pharmaceutical protein production. Since the finding that baculovirus can efficiently transduce mammalian cells, the applications of baculovirus have been greatly expanded. Although AcMNPV (Autographa californica multiple NPV) fails to replicate in vertebrate cells, it does express alien genes with levels of expression that are dependent on the strength of the promoter used to drive transcription of the foreign gene. Following these findings, subsequent studies have rapidly expanded the list of permissive cells that include cell lines originating from human, rodent, porcine, bovine and even fish sources. Many tries have been done to study the mechanism of baxulovirus entry into mammalian cells, but the events responsible for virus uptake and detailed mechanisms of intracellular movement and nuclear entry of the virus are still largely unknown. The application of modified baculoviruses for in vivo gene delivery has also been demonstrated. In contrast to other commonly used viral vectors, baculoviruses have the unique property of being incapable of initiating a replication cycle and producing infectious virus in mammalian cells. The AcMNPV genome is large, thus rendering the virus flexibility to carry multiple genes or large inserts. The recombinant viruses can be readily constructed and produced to high titers simply by infecting insect cells, initiate little to none microscopically observable cytopathic effects on mammalian cells and have a good biosafety profile. These attributes will undoubtedly lead to the increased application and continued development of this system for efficient gene delivery into mammalian cells.

Abstract:Baculoviruses are insect‐specific pathogens which have been employed for the natural control of insect pests for many years. They present an alternative to the use of chemical agents for crop protection. Their restricted host range enables particular pest species to be targeted, and this prevents any loss of beneficial insects from a treated area. However, their large‐scale development as insect control agents has been limited because they take longer to kill the host following infection than rapidly‐acting chemical insecticides. The aim of genetic modification of baculoviruses is to increase their speed of action, while preserving their beneficial characteristics, so creating a fast‐acting, environmentally friendly insecticide. The prospects for achieving this are discussed in this paper, with consideration of scientific progress and the influence of economic and legislative factors.

Abstract:Baculovirus expression vector system (BEVS) has been successfully applied to the over-expression of various proteins, thus providing sufficient materials for vaccine research. Compared to other systems, BEVS has many advantages: baculovirus solely being parasitic in invertebrates, the resultant products conferring high safety to mammalian, high expression level of recombinant proteins, preferable folding for eukaryotic protein, proper post-translational modification required for biological function, suitable for multiple genes co-expression and large-scale production with serum-free culture media. To better understand the advantages and prospective of BEVS for the vaccine research, this article will review the development of BEVS and its application on vaccine research.

Abstract:With continuous improvement of the baculovirus vector and the screening methods,the maximum recombinant virus efficiency is 100% now through Bac-to-Bac baculovirus method,which shortens the time to construct recombinant vectors and improves work efficiency greatly.In addition,the researchers developed some insect baculovirus expression vectors with expanded host range could produce high level recombinant proteins in silkworm or pupae.Insect baculovirus expression system is an excellent eukaryotic expression system with advantages of the extremely high expression of foreign proteins and the ability of posttranslational modification.There are about one thousand foreign proteins been produced successfully in this system so far.The baculovirus expression vector system,foreign gene expressions and its application in agriculture field were reviewed.

Abstract:Baculoviruses are insect pathogens widely used as biotechnological tools in different fields of life sciences and technologies. The particular biology of these entities (biosafety viruses 1; large circular double-stranded DNA genomes, infective per se; generally of narrow host range on insect larvae; many of the latter being pests in agriculture) and the availability of molecular-biology procedures (e.g., genetic engineering to edit their genomes) and cellular resources (availability of cell lines that grow under in vitro culture conditions) have enabled the application of baculoviruses as active ingredients in pest control, as systems for the expression of recombinant proteins (Baculovirus Expression Vector Systems—BEVS) and as viral vectors for gene delivery in mammals or to display antigenic proteins (Baculoviruses applied on mammals—BacMam). Accordingly, BEVS and BacMam technologies have been introduced in academia because of their availability as commercial systems and ease of use and have also reached the human pharmaceutical industry, as incomparable tools in the development of biological products such as diagnostic kits, vaccines, protein therapies, and—though still in the conceptual stage involving animal models—gene therapies. Among all the baculovirus species, the Autographa californica multiple nucleopolyhedrovirus has been the most highly exploited in the above utilities for the human-biotechnology field. This review highlights the main achievements (in their different stages of development) of the use of BEVS and BacMam technologies for the generation of products for infectious and noninfectious human diseases.Key points• Baculoviruses can assist as biotechnological tools in human health problems.• Vaccines and diagnosis reagents produced in the baculovirus platform are described.• The use of recombinant baculovirus for gene therapy–based treatment is reviewed.

Abstract:The use of baculoviruses (BVs) as bio-insecticides could be enhanced by incorporation of genes leading to reduced damage to crops by the infected host insect during the period of disease incubation. The risks associated with release of genetically engineered BVs can be best assessed against a comprehensive background of knowledge of their natural ecology. This paper summarises what is known of BV ecology and presents a structured programme of further research employing a virus belonging to this group which has been genomically labelled with a non-coding synthetic oligonucleotide sequence. Such a marker would permit an unique BV population to be traced through the environment in space and time.
