Mechanism by which EGCG inhibits proliferation of collegen Ⅱ-specific T cells

Abstract:To study the mechanism for inhibition of the collegen Ⅱ-specific T cell proliferation by EGCG,the mouse model for collegen-induced arthritis(CIA) was established by immunization of mice with collegen Ⅱ(CⅡ).The proliferation of CⅡ-specific CD4+T cells was examined by 3H-TdR incorporation method.The proliferation of CⅡ-specific CD4+T cells in the presence of CD11b+APC was examined in two ways: either the CⅡ-specific CD4+T cells were pretreated,or the APC were pretreated,by EGCG.In addition,apoptosis of CⅡ-specific CD4+T cells and the expression levels of APC surface markers were evaluated by flow cytometry after treatment with different concentrations of EGCG,the percentage of Treg was also examined.The results showed that inhibition of the proliferation of CⅡ-specific T cell by EGCG was dose-dependent,and EGCG influenced antigen presentation function of APCs.Moreover,EGCG can induce apoptosis of CⅡ-specific CD4+ T cells,up-regulate expression levels of the marker PD-L1 on APCs,but EGCG can't increase the percentage of Treg cells.These results justify further in vivo research for effect of EGCG on CIA and thereby lay foundation for clinical research for EGCG therapy of rheumatoid arthritis.
