The Status and Significance of Loss Imprint of IGF2 Gene in Renal Carcinoma

Abstract:Objective Exploring the status and significance of loss of imprinting(LOI)of IGF-Ⅱ gene in renal cell carcinoma.Methods Restriction fragment length polymorphism(RFLP)analysis were used to detect the imprinting status of IGF-Ⅱ gene in 75renal cell carcinoma tissues and matched histological normal kidney tissues.Results There were 58renal cell carcinoma cases with heterozygous IGF-Ⅱ gene(77.3%,58/75).In these 58cases,26cases had loss of imprinting(LOI)of IGF-Ⅱ gene both in carcinoma tissues and matched histological normal kidney tissues(44.8%,26/58),and 5cases had IGF-Ⅱ LOI in carcinoma tissues(8.62%,5/58),and 1cases had IGF-Ⅱ LOI in matched histological normal kidney tissues(1.72%,1/58).IGF-Ⅱ LOI occurred on both genders and all ages,there was no statistical differences.In 58heterozygous cases,there were 31(53.4%,31/58)and 27(46.6%)cases had IGFⅡ LOI in renal carcinoma tissues and matched histological normal kidney tissues,respectively.Statistical analysis showed that there was no statistical significant between the IGF-Ⅱ LOI in renal cell carcinoma tissue and the matched histological normal kidney tissues.Conclusion There is no significant correlation between IGF-Ⅱ LOI and the gender and age of renal cell carcinoma patients.IGF-Ⅱ LOI incidence is close in renal cell carcinoma tissues and matched histological normal kidney tissues,which indicates that IGF-Ⅱ LOI may occurs in the early stage of the disease.
