Abstract:The new promiscuous activity of lipase from porcine pancreas, type II (PPL II), has been observed to catalyze the direct asymmetric aldol reaction of heterocyclic ketones with aromatic aldehydes. PPL II showed favorable catalytic activity and had a good adaptability to different substrates in the reaction. The enantioselectivities of up to 87% ee and diastereoselectivities of up to 83:17 (anti/syn) were achieved. It is interesting that PPL II possesses the function of aldolase in organic solvents.

Abstract:Esterase BioH, which is obligatory for biotin synthesis in Escherichia coli, was found to exhibit a promiscuous ability to catalyse Aldol and Knoevenagel reactions with moderate to good yields. The reaction conditions including organic solvent, molar ratio of ketone to aldehyde, enzyme amount, and reaction time were investigated to evaluate the effect of different reaction conditions on yield. Target compounds were afforded in the best yield of 91.2% for Aldol reaction and 54.7% for Knoevenagel reaction. In addition, because the enzyme could be prepared with a low cost, this protocol could provide an economic route to conduct Aldol and Knoevenagel reactions, which expand the field of enzymatic promiscuity.

Abstract:Several proteases, especially pepsin, were observed to directly catalyze asymmetric aldol reactions. Pepsin, which displays well-documented proteolytic activity under acidic conditions, exhibited distinct catalytic activity in a crossed aldol reaction between acetone and 4-nitrobenzaldehyde with high yield and moderate enantioselectivity. Fluorescence experiments indicated that under neutral pH conditions, pepsin maintains its native conformation and that the natural structure plays an important role in biocatalytic promiscuity. Moreover, no significant loss of enantioselectivity was found even after four cycles of catalyst recycling, showing the high stability of pepsin under the selected aqueous reaction conditions. This case of biocatalytic promiscuity not only expands the application of proteases to new chemical transformations, but also could be developed into a potentially valuable method for green organic synthesis.

Abstract:PPL, lipase from porcine pancreas, is first reported to catalyze direct asymmetric aldol reactions between aromatic aldehydes and cyclic ketones. More importantly, the catalytic activity of PPL was greatly promoted by a small quantity of water at 37 degrees C. A wide range of aromatic aldehydes reacted with cyclic ketones to provide the corresponding aldol products with high yields (up to 99%) and moderate to good stereoselectivity (up to 90%ee and 99:1 dr).

Abstract:Porcine pancreas lipase type II (PPL II) exhibited unnatural catalytic activity in direct asymmetric aldol reactions between cyclic ketones and aromatic or heteroaromatic aldehydes in acetonitrile in the presence of phosphate buffer. A wide range of substrates was accepted by the enzyme to afford the corresponding aldol products in low to high yields (10 - 98%), with moderate to excellent enantioselectivities (53 - 94% ee, for anti-isomers) and low to moderate diastereoselectivities (48/52 - 87/13 dr, anti/syn). This methodology expands the application of PPL II, and it might be developed into a potentially valuable method for sustainable organic synthesis.

Abstract:Ten lipases and esterases have been examined to catalyse the reaction between p-nitrobenzaldehyde and methyl vinyl ketone, the Baylis–Hillman reaction, to form 3-[hydroxyl-(4-nitrophenyl)-methyl]-but-3-en-2-one. Among these enzymes, Escherichia coli BioH esterase had the best activity. Optimal conditions for this reaction were: 0.1 mmol aldehyde, 0.1 mmol activated alkene, 30 mg E. coli BioH, 1 ml acetonitrile at 30 °C for 96 h. In addition to the named substrates, four other aldehydes and three activated alkenes were also investigated to determine the substrate range of the enzyme. The structures of nine products were confirmed by NMR and yields of the corresponding products ranged from 21 to 46 %.

Abstract:A facile tandem route to α,β-unsaturated aldehydes was developed by combining the two catalytic activities of the same enzyme in a one-pot strategy for the aldol reaction and in situ generation of acetaldehyde. Lipase from Mucor miehei was found to have conventional and promiscuous catalytic activities for the hydrolysis of vinyl acetate and aldol condensation with in situ formed acetaldehyde. The first reaction continuously provided material for the second reaction, which effectively reduced the volatilization loss, oxidation, and polymerization of acetaldehyde, as well as avoided a negative effect on the enzyme of excessive amounts of acetaldehyde. After optimizing the process, several substrates participated in the reaction and provided the target products in moderate to high yields using this single lipase-catalyzed one-pot biotransformation. One-pot tandem approach for the synthesis of α,β-unsaturated aldehydes was developed under the catalysis of lipase from M. miehei (MML), firstly. MML displayed conventional and promiscuous catalytic activities for hydrolysis of vinyl acetate and aldol condensation with in situ formed acetaldehyde.

Abstract:Six lipases were screened for their ability to catalyse the Knoevenagel condensation between benzaldehyde and methyl cyanoacetate. Lipase from porcine pancreas tolerated a variety of functional groups on the aromatic ring, produced the highest yields, and also catalysed transesterification of the product in the presence of an alcoholic cosolvent. We show that, in organic solvents, lipase from porcine pancreas has higher activity for this "promiscuous" reaction than for naturally occurring esterification catalysis.

Abstract:This work, for the first time, demonstrated the catalytic promiscuity of an acyl-peptide releasing enzyme from Sulfolobus tokodaii (ST0779) for aldol addition reaction, which shows accelerated activity at elevated temperature. The turnover number kcat (s−1) of this thermostable enzyme at 55°C is 7.78-fold higher than that of porcine pancreatic lipase (PPL) at its optimum temperature of 37°C, which is one of the best reported enzymes for promiscuous catalysis of aldol reaction; and the molecular catalytic efficiency kcat/Km (M−1s−1) adds up to 140 times higher than PPL.

Abstract:The catalytic promiscuity of pepsin from porcine gastric mucous was observed in catalysis of the direct asymmetric aldol reactions of aromatic aldehydes with acetones, which were substituted by hydroxy-, dihydroxy-, methoxy- and benzyloxy- for the synthesis of diol compounds in acetonitrile. This biocatalysis was also applicable to the aldol reactions of cyclic or hetereocyclic ketones with aromatic aldehydes. Yields of up to 87%, diastereoselectivities of up to >99/1 dr and enantioselectivities of up to 75% ee were achieved.

Abstract:Enzymes are renowned for their catalytic efficiency and selectivity, but many classical transformations in organic synthesis have no biocatalytic counterpart. Aldolases are prodigious C–C bond-forming enzymes, but their reactivity has only been extended past activated carbonyl electrophiles in special cases. To probe the mechanistic origins of this limitation, we use a pair of aldolases whose activity is dependent on pyridoxal phosphate. Our results reveal how aldolases are limited by kinetically favourable proton transfer with solvent, which undermines aldol addition into ketones. We show how a transaldolase can circumvent this limitation, enabling efficient addition into unactivated ketones. The resulting products are highly sought non-canonical amino acids with side chains that contain chiral tertiary alcohols. Mechanistic analysis reveals that transaldolase activity is an intrinsic feature of pyridoxal phosphate chemistry and identifies principles for extending aldolase catalysis beyond its previous limits to enable convergent, enantioselective C–C bond formation from simple starting materials.

Abstract:Unnatural catalytic activity of trypsin from porcine pancreas for direct asymmetric aldol reaction was discovered. The reactions between aromatic aldehydes and various ketones gave products in moderate yields and enantioselectivities in the presence of a small amount of water. The influences of solvent, water content, temperature, mole ratio of substrates, and enzyme concentration were investigated. The mechanism of trypsin-catalyzed aldol reaction was discussed. This enzymatic promiscuity widens the application of trypsin to new chemical transformations.

Abstract:The unnatural ability of nuclease p1 from Penicillium citrinum was first discovered to catalyze asymmetric aldol reactions between aromatic aldehydes and cyclic ketones under solvent-free conditions. The excellent enantioselectivities of up to 99% ee and high diastereoselectivities of up to >99 : 1 (anti/syn) were achieved. This nuclease p1 catalyzed reaction provided a novel case for new activities of existing enzymes, which widens the applicability of this biocatalyst in organic synthesis.
