Genetic Diversity Analysis of 44 Shares of Hibiscus Cannabinus L. Germplasm Resources Using ISSR Molecular Marker

Abstract:[Objective] This study was to reveal the genetic diversity and genetic relationship of the kenaf(Hibiscus cannabinus L.) resources from different origins, thus providing basis for genetic improvement and molecular marker-assisted breeding of kenaf. [Method] Ninety one ISSR molecular markers were used for amplification on 44 shares of kenaf germplasm resources, of which 21 showing good diversity and clear bands were chosen for PCR amplification. Based on amplification results, the genetic similarity coefficients among kenaf germplasm resources were calculated by using analytic software NTSYSpc-2.10e, and phylogenetic tree was then established via UPGMA. [Result] Totally 169 bands were amplified using the 21 screened primers, averagely 8.05 bands were amplified from each primer. Of them, 141 bands were polymorphic, accounting for 83.4%. When genetic similarity coefficient 0.887 was used as criterion L1, these 44 shares of kenaf germplasm could be classified to be 32 shares of cultivars and 12 shares of wild type or half-wild type varieties. When genetic similarity coefficient 0.897 was used as criterion L2, these 32 shares of cultivars could be further grouped into four sub-clusters. The genetic diversities between cultivars and wild type or half-wild type varieties were between 0.46-0.91, showing huge hereditary difference; while that among 32 cultivars were between 0.85-0.97, suggesting that genetic relationships among cultivars are relatively close and their genetic similarities are rather narrow. [Conclusion] ISSR could well determine the genetic similarities among kenaf germplasm resources and provide valuable molecular information for selecting parents of hybrid cross, which can lay a good foundation for DNA mapping of kenaf germplasm resources.

Abstract:Hibiscus rosa-sinensis (H. rosa-sinensis) has been largely used in traditional medicine. This study aims to review the pharmacological and phytochemical properties of Hibiscus rosa-sinensis L and also summarize the pharmacological, photochemical, and toxicological characteristics of H. rosa-sinensis. The current review focuses on the distribution, chemical content, and main uses of H. rosa-sinensis. Various scientific databases, including ScienceDirect, Scopus, PubMed, Google Scholar, etc., were used. Correct plant names were verified from plantlist.org. The results were interpreted, analyzed, and documented based on bibliographic information. This plant has been frequently used in conventional medicine due to its high concentration of phytochemicals. All its parts contain numerous chemical compounds, such as flavonoids, tannins, terpenoids, anthocyanins, saponins, cyclopeptide alkaloids, and vitamins. More interestingly, the roots of this plant contain glycosides, tannins, phytosterols, fixed oils, fats, flavonoids, saponins, gums, and mucilages. The leaves contain alkaloids, glycosides, reducing sugars, fat, resin, and sterols. The stem contains other chemical compounds, such as ß-sitosterol, teraxeryl acetate, cyclic sterculic, and malvalic acids. Finally, the flowers contain riboflavin, thiamine, apigenidine, oxalic acid, citric acid, quercetin, niacin, pelargonidine, and ascorbic acid. This species has a wide variety of pharmacological applications, such as antimicrobial, antioxidant, antidiabetic, anti-inflammatory, antihypertensive, antifertility, antifungal, anticancer, hair growth-promoting, antihyperlipidemic, reproductive, neurobehavioral, antidepressant, and antipyretic activities. Finally, toxicological studies have shown that higher doses of extracts from the plant are safe.
Floral syndrome and breeding characters of Iris speculatrix

Abstract:Through the specific field observation of Iris speculatrix, its outcrossing index , pollen vitality and stigma receptivity , the ratio of pollen and ovule , artificial pollination and bagging were investigated to understand the flower feature , blossom dynamic and breeding system .Results showed that under the natural climate of Hangzhou , the flow-ering stage of populations was about 35 d from May to June, and the majority of plants kept blooming in 8th May to 20th May.The flowering stage for individual plant was 3-6 days.The life span of a single flower was about 3 d. The flowers, setting on the same inflorescence , were monoecism.Male and female flowers mature at the same time , and the distance between anther and stigma was about 3 mm.The out cross index(OCI)was 4, and pollen-ovule ratio ( P/O) was 552 .Taking the results of the emasculated , bagged and artificial pollination experiment together , it indi-cated that Iris speculatrix flower is a mixed-crossing system with out-crossing affinity as the main breeding form and concomitance out-crossing.Artificial pollination on fruit set rate was higher than the natural pollination .It was con-firmed that under natural conditions , the reproductive system of Iris speculatrix was affected by floral organ structure and environmental factors .

Abstract:Hibiscus (Hibiscus syriacus L.) is known as a horticultural plant of great ornamental and medicinal value. However, the effect of NaCl stress on hibiscus seedlings is unclear. Little is known about H. syriacus ‘Duede Brabaul’ (DB) and H. syriacus ‘Blueberry Smoothie’ (BS). Here, the effects of solutions with different concentrations of NaCl on the organic osmolytes, ion accumulation, and antioxidant enzyme activity of hibiscus seedling leaves were determined. The results showed that the Na+/K+ ratio was imbalanced with increasing NaCl concentration, especially in BS (range 34% to 121%), which was more sensitive than DB (range 32% to 187%) under NaCl concentrations of 50 to 200 mM. To cope with the osmotic stress, the content of organic osmolytes increased significantly. Additionally, NaCl stress caused a large increase in O2·− and H2O2, and other reactive oxygen species (ROS), and antioxidant enzyme activity was significantly increased to remove excess ROS. The expression level of genes related to salt tolerance was significantly higher in DB than that in BS under different NaCl concentrations. Taken together, DB possessed a stronger tolerance to salt stress and the results suggest membrane stability, Na+/K+, H2O2, catalase and ascorbate peroxidase as salt tolerance biomarkers that can be used for gene transformation and breeding in future hibiscus research.

Abstract:Background: The flower colour of H. syriacus 'Qiansiban' transitions from fuchsia to pink-purple and finally to pale purple, thereby enhancing the ornamental value of the cultivars. However, the molecular mechanism underlying this change in flower colour in H. syriacus has not been elucidated. In this study, the transcriptomic data of H. syriacus 'Qiansiban' at five developmental stages were analysed to investigate the impact of flavonoid components on flower colour variation. Additionally, five cDNA libraries were constructed from H. syriacus 'Qiansiban' during critical blooming stages, and the transcriptomes were sequenced to investigate the molecular mechanisms underlying changes in flower colouration. Results: High-performance liquid chromatography‒mass spectrometry detected five anthocyanins in H. syriacus 'Qiansiban', with malvaccin-3-O-glucoside being the predominant compound in the flowers of H. syriacus at different stages, followed by petunigenin-3-O-glucoside. The levels of these five anthocyanins exhibited gradual declines throughout the flowering process. In terms of the composition and profile of flavonoids and flavonols, a total of seven flavonoids were identified: quercetin-3-glucoside, luteolin-7-O-glucoside, Santianol-7-O-glucoside, kaempferol-O-hexosyl-C-hexarbonoside, apigenin-C-diglucoside, luteolin-3,7-diglucoside, and apigenin-7-O-rutinoside. A total of 2,702 DEGs were identified based on the selected reference genome. Based on the enrichment analysis of differentially expressed genes, we identified 9 structural genes (PAL, CHS, FLS, DRF, ANS, CHI, F3H, F3'5'H, and UFGT) and 7 transcription factors (3 MYB, 4 bHLH) associated with flavonoid biosynthesis. The qRT‒PCR results were in good agreement with the high-throughput sequencing data. Conclusion: This study will establish a fundamental basis for elucidating the mechanisms underlying alterations in the flower pigmentation of H. syriacus.

Abstract:The pollens of 2 cultivators from Hibiscus syriacus were studied.The pollen viability were tested by means of TTC and germination in vitro.The results showed that only when in vitro medium,the pollen germination rate could be improved and represent the pollen viability.The pollen was used as explant and the sucrose solution as medium.The germination of the pollen medium of Hibiscus syriacus was observed,The pollen began germinating with 15% sucrose concentration and 15mg/L boric acid under 30℃ after 30 minetes.

Abstract:The present study is an attempt to review the history of super hybrid rice breeding in China and abroad and an introduction to advancement in super hybrid rice breeding in China.At present,a great breakthrough in utilizing subspecies heterosis of indica and japonica rice and plant type breeding has been obtained in super hybrid rice breeding in China,and super hybrid rice has been extended for large scale cultivation.But,there are many problems existing in super hybrid rice cultivation,such as poorer adaptability,lower seed setting rate,bad grain quality,worse resistance to biotic and abiotic stresses,fewer double-cropping super rice varieties(combinations),imperfect breeding theory and technology.Therefore,some suggestions and research prospects have been put forward for overcoming these problems.

Abstract:Sea buckthorn (Hippophae rhamnoides L.), as an economically and ecologically valuable plant with rich nutritional and bioactive compounds, has garnered significant interest. The demand for Sea buckthorn has explosive growth, highlighting the urgent need for the cultivation of fast-growing, high-quality Sea buckthorn seedlings. However, there are still some controversies in Sea buckthorn germplasm resource research. This review provides a comprehensive summary of the recent research findings on Sea buckthorn plants, encompassing their classification, distribution, propagation methods, medical functions, and valorization. It aims to offer strong support for the industrial utilization of the Sea buckthorn and explores the prospects for molecular breeding in Sea buckthorn.

Abstract:The hollies (Ilex L., Aquifoliaceae) form a large (>669 spp.) genus of forest trees and shrubs, which is almost cosmopolitan in mesic environments but most diverse in subtropical China and montane South America. Throughout the range of the genus, Ilex species have been utilized as beverages, medicines, ornamentals, honey plants, timber, and for various other minor uses. Recent studies on the genomics, evolution, and biogeography of Ilex now make it possible to take a systematic approach to understanding and expanding the economic importance of the genus, but information on existing uses is scattered among numerous published and unpublished sources. We therefore review the existing literature on utilization of Ilex species, supplementing this with information from the grey literature and product websites. We show that, despite the number and diversity of known uses, most Ilex species are not known to be utilized at present, suggesting considerable unrealized potential. We highlight gaps in our knowledge and opportunities for expanded usage. Finally, we discuss how the availability of a new phylogeny and whole genome can assist screening of additional wild species for economic potential and facilitate breeding programs for species already under cultivation.

Abstract:Blue honeysuckle, also named as haskap or honeyberry, an emerging fruit worldwide, with the species being Lonicera L., originating from the north hemisphere, exhibits high genetic heterogeneity and abundant genetic variation in fruit and leaf traits. However, there are few papers on the research progress of blue honeysuckle breeding and cultivars. The International Union for the Protection of New Varieties of Plants (UPOV) is an effective plant variety protection system, and blue honeysuckle cultivars from different countries and regions are registered in the UPOV database. In this review, we summarized the origin, domestication and germplasm collection, breeding categories and supporting techniques of blue honeysuckle, and the information on registered blue honeysuckle cultivars was analysed based on the UPOV database. Finally, the paper put forward the challenges of blue honeysuckle in development, breeding direction and perspectives to promote the development of blue honeysuckle.

Abstract:The dried flowers of Hibiscus sabdariffa (HS), available worldwide, have various applications in both non-medicinal and medicinal fields. The growing global interest in the health benefits of HS is linked to its potential prevention or management of non-communicable diseases. The aim of this research was to find the optimal extraction method that ensures the maximum yield of multiple beneficial bioactive components, such as polyphenols, anthocyanins, vitamin C, β-carotene, antioxidant activity, free radical scavenging activity DPPH and ferric reducing antioxidant power (FRAP). To this end, stirring, pulsed electric field, and ultrasound-assisted extraction were evaluated, either alone or in combination. Under optimized extraction conditions, the obtained extract exhibited an elevated total polyphenol content (37.82 mg of gallic acid equivalents/g dry weight (dw)), total anthocyanin content (610.42 μg of cyanidin equivalents/g dw), total carotenoids content (921.84 μg of β-carotene equivalents/g dw), and ascorbic acid content (507.44 mg/100 g dw). Remarkably, the extracts exhibited strong antioxidant properties (487.51 μmol of ascorbic acid equivalents (AAE)/g dw and 243.42 μmol AAE/g dw as evidenced by FRAP and DPPH assays, respectively). This research advances the parameters that should be employed to produce the optimal and nutritionally enhanced HS flower extracts, that can be used in the commercial sector.

Abstract:Changium smyrnioides Wolff, a monotypic species of family Umbellaceae, has a narrow distribution area and can found only in Yangtze River basin in China; the widespread Anthriscus sylvestris (L.) Hoffm can be found from temperate to subtropical zone in Asia, Europe and North America. Both species have fleshy roots, which are prized in Chinese medicine. Because of the land use, C. smyrnioides has now become an endangered species, while A. sylvestris do not endangered. Comparative studies on the ecological strategies based on the biological and ecological traits of the two species in present paper, aimed to find out the endangered mechanism of C. smyrnioides. Research was conducted at Hangzhou, Ningbo, Nanjing, Chuzhou and Anqing. The shoot of C. smyrnioides has only the basal leaf, which ensures it can capture light efficiently at ground level. The aboveground leaf area ratio (LAR_a) of C. smyrnioides was found to be higher than A. sylvestris, suggesting it can capture more light per unit of biomass. After spring time, C. smyrnioides loses its survival competitiveness as other herbaceous species can grow rapidly and impedes sunlight. C. smyrnioides is not a shade-tolerant species, that may explain why C. smyrnioides are typically found in small population under defoliated trees. Conversely, A. sylvestris can increase light absorption and out-compete other herbaceous by its distinct ability to expand densely and occupies extra horizontal and vertical space. In a word, C. smyrnioides is more effective in light capture and water acquisition than A. sylvestris, but the ecological strategy it with cause it specialized or overspecialized to the narrow environment, which were limited under the deciduous trees in subtropical evergreen broad-leaved forest zone in winter. As the human disturbance (digging for medicinal use and deforestation) becoming serious, C. smyrnioides becoming rare and an endangered plant.

Abstract:Orchidaceae constitute one of the largest families of angiosperms. They are one of the most ecological and evolutionary significant plants and have successfully colonized almost every habitat on earth. Because of the significance of plant biology, market needs and the current level of breeding technologies, basic research into orchid biology and the application of biotechnology in the orchid industry are continually endearing scientists to orchids in Taiwan. In this introductory review, we give an overview of the research activities in orchid biology and biotechnology, including the status of genomics, transformation technology, flowering regulation, molecular regulatory mechanisms of floral development, scent production and color presentation. This information will provide a broad scope for study of orchid biology and serve as a starting point for uncovering the mysteries of orchid evolution.

Abstract:Hibiscus syriacus L. var. Shigyoku is a new double-flowered bluish-purple variety in China that changes color during flower development from bluish-purple to light purple. There is limited information on the anthocyanin accumulation patterns and associated transcriptome signatures in Shigyoku from D1 (bud) to open flower (D3). Here, we employed a combined transcriptome and metabolome approach to understanding the mechanism of this color change. Our results demonstrate that cyanidins, pelargonidins, delphinidins, petunidins, peonidins, and malvidins were differentially accumulated in Shigyoku petals. The anthocyanin biosynthesis started in D1, was significantly upregulated in D2 (semi-open flower), and reduced in D3. However, malvidins, pelargonidins, and peonidins could be associated with the bluish-purple coloration on D2. Their reduced accumulation in D3 imparted the light purple coloration to Shigyoku petals on D3. Significant contributions in the color change could be associated with the expression changes in anthocyanin biosynthesis genes i.e., LARs, ANSs, DFRs, UGT79B1, C3'Hs, 3ATs, and BZ1s. The UFGTs were associated with the higher accumulation of glycosylated anthocyanins in D2 and D3. Furthermore, the changes in the expressions of the MYB and bHLH transcription factors were consistent with the anthocyanin accumulation. Finally, we discussed the possible roles of Jasmonic acid, auxin, and gibberellic acid signaling in regulating the MBW complex. Taken together, we conclude that H. syriacus petal coloration is associated with anthocyanin biosynthesis genes, the MBW complex, and phytohormone signaling.
