Abstract:Dengzhanxixin (DZXX), the dried whole plant of Erigeron breviscapus (Vaniot) Hand.-Mazz., belonging to Compositae and first published in Materia Medica of South Yunnan by Lan Mao in the Ming Dynasty (1368 AD–1644 AD), is included in Medicinal Materials and Decoction Pieces of the 2020 edition of the Pharmacopeia of the People’s Republic of China . Its main chemical components are flavonoids that mainly include flavonoid, flavonols, dihydroflavones, flavonol glycosides, flavonoid glycosides, coffee acyl compounds, and other substances, such as volatile oil compounds, coumarins, aromatic acids, pentacyclic terpenoids, phytosterols, and xanthones. Among them, scutellarin and 1,5-dicoffeoylquininic acid are the main active components of DZXX. DZXX has pharmacological effects, such as improving cerebral and cerebrovascular ischemia, increasing blood flow, inhibiting platelet aggregation, promoting antithrombotic formation, improving microcirculation, reducing blood viscosity, protecting optic nerves, exhibiting anti-inflammatory properties, scavenging free radicals, and eliciting antioxidant activities. It is widely used in the treatment of cardiovascular and cerebrovascular ischemic diseases, kidney diseases, liver diseases, diabetic complications, and glaucoma. Pharmacokinetic studies have shown that the active components of DZXX have a low bioavailability and a high elimination rate in vivo. Nevertheless, its utilization can be improved through liposome preparation and combination with other drugs. Acute and subacute toxicity studies have shown that DZXX is a safe medicinal material widely used in clinical settings. However, its target and drug action mechanism are unclear because of the complexity of its composition. In this paper, the clinical application and pharmacological toxicology of DZXX are reviewed to provide a reference for further studying its active components and action mechanism.

Qualitative and Quantitative Analysis of Polyphenols in Erigeron Breviscapus

Abstract:Objective: To establish qualitative and quantitative analysis methods for quality evaluation of Erigeron breviscapus.Methods: The separation was performed on an Alltima C18 reverse-phase column(250 mm × 4.6 mm,5 μm);the mobile phase consisted of water containing 0.1% formic acid and acetonitrile with gradient elution at the flow rate of 1.0 mL.min-1,and the detection wavelength was 320 nm.A combination of ion trap tandem mass spectrometer(Trap-MSn) and time-of-flight mass spectrometer(TOF-MS) was applied to qualitative analysis under negative ion mode.The contents of 5 marker compounds,chlorogenic acid(3-O-CQA),scutellarin,3,5 O-dicaffeoylquinic acid(3,5-O-diCQA),1,5-O-dicaffeoylquinic acid(1,5-O-diCQA) and 4,5-O dicaffeoylquinic acid(4,5-O-diCQA),as well as total polyphenol of 11 samples of different origin and different parts of Erigeron breviscapus were determined by HPLC and UV,respectively.Results: Totally 14 peaks were identified based on the retention time,on-line UV spectra and MS spectra.The quantitative analysis results showed that there were obvious differences among different origins and different parts of Erigeron breviscapus.Conclusion: The established methods are simple and reliable,which are helpful for quality control of Erigeron breviscapus.

Abstract:Erigeron breviscapus is an important herbal drug for the treatment of cardiovascular and cerebral vessel diseases. In this study, phenolic compounds were extracted from the whole plant of E. breviscapus and analyzed by high-performance liquid chromatography/electrospray ionization mass spectrometry. A total of 53 compounds were identified or tentatively characterized based on their UV and mass spectra. These compounds included caffeoylquinic acids (CQAs), CQA glucosides, malonyl-CQAs, acetyl-CQAs, caffeoyl-2,7-anhydro-3-deoxy-2-octulopyranosonic acids (CDOAs), caffeoyl-2,7-anhydro-2-octulopyranosonic acids (COAs), flavones, flavonols and flavonones. Most of them were reported for the first time from E. breviscapus and nineteen of them belonged to new compounds. The MS(n) spectra of CQA glucosides were similar to CQAs and they were discriminated by their retention times. No caffeic acid related ions (X(0) (-), Y(0) (-) and Z(0) (-)) were observed in MS(n) spectra of acyl-CQAs compared to those of CQAs. Fragment ions ((2,5)O(-), (3,6)O(-) and (4,6)O(-)) corresponding to ring cleavage were shown in MS(n) spectra of CDOAs and COAs, characteristic of this class of compounds. The 5,6,7-trihydroxyl-substituted flavones were dominant in E. breviscapus. Their [A--H](-) ions underwent the loss of a molecule of H(2)O, followed by the loss of CO, which was used to discriminate from other hydroxyl-substituted flavones. Our results are the first comprehensive analysis of E. breviscapus constituents and will be helpful for the quality control of the herb of E. breviscapus and its related preparations.

Abstract:Ethnopharmacological relevance: Erigeron breviscapus is a common medicine of eight ethnic minorities, including Miao, Naxi, and Yi. As early as the Ming Dynasty (AD 1368-1644), Lanmao's Materia Medica of Southern Yunnan (AD 1436) recorded that the medicine is used for the treatment of "Zuo tan you huan." In modern pharmacological research, Erigeron breviscapus injection is the most commonly used preparation in the treatment of ischemic stroke caused by acute cerebral infarction, but its mechanism of action in the treatment of ischemic stroke is not well understood. Aim of the study: In this study, a metabonomics study based on ultraperformance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-QTOF-MS) was used in investigating the effect of a traditional Chinese medicine preparation Erigeron breviscapus injection on the rat model of focal cerebral ischemia-reperfusion and the affinity of its main components with the targets of mitochondrial apoptotic pathways. Materials and methods: This study used molecular docking technology to verify the effective binding ability of main effective components of Erigeron breviscapus injection to target proteins related to mitochondrial apoptosis pathway. This study developed a metabonomics method based on the ultra-performance liquid chromatography combined with quadrupole time-of-flight tandem mass spectrometry (UPLC Q-TOF MS) to evaluate the efficacy and study the mechanism of traditional Chinese medicine preparation. With pattern recognition analysis (principal component analysis and partial least squares-discriminate analysis) of urinary metabolites, a clear separation of focal cerebral ischemia-reperfusion model group and healthy control group was achieved. Results: Erigeron breviscapus injection can significantly reduce the area of cerebral infarction, improve tissue morphological lesion in rats, and can increase the number of Nissl bodies. It may be a promoting factor by inhibiting hippocampal nerve cell apoptosis and Bax protein expression and by exerting effects against ischemia reperfusion after the induction of apoptosis. Thus, it plays a role in brain protection. Moreover, it can considerably promote the recovery of neurological deficiency signs in advance. Meanwhile, Erigeron breviscapus decreased malondialdehyde content and T-NOS activity. Its curative effect from strong to weak order: low dose > high dose > medium dose. The representative components of Erigeron breviscapus have good affinity with the active sites of mitochondrial apoptosis-related proteins. Metabolomics found that the potential biomarkers regulated by breviscapine are kynurequinolinic acid, succinylornithine, and leucine proline. It is speculated that it may participate in TRP-kynurequinolinic acid and succinylornithine-urea cycle-NO metabolic pathways. Conclusions: This paper revealed the potential biomarkers and metabolic pathways regulated by Erigeron breviscapus. It was speculated that the mechanism is related to its inhibition of mitochondrion-mediated apoptosis. Erigeron breviscapus could restore the metabolic profiles of the model animals to normal animal levels. The mechanism may be related to the potential biomarkers of quinolinic acid, succinylornithine, and leucine proline and the metabolic pathways involved. However, the exact mechanism by which Erigeron breviscapus inhibits mitochondrion-mediated apoptosis remains to be further explored.

Abstract:Erigeron breviscapus is an important medicinal plant in Compositae and the first species to realize the whole process from the decoding of the draft genome sequence to scutellarin biosynthesis in yeast. However, the previous low‐quality genome assembly has hindered the optimization of candidate genes involved in scutellarin synthesis and the development of molecular‐assisted breeding based on the genome. Here, the E. breviscapus genome was updated using PacBio RSII sequencing data and Hi‐C data, and increased in size from 1.2 Gb to 1.43 Gb, with a scaffold N50 of 156.82 Mb and contig N50 of 140.95 kb, and a total of 43,514 protein‐coding genes were obtained and oriented onto nine pseudo‐chromosomes, thus becoming the third plant species assembled to chromosome level after sunflower and lettuce in Compositae. Fourteen genes with evidence for positive selection were identified and found to be related to leaf morphology, flowering and secondary metabolism. The number of genes in some gene families involved in flavonoid biosynthesis in E. breviscapus have been significantly expanded. In particular, additional candidate genes involved in scutellarin biosynthesis, such as flavonoid‐7‐O‐glucuronosyltransferase genes (F7GATs) were identified using updated genome. In addition, three candidate genes encoding indole‐3‐pyruvate monooxygenase YUCCA2 (YUC2), serine carboxypeptidase‐like 18 (SCPL18), and F‐box protein (FBP), respectively, were identified to be likely related to leaf development and flowering by re‐sequencing 99 individuals. These results provided a substantial genetic basis for improving agronomic and quality traits of E. breviscapus, and provided a platform for improving other draft genome assemblies to chromosome‐level.

Abstract:OBJECTIVE To investigate the chemical constituents from Erigeron multiradiatus(Wall.) Benth..METHODS Column chromatographic techniques were used for isolation and purification of chemical constituents of the plant and the structures were elucidated by chemical and spectral methods.RESULTS Ten compounds were isolated and indentified as: β-sitosterol(Ⅰ),kaempferol(Ⅱ),Apigenin(Ⅲ),Isorhamnetin(Ⅳ),P-hydroxybenzoic acid(Ⅴ),1-hydroxy-2,3,5-trimethoxy xanthone(Ⅵ),Wogonin(Ⅶ),Quercetin(Ⅷ),Emodin(Ⅸ),3,5-dimethoxy-3',4':2",3"-dihydro-furanchalcone(Ⅹ).CONCLUSION Compound Ⅰ,Ⅳ,Ⅵ,Ⅶ,Ⅸ,Ⅹ are isolated from this plant for the first time.

Abstract:A low-temperature, timesaving, lower solvent consumption, and energy cost and multi-stage countercurrent extraction (MCCE) technique was developed for pilot-scale production of scutellarein from Erigeron breviscapus (Vant.) Hand-Mazz. The optimum conditions of MCCE process were obtained using the orthogonal array design method, i.e., 70% (v/v) of ethanol water solution, 16L/kg of solvent to herbal sample ratio, 45C of extraction temperature and 30min of extraction time. A comparison between the MCCE technique and single pot extraction (SPE) under respective optimized operation conditions was made and time courses of scutellarein of MCCE and SPE processes were plotted, indicating that the MCCE technique can lower the extraction temperature and decrease five-sixth of the extraction time and two-thirds of the solvent consumption at the equivalent extraction yield of scutellarein.

Abstract:Ethnopharmacological relevance: Angelica biserrata (R.H. Shan & C.Q. Yuan) C.Q. Yuan & R.H. Shan (Angelica pubescens Maxim. f. biserrata Shan et Yuan) (A.biserrata) is a widely used traditional Chinese medicine; its roots known as 'Duhuo' in China. The herb is used for expelling wind, eliminating dampness, and terminating pain. Moreover, it is used for treating the onset of anemofrigid-damp arthralgia, pain of the waist and knee and headache caused by latent wind pathogenic factor or damp-cold pathogenic factor. A.biserrata is slightly warm, bitter and pungent in taste, and it is well distributed in regions such as Sichuan and Hubei Provinces. Aim of the study: This review aims to provide critical summary of the current evidence on A.biserrata. In particular, the progress of studies in the fields of botany, ethnopharmacology, phytochemistry, pharmacology and toxicity are discussed. Possible directions for future research are also briefly proposed. Materials and methods: Information on A.biserrata was collected from the internet database PubMed, Elsevier, China Knowledge Resource Integrated databases, ResearchGate, Web of Science, Wiley Online Library and Europe PMC using a combination of various relevant keywords. Other published books providing an overview of extant literature studies were considered for reference if they are related to the taxonomy, traditional knowledge, phytochemistry, pharmacology and toxicity of the plant. Results: A substantial proportion of the isolated and identified compounds of the herb were reported to be coumarins and volatile oils. Biological effects, such as neuroprotective, anti-tumor, anti-arthritis, anti-inflammatory, and sedative, were also validated in In vitro and in vivo studies. Therapeutic effects are attributed to the bioactivities of the naturally occurring compounds in this herb. Conclusions: A.biserrata has been proven as a valuable medicinal sources from traditional herb. Some conventional uses has been evaluated by pharmacological investigation. Although the crude extracts of A.biserrata has been emerged to possess more pharmacological activities, it is now time to isolate and identify more active chemical constituents by Bioactivity-Guided and elucidate their structure-activity relationship. More designed investigations are need to focus on understanding the multi-target network pharmacology, clarity the molecular mechanism of action and efficacy as well as identifying the effective doses of A.biserrata. In addition, A.biserrata is not fully assessed regarding its safety. Further studies are essential to investigate its toxicity on human. It's useful to provide identify its underlying therapeutic remedy and economic value of developing new medicine in the future.
Sulfur fertiliser enhancement of Erigeron breviscapus (Asteraceae) quality by improving plant physiological responses and reducing soil cadmium bioavailability

Abstract:AbstractErigeron breviscapus (Vant.) Hand.-Mazz. is an important medicinal plant; however, its quality is severely diminished by cadmium (Cd) pollution. Sulfur fertilisation can improve the production and application of E. breviscapus. This study examined Cd stress alleviation in the soil–plant system and determined the plant growth response after the application of sulfur fertiliser. The soil Cd concentration used in the treatments was 100 g·kg−1, and the sulfur fertiliser application rates were 0.1, 0.2, and 0.3 g·kg−1. Using pot experiments, we explored the impacts of high, medium, and low amounts of sulfur fertiliser on Cd accumulation and the quality and activity of E. breviscapus. The results showed that the application of sulfur fertiliser promoted Cd transformation to residual Cd under oxidation conditions, reducing Cd accumulation in E. breviscapus. Throughout the growth period, the application of sulfur fertiliser increased the soluble protein content and antioxidant enzyme activity, which alleviated Cd toxicity. The net photosynthetic rate, transpiration rate, intercellular CO2 concentration, chlorophyll level, and leaf width increased significantly. The biomass content of E. breviscapus also increased. Sulfur fertiliser improves the quality of herbaceous medicinal plants by reducing Cd accumulation and increasing scutellarin, chlorogenic, isochlorogenic acid B, and isochlorogenic acid C contents. A reasonable application of sulfur fertiliser is essential for improving E. breviscapus quality. This study provides a new method to reduce the ecological risk of planting herbaceous medicinal plants in Cd-contaminated soil.

Abstract:Erycibes are members of the Convolvulaceae family, including more than 10 species worldwide that are distributed in tropical Asia. Some Erycibes species have long been used as traditional remedies for rheumatoid arthritis, fever, hepatitis, and liver injury in China and Thailand. A total of 152 compounds from Erycibes plants have been isolated and identified, categorized as flavonoids, coumarins, quinic acid derivatives, lignans, and alkaloids. Coumarins are the characteristic and active constituents of this species, including scopoletin and scopolin. Modern pharmacological studies have shown that the extracts and bioactive components of Erycibes plants exhibit several biological activities, including antiinflammatory, analgesic, hepatoprotective, anti‐gout, antitumor, antioxidation, and other therapeutic effects. However, in recent years, due to destructive exploitation and utilization, some Erycibes plants' natural resources have become rare or endangered. Developing substitutes is a strategy to alleviate the pressure on those endangered medicinal plant resources. To provide a scientific basis for the development and protection of those threatened Erycibes species, this review summarized the current status of the chemical compositions, pharmacological activities, quality control studies, and the development of substitutes for Erycibes plants. In particular, the rationale for use of Porana sinensis currently on the market is discussed.

Abstract:In this study, the volatile components of Erigeron sublyratus essential oils and their anti‐inflammatory and cytotoxic activities were investigated for the first time. Gas chromatography‐mass spectrometry (GC‐MS) analysis revealed that a total of 28 components were identified in the root and aerial part essential oils. Among them, cis‐lachnophyllum ester (53.4‐64.2%), followed by germacrene D (5.6‐8.6%), trans‐β‐ocimene (2.6‐7.5%), β‐caryophyllene (4.7‐6.8%), β‐myrcene (2.0‐6.3%), and (E)‐β‐famesene (4.8‐5.0%) were principal components. The root essential oil significantly inhibited nitric oxide (NO) production on LPS‐induced RAW264.7 cells (IC50 = 1.41 ± 0.10 μg/mL) as compared to standard, dexamethasone (IC50 = 5.43 ± 0.54 μg/mL). Besides, both root and aerial part essential oils exhibited cytotoxic activity against MCF‐7, SK‐LU‐1, and HepG2 (IC50 from 1.11 ± 0.04 to 1.70 ± 0.05 μg/mL). Molecular docking simulation results show that (E)‐β‐farnesene exhibits the strongest binding energy among the studied compounds with the VEGFR‐2 enzyme (ΔG = ‐7.295 kcal/mol), while δ‐cadinene demonstrates the strongest affinity (ΔG = ‐8.047 kcal/mol) towards the COX‐2 enzyme. Furthermore, hydrophobic interactions were indicated to be the main contributors to the binding ability in the studied protein‐ligand complex. These findings proposed that E. sublyratus can be exploited for its anti‐inflammatory and anti‐cytotoxicity potential.

Abstract:Erigeron Breviscapus is known as a natural plant medicinal material with special clinical effect. In this work, the seeds of Erigeron Breviscapus were irradiated by He-Ne laser (632.8nm, 6mw) with different doses. Plant physiology and biochemistry index such as total soluble sugar and chlorophyll content were analyzed one month after germination to study the He-Ne laser effect to the Erigeron Breviscapus seeds. This work may provide the experimental basis for good growth vigor and high yield genetic character screening of Erigeron Breviscapus treated by laser mutation breeding.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3>Euphorbia fischeriana Steud. (Euphorbiaceae) is a perennial herb distributed in grassland, hill slopes or gravel hillside, with average altitude of 100-600 meters. The whole grass of E. fischeriana is toxic with roots used as folk medicine to treat Zhushui, dyspepsia, abdominal distension, abdominal pain, cough, as well as external applications such as cure of scabies and tuberculosis of lymph nodes.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Aim of the review</h3>This systematic review aims to provide a detailed and in-depth summary about the reported advances in traditional uses, clinical applications, phytochemistry, pharmacology and toxicity of E. fischeriana, so as to offer fresh ideas and broader vision and insights for subsequent studies.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3>Various scientific data bases such as CNKI, Elsevier, Google Scholar, Pubmed, Science Direct, SciFinder Scholar and Web of Science were searched to collect information about E. fischeriana. Other relevant literatures were searched in 'Flora of China Editorial Committee', ancient books, Ph.D and Masters' Dissertation to get more data of E. fischeriana.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3>A total of 241 chemical constituents have been identified from the roots of E. fischeriana, including diterpenoids, triterpenoids, meroterpenoids, acetophenones, flavonoids, coumarins, steroids, phenolic acids, tannins, etc. Various pharmacological activities have been demonstrated, especially anti-tumor, antibacterial, anti-inflammatory, antiviral and anti-leukemia activities. Moreover, different investigations about clinical uses and toxicology of E. fischeriana indicated that attention should be paid to its usage and dosage.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3>The researches of E. fischeriana are excellent, but gap still remains. As a poisonous traditional Chinese medicine, there are not enough studies on the toxicity of E. fischeriana. In addition, scholars' research on the pharmacological mechanism of E. fischeriana focuses more on the anti-tumor activity, which can be broadened in the future. Presumably, chemical constituents and biological activities of diterpenoids and trace meroterpenoids in E. fischeriana deserve further research in-depth in the future, in order to provide low toxicity and high efficiency lead compounds. Meanwhile, further studies on other medicinal aspects may lay a foundation for the comprehensive development and utilization of E. fischeriana.

Abstract:Natural products, owing to their chemical diversity, biological activity, and historical success, continue to be a precious source of lead compounds and potential candidates for drug discovery. Siegesbeckia , in particular, presents a promising avenue for drug discovery due to its long history of usage in traditional Chinese medicine and its well-established clinical applications. Diterpenoids and sesquiterpenoids, the characteristic metabolites of Siegesbeckia , have consistently attracted considerable attention in related scientific research because of their diverse structures and extensive range of bioactivities, including anti-inflammatory and anti-cancer properties. Herein, we provide an overview of the phytochemistry, biosynthesis, pharmacological action, as well as clinical applications of Siegesbeckia . This paper will provide a valuable reference for advancing further research and development of Siegesbeckia .

Abstract:Scutellaria Linn., which belongs to the family Lamiaceae, is a commonly used medicinal plant for heat clearing and detoxification. In particular, the roots of S. baicalensis and the entire herb of S. barbata have been widely used in traditional medicine for thousands of years. The main active components of Scutellaria , including: baicalein, wogonin, norwogonin, scutellarein, and their glycosides have potential or existing drug usage. However, the wild resources of Scutellaria plants have been overexploited, and degenerated germplasm resources cannot fulfill the requirements of chemical extraction and clinical usage. Metabolic engineering and green production via microorganisms provide alternative strategies for greater efficiency in the production of natural products. Here, we review the progress of: pharmacological investigations, multi-omics, biosynthetic pathways, and metabolic engineering of various Scutellaria species and their active compounds. In addition, based on multi-omics data, we systematically analyze the phylogenetic relationships of Scutellaria and predict candidate transcription factors related to the regulation of active flavonoids. Finally, we propose the prospects of directed evolution of core enzymes and genome-assisted breeding to alleviate the shortage of plant resources of Scutellaria . This review provides important insights into the sustainable utilization and development of Scutellaria resources.

Abstract:A method was developed to identify and trace the geographic sources of Erigeron breviscapus using high-resolution mass spectrometry and chemometrics. The representative samples were collected from the geographic area of Honghe Dengzhanhua and other areas in Yunnan province and Guizhou province. The data points could be determined well using the PCA and PLS-DA diagram. A total of 46 characteristic compounds were identified from Honghe Dengzhanhua and within Guizhou province, but 37 compounds were different from Honghe Dengzhanhua and other counties in Yunnan province. Two biomarkers were found from three regions. Their structures were inferred as 8-amino-7-oxononanoic acid and 8-hydroxyquinoline, and they had the same molecular composition. This may suggest that a possible synthesis pathway can be proven in the future.

Abstract:Ethnopharmacological relevance: The aerial parts of Senecio scandens Buch.-Ham. (family Asteraceae) have a long history in traditional Chinese medicine as a treatment for various ailments, such as bacterial diarrhea, enteritis, conjunctivitis, and respiratory tract infections. Materials and methods: A bibliographic investigation of Senecio scandens was accomplished by analyzing secondary sources, including the Chinese Medicinal plantal Classics, the Internet (Google Scholar and Baidu Scholar), and scientific databases accepted worldwide (Pubmed, Scopus, Web of Science, SciFinder, and CNKI). These sources were scrutinized for available information about the uses of Senecio scandens in traditional Chinese medicine, phytochemistry, pharmacology, and toxicology. Results: Senecio scandens is a medicinal plant with a climbing woody stem. Phytochemical studies have shown the presence of numerous valuable compounds, such as flavonoids, alkaloids, phenolic acids, terpenes, volatile oils, carotenoids, and trace elements. Among them, PAs are the characteristic constituents, adonifoline is one of the index ingredients of Senecio scandens. Studies in modern pharmacology have demonstrated that extracts and compounds isolated from Senecio scandens show a wide spectrum of pharmacological activities, including anti-inflammatory, antimicrobial, anti-leptospirosis, hepatoprotective, anti-infusorial, antioxidant, antiviral, antitumoral, analgesic, mutagenic, and toxicological activities. Conclusions: Phytochemical and pharmacological studies have demonstrated that the extracts of the plant possess various pharmacological activities that can be attributed to the presence of various flavonoids, phenolic acids, and alkaloids. Newer technologies for qualitative and quantitative methods of PAs need to be developed to obtain better accuracy and sensitivity. Due to the toxicity of PAs present in this medicinal plant, the regulations on PAs of Senecio scandens were varied among different countries and regions. In China, the PAs toxicity of Senecio scandens ranking criteria is not well defined in the Chinese Pharmacopoeia 2010, and there is no strict uniform requirement on the PAs in western countries. We propose that the use of Senecio scandens should be reevaluated based on a set of criteria, which includes risk-benefit analysis and severity of the toxic effects, clinical and preclinical data to ensure safe use while continuing to satisfy the need for access to the medicinal plant.

Abstract:The transcription factors of WRKY genes play essential roles in plant growth, stress responses, and metabolite biosynthesis. Erigeron breviscapus , a traditional Chinese herb, is abundant in flavonoids and has been used for centuries to treat cardiovascular and cerebrovascular diseases. However, the WRKY transcription factors that regulate flavonoid biosynthesis in E. breviscapus remain unknown. In this study, a total of 75 EbWRKY transcription factors were predicted through comprehensive genome-wide characterization of E. breviscapus and the chromosomal localization of each EbWRKY gene was investigated. RNA sequencing revealed transient responses of 74 predicted EbWRKY genes to exogenous abscisic acid (ABA), salicylic acid (SA), and gibberellin 3 (GA3) after 4 h of treatment. In contrast, the expression of key structural genes involved in flavonoid biosynthesis increased after 4 h in GA3 treatment. However, the content of flavonoid metabolites in leaves significantly increased at 12 h. The qRT-PCR results showed that the expression patterns of EbWRKY11 , EbWRKY30 , EbWRKY31 , EbWRKY36 , and EbWRKY44 transcription factors exhibited a high degree of similarity to the 11 structural genes involved in flavonoid biosynthesis. Protein-DNA interactions were performed between the key genes involved in scutellarin biosynthesis and candidate WRKYs . The result showed that F7GAT interacts with EbWRKY11, EbWRKY36, and EbWRKY44, while EbF6H has a self-activation function. This study provides comprehensive information on the regulatory control network of flavonoid accumulation mechanisms, offering valuable insights for breeding E. breviscapus varieties with enhanced scutellarin content.

Abstract:Background: Scutellariae radix (SR) is the dried root of Scutellaria baicalensis Georgi. It has a long history of ethnic medicinal use, traditionally recognized for its efficacy in clearing heat， drying dampness, eliminating fire， removing toxins , stopping bleeding and tranquilizing fetus to prevent miscarriage. Clinically, it is used to treat cold, fever, migraine, hand-foot-and-mouth diseases, liver cancer and inflammatory diseases. Purpose: The review aims to provide a comprehensive reference on the ethnobotanical uses, processing, phytochemistry, pharmacological effect, quality control and influence factors of biosynthesis for a deeper understanding of SR. Results and conclusion: A total of 210 isolated components have been reported in the literature, including flavonoids and their glycosides, phenylpropanoids, phenylethanoid glycosides, phenolic acids, volatile components, polysaccharides and others. The extract of SR and its main flavonoids such as baicalin, baicalein, wogonin, wogonoside, and scutellarin showed antioxidant, anti-inflammatory, anti-tumor, antiviral, hepatoprotective, and neuroprotective effects. However, further studies are required to elucidate its mechanisms of action and clinical applications. The pharmacodynamic evaluation based on traditional efficacy should be conducted. Although various analytical methods have been established for the quality control of SR, there are gaps in the research regarding efficacy-related quality markers and the development of quality control standards for its processed products. The regulatory mechanisms of flavonoids biosynthesis remain to be explored while the influence of environmental and transcription factors on the biosynthesis have been studied. In conclusion, SR is a promising herbal medicine with significant potential for future development.
