Effect of N-acetylcysteine on expressions of MCP-1 and MIP-2 in rat lung with acute pancreatitis

Abstract:Objective To explore the potential role of MCP-1 and MIP-2 in acute pancreatitis(AP)-associated lung injury,and evaluate the effect of N-acetylcysteine(NAC)on the expressions of monocyte chemoattractant protein(MCP-1)mRNA and macrophage inflammatory protein(MIP-2)mRNA in the lungs.Methods Thirty-five SD rats were randomly divided into three groups of sham-operation group(SO),acute necrotizing pancreatits(ANP)3,6 and 12 h groups,and NAC-pretreated 3,6 h and 12 h groups.ANP was induced by retrograde injection 4% sodium taurocholate into the bilipancreatic duct.NAC-pretreated groups were injected NAC(500 mg/kg)intraperitoneally 30 minutes before induction.Lung injury was evaluated by quantitating lung myeloperoxidase(MPO)activity,pulmonary microvascular permeability and pathological examination.The expressions of MCP-1 mRNA and MIP-2 mRNA were analyzedd by reverse transcriptase-PCR.Results Compared with group SO,the levels of MPO and pulmonary microvascular permeability in the lung significantly increased in group ANP(P<0.05 or P<0.01).The expressions of MCP-1 mRNA and MIP-2 mRNA in the lungs were upregulated obviously in group ANP.The expressions of MCP-1 mRNA and MIP-2 mRNA were positively correlated with the severity of lung injury.Compared with group ANP,the activity of intrapulmonary MPO,pulmonary microvascular permeability,and the expressions of MCP-1 mRNA and MIP-2 mRNA in the lungs were markedly decreased in NAC-pretreated groups.The lung injury was obviously ameliorated in NAC-pretreated groups as compared with that in group ANP.Conclusion The chemokines of MCP-1 and MIP-2 might play an important role in the pathogenesis of AP-associated lung injury.NAC might have benificial effects on AP-associated lung injury through downregulating the expressions of MCP-1 and MIP-2.
