Mechanism of 5-Aza-2′-deoxycytidine Inhibiting the Growth of CNE Cell Line

Abstract:Objective To explore the effect of 5-Aza-2′deoxycytidine on the growth of CNE cell line and find new target of nasopharyngeal carcinoma therapy.Methods CNE cells were treated by 5-Aza-2′-deoxycytidine with different concentrations.MTT was used to detect the effect of 5-Aza-2′-deoxycytidine on the growth of CNE cells.Methylation specific PCR was used to evaluate methylation status of DAPK gene.Cell apoptosis,the expression of DAPK mRNA and protein were detected by flow cytometry,reverse transcription polymerase chain reaction(RT-PCR) and immunocytochemistry,respectively.Results Each dose of 5-Aza-2′-deoxycytidine could suppress the growth of CNE cells.Promoter hypermethylation of DAPK gene existed and no expression of DAPK mRNA was found in CNE cells without 5-Aza-2′-deoxycytidine treatment.Hypermethylated DAPK gene was partly demethylated and 5-Aza-2′-deoxycytidine induced DAPK expression in a dose-dependent manner after CNE cells were treated with 5-Aza-2′-deoxycytidine.Immunocytochemistry revealed that the expression of DAPK protein was positive in experiment group,while negative in control group.Flow cytometer showed that proportion of apoptotic cells and cells in G_(1)stage were significantly increased,while those in G_(2)/M stages were reduced in CNE cells treated with 5-Aza-2′-deoxycytidine.Conclusion 5-Aza-2′-deoxycytidine can inhibit the growth of tumor cells by reactivating DAPK genes silenced by promoter methylation in CNE cells in vitro.
