Abstract:Taking Medicago sativa 'Golden Queen' as materials,the effects of different hormones concentrations on cotyledon node bud induction were studied to establish a regenerating system of Medicago sativa.The results showed that MS media without any hormones was the best media on cotyledon node bud induction.The best combination was 1 mg/L IBA+10 g/L Sucrose in the rooting stage.Hygromycin had inhibition effect on buds induction and growth.When the concentration reached 20 mg/L,induced bud would wither or die with time gone,so the best concentration was 20 mg/L.As a bacteriostatic agent,cef had no significant influence on regeneration in 700 mg/L,but with the concentration increased,the mortality rate of cotyledon node increased.
