Tht Effect Study of Oligchitosan on Sodium Urate Induced Acute Gouty Arthritis

Abstract:Objective: To investigate the effect of oligochitosan on acute gouty arthritis induced by sodium urate as well as its possible mechanisms. Methods: 48 SD male rats were randomly divided into 6 groups which include normal control group, model group, colchicine group and oligochitosan (low dose, medium dose, high dose) groups. The rats were administrated orally with different dose of drugs for 7 days. Animal models of acute gouty arthritis were made on the 4th day. The ankle perimeters at 0, 1h, 2h, 4h, 6h, 12h, 24h, 48h, 72h before and after sodium urate injection were measured separately and the difference was considered as the swelling level of gouty arthritis. After the last measurement, all the rats were anesthetized and the blood was collected for leucocyte count and IL-1β test. Results: (1) There was not obvious difference of ankle perimeters in experimental groups before sodium urate injection (P0.05). The ankle perimeters of rats increased after sodium urate injection and reached the peak after 12h, especially model group. The ankle perimeters of rats in oligochitosan groups and colchicine group declined in different degrees. After 24h, the swelling gradually subsidised and the swelling level of oligochitosan groups and colchicine group was lower than that of model group at 12h and 24h (P0.01). There was not obvious swelling peak in normal control group. (2)High and medium dose of oligochitosan groups could increase leucocytes of model rats (P0.01). (3) High and medium dose of oligochitosan groups could decrease IL-1β tendency of model rats (P0.01). Conclusion: Sodium urate intra-articular injection can successfully establish the rat model of acute gouty arthritis; Oligochitosan can relieve the attack of acute gouty arthritis and the curative effect of high and medium dose groups is stable.
