Cloning and Sequence Analysis of RAPD Markers Linked to the Resistant Powdery Mildew Genes in Chinese Wild Vitis

Abstract:Seven RAPD markers linked to the resistant powdery mildew (Uncinula necator) genes in Chinese wild Vitis (Vitis pseudoreticulata), OPW02-1756, OPO11-964, OPY13-661, OPB11-520, OPW05-766, OPV03-1365 and OPJ16-759, were cloned and sequenced, respectively. Nucleotide sequence data of these RAPD markers had been submitted to the GenBank databases, and the accession numbers are AY491396, DQ493890, DQ493892, AY491398, AY491397, AY392095 and AY392126, respectively. The sequence analysis indicated that there were some homologies for nucleotide sequence between these RAPD markers and V. vinifera or other lives. Besides these, OPO11-964 showed 83% identity with an EST sequence from multiple pathogen-infected sweet orange. OPO11-964 sequence had the biggest open reading frame of 444 bp encoding 147 amino acids and partial identity with 21 protein sequences unknown function from other lives. OPY13-661 sequence showed 89% identity with 2 EST sequences from abiotic stressed leaves of V. vinifera cv. Chardonnay. OPJ16-759 showed 83% to 100% identity with 14 EST sequences from abiotic stressed leaves of V. vinifera var. Chardonnay, and 94% to 100% identity with 3 EST sequences from water stressed berry of V. vinifera cv. Cabsau.
