Chondroprotective Effect of P38 Mitogen Activated Protein Kinase Inhibitor on Experimental Rats Osteoarthritis

Abstract:[Objective] To observe the protective effect of p38 mitogen activated protein kinase inhibitor SB203580 on cartilage of experimental rats osteoarthritis(OA).[Methods] 40 SD rats underwent unilateral anterior cruciate ligament transection(ACLT)and were randomly divided into A,B,C,D groups.Rats of group A and B received 0.1 ml intra-articular injection of SB203580 at 100,10 μm/L after surgery immediately for 6 weeks,once a week.Intra-articular of those in group C received 0.1 ml 0.9% sodium chloride,group D were not injected as control.On the 6th week after surgery,the rats were sacrificed.The cartilage changes,OA grade,cartilage surface class and Mankin inflammation class were compared.[Results] The cartilage changes,cartilage surface class and Mankin inflammation class in group C and D were more severe than those in experimental group A,B.[Conclusion] It is suggested that p38MAPK inhibitor SB203580 might reduce the severity of cartilage degeneration and have the chondroprotective effect.
