Identification and Fungicide Screening of Fungal Species Associated with Walnut Anthracnose in Shaanxi and Liaoning Provinces, China

Abstract:Walnut is cultivated around the world for its precious woody nut and edible oil. Recently, walnut infected by Colletotrichum spp. resulted in a great yield and quality loss. In August and September 2014, walnut fruits with anthracnose were sampled from two commercial orchards in Shaanxi and Liaoning provinces, and five representative isolates were used in this study. To identify the pathogen properly, four genes per region (internal transcribed spacer, glyceraldehyde-3-phosphate dehydrogenase, actin, and chitin synthase) were sequenced and used in phylogenetic studies. Based on multilocus phylogenetic analysis, five isolates clustered with Colletotrichum fioriniae, including its ex-type, with 100% bootstrap support. The results of multilocus phylogenetic analyses, morphology, and pathogenicity confirmed that C. fioriniae was one of the walnut anthracnose pathogens in China. All 13 fungicides tested inhibited mycelial growth and spore germination. Flusilazole, fluazinam, prochloraz, and pyraclostrobin showed the strongest suppressive effects on the mycelial growth than the others, the average EC50 values ranged from 0.09 to 0.40 μg/ml, and there was not any significant difference (P < 0.05). Pyraclostrobin, thiram, and azoxystrobin were the most effective fungicides on spore germination (P < 0.05), and the EC50 values ranged from 0.01 to 0.44 μg/ml. Pyraclostrobin, azoxystrobin, fluazinam, flusilazole, mancozeb, thiram, and prochloraz exhibited a good control effect on walnut anthracnose caused by C. fioriniae, and preventive activities were greater than curative activities. Pyraclostrobin at 250 a.i. μg/ml and fluazinam at 500 a.i. μg/ml provided the highest preventive and curative efficacy, and the values ranged from 81.3 to 82.2% and from 72.9 to 73.6%, respectively. As a consequence, mancozeb and thiram could be used at the preinfection stage, and pyraclostrobin, azoxystrobin, flusilazole, fluazinam, and prochloraz could be used at the early stage for effective prevention and control of walnut anthracnose caused by C. fioriniae. The results will provide more significant instructions for controlling the disease effectively in northern China.

Abstract:Abstract Leaf-associated microbiota is vital in plant-environment interactions and is the basis for micro-ecological regulation. However, there are no studies on the direct differences in microbial community composition between disease-susceptible and healthy walnut leaves. This study collected five samples of healthy and infected leaves (all leaves with abnormal spots were considered diseased leaves) from May to October 2018. Differences in fungal diversity (Chao1 index, Shannon index, and Simpson index) and community structure were observed by sequencing and analyzing diseased and healthy leaf microbial communities by Illumina HiSeq sequencing technology. The main fungal phyla of walnut leaf-associated were Ascomycota , Basidiomycota , and Glomeromycota . Diversity indices (Shannon and Chao1 index values) of healthy leaves differed significantly in the late stages of disease onset. The results showed that the fungal species that differed considerably between the healthy and infected groups differed, and the fungal species that differed significantly between the healthy and infected groups changed with the development of the leaf disease. Critical control time points were determined by analyzing the population dynamics of pathogenic fungi. Leaf-associated microorganisms are abundant and diverse, and fungal identification and diversity studies are helpful for developing more appropriate walnut management strategies

Abstract:Chemical fungicides were applied to suppress pathogen mycelial growth and conidial germination in the laboratory, and to control a gummosis disease in Japanese apricot and peach orchards caused by Botryosphaeria dothidea and Lasiodiplodia theobromae (Syn. = Botryodiplodia theobromae). Field applications were made by spraying or painting after lesion-scraping. Thiophanate-methyl consistently inhibited mycelial growth and conidial germination and controlled disease development in Japanese apricot trees. Recover No. 843 (humic acid plus other chemical elements) provided the best disease control on both tree types in the field and improved plant tissue regrowth in the area of the lesion. The discrepancies between smaller and larger lesions on trees of various ages were also discussed.

Abstract:Establishing a species-diversified cropping system to control crop diseases, insect pests and weeds is an important approach to sustainable agricultural development. This paper reviewed the researches on paddy field species-diversified cropping systems at home and abroad, and discussed the controlling effects and mechanisms of multiple species coexistence on rice diseases, pests and weeds control. The multiple species coexistence models such as rice-fish, rice-duck, rice-azolla-fish and rice-azolla-duck had effective controlling effects on Thanatephorus cucumeris (Frank) Donk, Nilaparata lugens Stal, Chaphalocrocis medinalis Gueneeand, and weeds. Other models like intercropping rice with Zizania caduciflora L. and with other wetland crops also had effective effects in controlling the occurrence and spread of Pyricularia grisea. Abstract:Introduction: Walnut blight is a serious bacterial disease that affects the yield and quality of walnuts. Pantoea agglomerans is one of the main causative agents of walnut blight. However, there have been few studies on the response of walnuts to P. agglomerans infection. Methods: In this study, the soluble sugar, photosynthesis, antioxidant enzyme activities, and secondary metabolites were measured, and the transcriptomic analysis was performed to determine the response of walnut tissue cultures to P. agglomerans infection. Results: After pathogen inoculation, the soluble sugar content decreased, and photosynthesis was inhibited. Antioxidant enzyme (superoxide dismutase and peroxidase) activities and secondary metabolites (phenol and flavonoid) contents increased, especially in the early stages of inoculation. Transcriptomic analysis revealed that the phenylpropanoid biosynthesis pathway is induced after infection, and pathogen infection promotes ABA and ethylene signal transduction and inhibits auxin signaling. In addition, SA and JA-related gene expression was altered after inoculation with P. agglomerans , and the FLS- and calcium-mediated disease resistance signaling pathways were activated. Furthermore, our results suggested an involvement of the R-protein RPM-mediated disease resistance pathway in the response of walnuts to bacterial infections. Discussion: Our findings indicated that phenylpropanoid biosynthesis, hormone signal transduction, and plant-pathogen interaction have key roles in pathogenic inoculation, which provide insights into the molecular mechanisms in the response of walnuts to P. agglomerans infection.

Abstract:The Rice black-streaked dwarf viral disease(RBSDV),transmitted by the small planthopper Laodelphax striatellus Fallen,is one of the most destructive diseases in rice.In order to control this disease,a provincial collaborative research was conducted across the Zhejiang province,China,during 2000 to 2005.The epidemiology,causes of resurgence and damage-yield loss relationship were understood and were applied to design the field sampling and forecast methods,and to determine the economic thresholds of the disease through control of the insect vector.A provincial disease alert system including the GPS,GIS and RS approaches,and an estimation model for virus-infestation in rice were established and were used for long-term forecast of the disease.A new management strategy was proposed which integrated the extension of RBSDV disease-and vector-resistant varieties,improvement of seedling nursery,healthy cultivation of diseased hill gap-filling by removing tillers from nearby healthy hills,and control of insect vectors in the seedlings stage to protect rice plants from the viral disease in field stages.An integrated RBSDV disease management technical system was therefore shaped for the Yangtze Delta region,and applied in large scale,effectively reduced the disease occurrence and damage,achieved significant economic and social effects.

Abstract:English walnut ( Juglans regia ), has high economic and ecological value. As an important tree species for eliminating poverty, it is planted in many Provinces of China. In 2021, new pathogenic fungi were observed in English walnut in Guangyuan City, Sichuan Province, China. The initial symptom of leaf infection is that the leaves are covered with small black spots, which gradually expand into larger brown spots. Most of the spots appeared at the edges of the leaves, and yellow whorls were observed at the junction between the spots and the healthy leaves. The pathogenic fungi were isoalted form collecting disease samples and purified by single-spore culturing. In vitro and field experiments showed that the pathogen could cause brown spots on walnut leaves. The inoculation experiment showed that the symptoms in the field experiment were the same as those observed on the spot; however, slight differences were observed in the in vitro experiment. Ten isolates were obtained from walnut leaves with brown spot symptoms, and these were further characterized based on morphology and DNA sequencing. ITS (internal transcribed spacer), LSU (large sub-unit rDNA), rpb 2 (second largest subunit of RNA polymerase) and tub 2 (beta-tubulin) gene regions were used to construct phylogenetic trees and determine the evolutionary relationships among the collected strains. The isolate was identified as Nothophoma quercina by morphological and polygene analyses. As far as we are aware, the brown spots on walnut leaves caused by N. quercina is the first report of its kind.

Abstract:Walnut ( Juglans regia L.) is a widely grown nut plant worldwide, including in Guizhou Province, located in southwest China. The high quality and special taste make Guizhou walnuts, particularly those produced in Hezhang County, a "Chinese National Geographical Indication Product" that substantially contributes to the local economy and grower's income. In July 2022, a serious occurrence of leaf spot disease was observed in a walnut plantation area, Shuitang Town, Hezhang County, Guizhou Province, China (27°07′67′′N, 104°64′61′′E). The causal agent was identified as Didymella segeticola through morphological characterization and amplification and sequencing of the internal transcribed spacer (ITS) region, beta-tubulin ( TUB ) gene, and glyceraldehyde-3-phosphate dehydrogenase ( G3PD ) gene. Koch's postulates, including re-isolation and identification, were performed to confirm its pathogenicity on healthy leaves. To our knowledge, this is the first report of D. segeticola causing leaf spot on walnuts worldwide. Further, to determine its biological characteristics, which could be utilized for future disease management, the effects of temperature, light, and carbon and nitrogen resources on mycelial growth, conidia production, and conidia germination and the effects of humidity on conidia germination were studied. The optimum temperature for mycelial growth of representative strain D. segeticola C27 was 20°C. Increasing the light period significantly decreased conidia production and conidia germination. Maltose and beef extract were the best carbon and nitrogen sources, respectively, for the pathogen. Conidia germination was enhanced at 90% humidity. In vitro screening of effective fungicides was conducted. Among the 20 screened fungicides, difenoconazole showed the best inhibition rate, with an EC 50 (concentration for 50% of the maximal effect) of 0.0007 μg/mL. Tetramycin also showed sufficient inhibitory effects against D. segeticola , with an EC 50 value of 0.0009 μg/mL. Our study provides new insights into the causal agent of walnut leaf spot in Guizhou, China, as well as the first pathogen characteristics and promising candidate fungicides for its management.

Abstract:Walnut is one of the important economic tree species in the world. For the past few years, with the continuous expansion of walnut planting area, walnut bacterial blight caused by Xanthomonas campestrispv. juglandis has critically affected walnut production and caused economic losses. In this research, by comparing the gene sequences of X. campestrispv. juglandis and its closely related bacterial species, the 16S rDNA gene sequence was selected, and the specific primers were evaluated using PCR. Subsequently, the real-time fluorescence PCR specific detection method of X. campestrispv. juglandis was established and optimized. The specificity of real-time PCR was verified for 23 strains isolated from walnut and its surrounding soil, including eight target pathogens. The specific amplification was possible even when the cDNA template concentration was 7.0×10-5 μg/mL, hence, the detection sensitivity of the standard method is 7.0×10-5 μg/mL. The reliability of the established real-time PCR detection method in field detection was verified by the identification of target bacteria in 8 randomly collected healthy leaves and leaves of suspected walnut blight. This method laid a foundation for the early diagnosis of X. campestrispv. juglandis.

Abstract:To define the relationship between the severity of maize dwarf disease and yield loss,experiments were conducted in Lin'an and Tonglu,Zhejiang Province,from 2011 to 2013.The disease incidence,damage and yield losses were studied on the widely-grown maize varieties'Jundan 20'and'Jundan 18'.The results showed that compared with healthy maize plants,infected plants had lower height,shorter internodes and fruiting ears,narrower and shorter leaves,fewer filled grains and lower grain weight.The biomass of plants with disease grades of 1-4 decreased by 28.76％-81.31％,grain yield per ear decreased by 32.29％-98.91％.Yield losses (1.03％-32.13％) were significantly correlated with disease incidence (2.31％-39.44％).The formula of the relationship between damage and yield loss was developed by regression analysis.The economic threshold was calculated and the index was set for the disease control on the basis of the current conditions and economic benefits of maize production.

Swingle , in order to reduce their harm and accelerate the production.Methods Literature search and field investigation were used.Results The status and control measures of such diseases as tristeza,huanglongbing,authracnose and cankerous disease were summarized,and the reasons and spread fashions of them were analyzed.Moreover, the general control measures were established.Conclusion To control them effectively, prevention is most important, and agricultural methods,biologic methods and chemical germicides should be used together.

Abstract:Aimed to realize high yield and high quality growing of fine varieties of walnut,integrated measures including low-cost orchard establishment,training and pruning,fertilization,irrigation,flower management,frost protection,pest and disease control were studied in this paper.The results show that these measures combination worked effectively in realizing the expected target with the 254.65 kg/667 m2 of a yield of 5-year-old Xiangling,Grafted after rootstock planting and 342.54 kg/667 m2 of a yeild of 5-year-old Yuanfeng,Grafted after rootstock planting respectively.

Abstract:The traditional rice-fish ecosystem is one of the outstanding examples of the globally important agricultural heritages and is a sustainable form of agriculture.By field sampling and controlled experiments,we studied the application of pesticides and occurrence of disease,pests and weeds through the rice production process in the ecosystem.The results obtained from field sampling showed that there is no significant difference of rice grain yield found between the rice monoculture and the traditonal rice-fish coculture.The usage of agrochemicals for disease,pest and weeds control in rice-fish coculture ecosystem was 3.13±0.31 kg.hm-2 fewer that applied in rice monoculture ecosystem.,and the times of pesticides application in traditional rice-fish coculture was 3.4 less than that in rice monoculture ecosystem.The usage of agrochemicals was significantly negatively correlated with the fish population density in the traditional rice-fish coculture ecosystem.The results from field experiments showed that the rice grain yield in traditional rice-fish coculture ecosystem was significantly higher than that in rice monoculture with chemically control of disease,pests and weeds.The occurrence of diseases,pests and weeds in tradtional rice-fish coculture significantly decreased.

Abstract:Detecting and classifying diseases in agricultural crops is crucial for ensuring crop health and maximizing yields. In the case of walnut trees, timely identification of fungal diseases is essential to prevent their spread and minimize economic losses. Traditional methods of disease diagnosis rely on visual inspection by experts, which can be time-consuming and prone to human errors. In this study, we propose an intelligent system for the detection and classification of walnut fungi diseases using machine-learning techniques. The system leverages the power of image processing and pattern recognition algorithms to automate the identification process, enabling rapid and accurate diagnosis. The system is trained using a dataset comprising high-resolution images of healthy walnut leaves and leaves affected by various fungal diseases. We extract relevant features from the images and employ state-of-the-art machine learning algorithms, such as back-propagation neural network (BPNN), to learn the complex patterns associated with each disease. To evaluate the performance of our system, we conducted extensive experiments on a real-world dataset, achieving high accuracy rates for disease detection and classification. The system successfully identifies common walnut fungi diseases, including walnut blight, anthracnose, and powdery mildew, with accuracy rates exceeding 90%. Moreover, it exhibits robustness against variations in lighting conditions and leaf orientations. Our results demonstrate the potential of machine learning techniques in revolutionizing disease diagnosis in the agricultural domain. The proposed system offers a cost-effective and scalable solution for farmers and agronomists to monitor the health of walnut trees, detect diseases at an early stage, and apply targeted treatments. By providing accurate and timely diagnoses, this technology can contribute to reducing crop losses and improving overall productivity in walnut cultivation.
