Genetic Transformation of Potato with Bt-CryV Gene

Abstract:Bt-CryV gene was transferred into the potato cultivar Dongnong 303 by agrobacterium-mediated method. Microtuber discs were used as explants. A regeneration system for potato transformation was established when several factors, such as the kind and concentration of different phytohormones and antibiotics, the dormant stages of microtuber, the time of co-culture, and the method of selection, were optimized. The 21 transgenic plants were regenerated and selected on MS basal medium supplemented with Kan 100 mg·L-1, of which 10 transformants were confirmed by the test of PCR and PCR-Southern Blot.
