Abstract:The practical and diversified functionalization of ubiquitous C(sp3)–H bonds is an economically attractive yet chemically challenging approach to build molecular complexity from abundant chemical raw materials. Herein, we have identified a very robust and practical FeCl3/HCl photocatalytic system that enables greatly efficient C–H alkylation, oxidation, chlorination, fluorination, amination, alkynylation, and sulfonylation with total turnover number (TON) of up to 9,900. The aliphatic C–H activation is mediated by chlorine radical, in situ generated from a photoactive tetrahedral [FeIIICl4]− via ligand-to-metal charge transfer excitation, and the use of HCl as co-catalyst significantly improves the total TON by more than one order of magnitude.

Abstract:Vinyl cations exhibit remarkable reactivity towards arene C-H functionalizations. This computational study revealed the key mechanistic details of intramolecular C-H vinylation through a vinyl cation intermediate. Based on the reaction mechanism, the effects of substitution, ring strain and tether length on the reactivity of the vinyl cation were elucidated.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Herein, the first electrochemical strategy for direct acyloxylation of benzylic C(sp3)–H with amides is described. The method features transition metal- and oxidant-free conditions with H2 evolution, the excellent functional group (including fluoro, chloro, bromo, iodo, cyano, trifluoromethyl, aldehyde, ketone, carboxyl, ester, and ether) tolerance, up to 82% yield in an operation-friendly undivided cell under the open air, and ease of scale up. A number of well-designed mechanistic experiments [e.g., radical trapping, electron paramagnetic resonance (EPR) and isotope labeling] strongly support the radical-involved process of benzylic C(sp3)–H functionalization and the process of amide C–N bond breaking.

During the recent years, significant progress has been made in this fascinating field with various functionalized alkenes, heterocycles and carbocycles being obtained. This article will focus on the recent achievements in the field of directing-group-assisted vinylic C-H bond functionalization.

Abstract:A chlorobenzylation of terminal alkynes through Fe(II)promoted benzylic C(sp3) H bond functionalization in the presence of NCS as a chloride source was developed. Compared with previous methods to prepare polysubstituted alkenyl halides, the presented procedure provides an efficient alternative with high atom and step economy under mild conditions. The transformation was established to proceed through a single -electron transfer (SET) process with benzyl cations as key intermediates.

Abstract:A copper-catalyzed 1,1-difunctionalization of terminal alkynes was achieved via a three-component reaction, providing a variety of vinyl sulfones with good yields and excellent chemo- and stereoselectivity. Preliminary mechanistic studies indicated that the reaction probably underwent a Cu-catalyzed formal C–H insertion to produce an allene intermediate, which was then trapped by a sulfonyl anion to give the corresponding product.

Abstract:Abstract Methods that enable site selective acylation of sp 3 C-H bonds in complex organic molecules are not well explored, particularly if compared with analogous transformations of aromatic and vinylic sp 2 C-H bonds. We report herein a direct acylation of benzylic C-H bonds by merging N-heterocyclic carbene (NHC) and photoredox catalysis. The method allows the preparation of a diverse range of benzylic ketones with good functional group tolerance under mild conditions. The reaction can be used to install acyl groups on highly functionalized natural product derived compounds and the C-H functionalization works with excellent site selectivity. The combination of NHC and photoredox catalysis offers options in preparing benzyl aryl ketones.

Abstract:The direct functionalization of C–H bonds has drawn the attention of chemists for almost a century. C–H activation has mainly been achieved through four metal-mediated pathways: oxidative addition, electrophilic substitution, σ-bond metathesis and metal-associated carbene/nitrene/oxo insertion. However, the identification of methods that do not require transition-metal catalysts is important because methods involving such catalysts are often expensive. Another advantage would be that the requirement to remove metallic impurities from products could be avoided, an important issue in the synthesis of pharmaceutical compounds. Here, we describe the identification of a cross-coupling between aryl iodides/bromides and the C–H bonds of arenes that is mediated solely by the presence of 1,10-phenanthroline as catalyst in the presence of KOt-Bu as a base. This apparently transition-metal-free process provides a new strategy with which to achieve direct C–H functionalization.

Abstract:Direct C–H functionalization has recently emerged as one of the most efficient strategies to access structurally complex molecules from readily accessible feedstocks in an atom- and step-economic manner. In particular, enantioselective C–H activation has garnered increasing attention by enabling chemists to efficiently assemble valuable chiral compounds by asymmetrically manipulating C–H bonds into useful functionalities. Apart from the extensively studied C–C bond formation, very few endeavors have been focused on the C–X formation analogs. Motivated by the utility of the latter approach in constructing academically and industrially important heteroatom-containing chiral compounds, we provide herein an overview on C–X forming asymmetric C–H activation reactions proceeding through C–H metalation. The advancements are organized according to the employed catalytic systems, which include Pd(II) catalysis, group-9 CpxM(III) catalysis, monovalent group-9 metal catalysis, and multi-boryl/silyl Ir(III) catalysis, with emphasis on the design philosophy, mechanism, and mode of enantiocontrol.

Abstract:The results from a direct palladium-catalyzed cross-coupling reaction between a variety of olefins with acrylate through vinylic C-H activation that give the corresponding butadienes in moderate to good yields and stereoselectivities are reported. Given the reaction's ability to tolerate a wide range of styrenes, aliphatic alkenes and acrylates, this atom- and step-economical methodology offers a straightforward method to forge new C-C bonds of the valuable diene products from readily available starting materials under very mild reaction conditions.
