Stromal cell derived factor-1 alpha promotes neovascularization during cutaneous wound healing

Abstract:BACKGROUND:Stromal cell derived factor-1α(SDF-1α)has angiogenesis property,which belongs to the family of CXC chemokines,and plays an important role during tissue/organ injury healing.However,the effect of SDF-1αon cutaneous wound healing is poorly understood.OBJECTIVE:To investigate the effects of SDF-1αon neovascularization during cutaneous wound healing.DESIGN,TIME AND SETTING:The design was random control animal experiment,which was finished at the Sixth Department,Research Institute of Surgery,Daping Hospital,and the Third Military Medical University from June to August of 2008.MATERIALS:Sixty SPF-grade Kunming mice,irrespective of the gender,with weight of 25-30 g.The SDF-1αantibody was provided by Santa Cruz Biotechnology Company(USA).METHODS:Mice full-thickness wound models were prepared,and divided into control,10 mg/L and 20 mg/L antibody groups,with 20 animals in each group.Anti-SDF-1αwith 10 mg/L and 20 mg/L were injected into full-thickness wound site of mice for successive 6 days after trauma.MAIN OUTCOME MEASURES:CD146 mRNA in wound site was detected by reverse transcription-polymerase chain reaction(RT-PCR)analysis,and the microvascular density and rate of wound contraction were observed at days 3,4,5,6,7 after trauma.RESULTS:Compared with the control group,the expressions of CD146 mRNA and microvascular density of 10 mg/L and 20 mg/L of anti-SDF-1α groups were greater at days 3 and than days 5,6,7,which was smaller than the control group at each day after trauma(P < 0.05).The rate of wound contraction of 10 mg/L in the anti-SDF-1αgroup was decreased than the control group at days 6 and 7,and that of 20 mg/L was lower than the control group at days 5,6 and 7(P < 0.05),but there was significant difference between 10 mg/L and 20mg/L of anti-SDF-1αgroups at day 7(P < 0.05).CONCLUSION:Neovascularization of wound can be decreased by blocking SDF-1α expression,which may influence the wound healing progress.
