Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:Ethyl and phenyl oxanilates were readily prepared from N-(2-bromobenzyl)anilines and oxalyl chloride monoethyl and monophenyl esters, respectively. It was found that the ethyl oxanilate survived in the presence of K2CO3 in DMA at 120 degrees C and underwent an intramolecular direct arylation using Pd(OAc)(2)-dppf, furnishing the 5,6-dihydrophenanthridine derivative. In contrast, the corresponding phenyl oxanilates decomposed upon exposure to K2CO3 in DMA at 120 degrees C and were transformed into N-arylisoindolin-1-ones via Pd(OAc)(2)-dppf-catalyzed intramolecular decarbonylative coupling. Except for the 4-methoxy-substituted oxanilic acid phenyl ester, other phenyl oxanilates possessing electron-withdrawing (NO2, Cl) and weak electron-donating (Me) substituents provided the N-arylisoindolin-1-ones in 43-80% yields.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:Certain imines lead to the formation of imidazolidine derivative in THF with Sni/TiCl4 system. It was found that products of reductive coupling could be obtained in below 20% yields without 1,2-dibromoethane, while products of reductive coupling in moderate yields when I 2-dibromoethane was added. The structures of all products were confirmed by IR, H-1 NMR and MS spectra. According to the results of the experiment, a possible mechanism was proposed.

Abstract:A formal (3+2) annulation reaction between isatin-derived N-tosylhydrazones and o-(trimethylsilyl)aryl triflates allows facile synthesis of spiro[indazole-3,3'-indolin]-2'-ones in high to excellent yields. Furthermore, indazolo[2,3-c]quinazolin-6(5H)-ones could be accessed from the same starting materials in one-pot fashion via an annulation/rearrangement pathway.

Abstract:An one pot method of synthesis of 1,2,4,5-tetraphenyl-1,3-imidazole derivatives was developed.Six products were synthesized using benzoyl(1), benzaldehyde(2a), aniline(3) and ammonium acetate(4) as starting material with immobilized montmorillonite/iodine as catalyst.The structures were confirmed by 1H NMR and IR.The reaction conditions were optimized.The results showed that under the optimum reaction conditions{using 10 mol%MMT/I2 as catalyst, 10 mol%DABCO as base additive, and n-butanol as the solvent, raw material molar ratio r[n(1)∶n(2a) ∶n(3) ∶n(4)]=1.0 ∶2.0 ∶2.0 ∶8.0, reaction at 110 ℃ for 16 h}, the highest yield was 91.5%.

Abstract:(2S, 3R)-2-acetooxymethanyl-3-(P-nitrophenyl)-N-tosylaziridine 5 was synthesized from (1S, 2S)-2-amino-1-(4-nitrophenyl)-1, 3-propanediol 1 in four steps with a 24.8% overall yield. This reaction is stereospecific and occurs an expectable configuration inversion at α-carbon atom of benzyl group. The structure of substituted aziridine was determined on FT-IR, NMR, MS and so on.

Abstract:We developed an efficient method for synthesis of tetrafunctionalized 2-imidazolines using TfOH-catalyzed formal [3+2] cycloaddition of N-tosylaziridine dicarboxylates and nitrites. This is the first report of C-N bond cleavage of N-tosylaziridine dicarboxylates catalyzed by Bronsted acid and the reaction worked well over wide scope of substrates in good to excellent yields under mild conditions. The method has the potential to be applied to pharmaceutical design and synthesis of tetrafunctionalized 2-imidazolines. (C) 2019 Published by Elsevier Ltd.

Abstract:The synthesis of novel imidazole derivatives via immobilized α-acylamino ketones is reported in this article. The key intermediates were prepared from the Wang-piperazine resin-supported Fmoc-amino acids. After their sulfonylation with 4-nitrobenzenesulfonyl chloride (4-Nos-Cl), followed by alkylation with α-bromoketones and cleavage of Nos group, the resulting α-acylamino ketones were reacted with Fmoc-isothiocyanate. The corresponding Fmoc-thioureas were subjected to the Fmoc-cleavage and spontaneous ring-closure to imidazole scaffold. The resulting imidazole-thiones were alkylated with alkyl halides and oxidized using meta-chloroperbenzoic acid ( mCPBA). Trifluoroacetic acid (TFA)-mediated cleavage yielded the corresponding trisubstituted 2-alkylsulfonyl imidazoles in good crude purity and acceptable overall yields. In the case of sulfides, prepared from alkyl bromides, the unexpected products brominated at the C4 position of the imidazole were obtained.

Abstract:In this work, 10 new 4‐(4‐nitro‐1H‐imidazol‐1‐yl)butyl 1‐aryl‐1H pyrazole‐4‐carboxylates 2(a–j) were synthesized, in 18%–85% yields, from N‐alkylation reaction of 4(5)‐nitroimidazole. Imidazole is one of the most important heterocyclic rings. This nucleus can be found in a plethora of substances in different knowledge areas such as medicinal, agrochemical, polymer, and dyestuff. N‐alkylation reactions involving N − 1 atom is a remarkable tool to synthesize many imidazole derivatives which can be attached to a great variety of functional groups. In this work, we synthesized 10 new intermediates 4‐chlorobutyl 1‐aryl‐1H‐pyrazole‐4‐carboxylates 3(a–j) from 1‐aryl‐1H‐pyrazole‐4‐carbonyl chlorides, obtained from 1‐aryl‐1H‐pyrazole‐4‐carboxylic acids 4(a–j), and 4‐chlorobutan‐1‐ol 1, generated by THF ring‐opening promoted by hydrochloric acid. Compounds 3(a–j) promoted N‐alkylation of 4(5)‐nitro‐1H‐imidazole to afford the targets 4‐(4‐nitro‐1H‐imidazol‐1‐yl)butyl 1‐aryl‐1H‐pyrazole‐4‐carboxylates 2(a–j) in 18%–85% yields.

Abstract:A one-pot procedure has been developed to prepare the derivatives of 2-arylimidazo(2, 1-a)-isoquinolines (2a similar to 2b), 2-arylimidazo(1, 2-a) quinolines (4a similar to 4b) and 2-arylimidazo(1, 2-a) phthalazines (6a similar to 6c) in moderate yields (51% similar to 62%) by the reaction of isoquinolinium salts, quinolinium salts or phdialazinium salts with ammonium acetate in the presence of tetrakispyridine cobalt(II) dichromate (TPCD).

Abstract:The umpolung of N-tosylhydrazones was reported for the first time. Synthesis of N-cyano tosylhydrazones was developed, and various N-cyano tosylhydrazones were prepared in good yields. An umpolung N-tosylhydrazones mechanism was proposed. Meanwhile, the [3,3]-sigmatropic rearrangement of N-cyano tosylhydrazones was examined, and the corresponding 2-aminoimidazoles were given.

Abstract:Imidazoles annulated to saturated six‐membered cycles featuring S, SO2, NH, NCbz have been prepared in bulk quantities. The starting previously unknown cyclic α‐amino ketones were obtained via the Neber rearrangement. Unstable tosyl ketoximes were used without purification immediately after their preparation. Marckwald reaction with desulfurization using Raney nickel led to the formation of the annulated imidazoles. A synthetic approach to imidazoles annulated to saturated six‐membered cycles featuring S, SO2, NH, NCbz was developed. It was achieved by combining the Neber rearrangement and the Marckwald reaction. The Neber rearrangement applied to cyclic ketones allowed us to prepare in hundred gram quantities previously unknown α‐amino ketones. Unstable tosyl ketoximes were used without purification immediately after their preparation. α‐Amino ketones react with potassium thiocyanate and cyanate in almost a quantitative yield affording highly pure imidazol‐2‐thiones or imidazol‐2‐ones. Desulfurization of imidazole‐2‐thiones using Raney nickel led to the formation of previously unknown 2‐unsubstituted fused imidazoles in high yields.

Abstract:We report herein a general and effective system achieving cyclization of β-trifluoromethyl enones with amidines in the presence of 1,3-diiodo-5,5-dimethylhydantoin (DIH), which affords a range of trifluoromethylated 2-imidazolines in synthetically useful yields with good diastereoselectivities (up to 95% yield, up to 98:2 dr) and good functional group tolerance. Furthermore, the one-pot synthesis of trifluoromethylated imidazoles via sequential cyclization and oxidation is demonstrated. More significantly, the reaction mechanism was verified by ESI-MS studies of possible intermediates, and a reasonable reaction mechanism was proposed.
