Abstract:Eg5 is identified as a microtubule-dependent motor protein and has been suggested to play important role in cancer development. The aim of this study was to detect the expression of Eg5 and its clinicopathological characteristics in patients with laryngeal squamous cell carcinoma. One-step quantitative reverse transcription-polymerase chain reaction (qPCR) test with 20 fresh-frozen LSCC. samples and immunohistochemistry (IHC) analysis in 137 LSCC cases were performed to investigate the relationship between Eg5 expression and the clinicopathological characteristics of LSCC. The Eg5 mRNA and protein levels were significantly higher in LSCC tissues compared with corresponding non-cancerous tissues (p < 0.05). Eg5 protein expression was significantly correlated to lymph node metastasis (p = 0.021) and TNM stage (p = 0.030). Kaplan-Meier survival and multivariate analysis with Cox regression model indicated that high Eg5 expression (p = 0.031) and TNM stage (p = 0.011) were independent factors to predict unfavourable prognosis for patients with LSCC. The data suggested that Eg5 may be identified as a novel prognostic biomarker and targeting Eg5 seems to be a novel strategy for LSCC treatment.
Expression of LN-5 in lung adenocarcinoma tissue and its relationship with EGFR mutations

Abstract:Objective To investigate the clinical significance of the expression of laminin-5 (LN-5) in lung adenocarcinoma and its relationship with mutations of epidermal growth factor receptor (EGFR). Methods From January 2015 to December 2015, specimens from 57 cases of lung adenocarcinoma and adjacent normal tissues with pathological confirmation after surgical resection were collected. LN-5 expression was detected using immunohistochemical SP methods, and EGFR mutations were assessed using liquichip technology. The expression of LN-5 and its relationship with EGFR mutations were analyzed. Results The positive expression of LN-5 in adenocarcinoma tissues was 21.1％ (12/57), higher than 1.7％ (1/57) of adjacent normal tissues (P<0.05). The incidence of EGFR mutations (E19+E21) was 43.9％ (25/57), while that of wild type+E20 was 56.1％ (32/57). The expression of LN-5 was associated with lymph node metastasis and EGFR mutation (P<0.05), but not with TNM stage (P>0.05). There was a negative correlation between the expression of LN-5 and EGFR mutation(r =-0.283 ,P<0.05). Conclusion The expression of LN-5 is up-regulated in lung adenocarcinoma and is correlated with EGFR mutation status.
Effect of Fibulin-5 on Cell Proliferation and Invasion in Human Gastric Cancer Patients

Abstract:Objective: To explore the effect of Fibulin-5 expression on cell proliferation and invasion in human gastric cancer patients. Methods: Fibulin-5 expression was detected in 56 samples of surgically resected gastric cancer and paired noncancerous tissues using qRT-PCR and immunoblotting. Fibulin-5 was knocked down by Fibulin-5 shRNA in MGC-803 cells, then BrdU cell proliferation and transwell invasion assays were used to determine cell proliferation and invasion. Results: The level of Fibulin-5 mRNA in gastric cancer tissues was significantly higher as compared with that in normal tumor adjacent tissues (P<0.05). Otherwise, the level of Fibulin-5 protein in cancer and noncancerous tissues was consistent with mRNA expression (P<0.05). Fibulin-5 protein expression in tumor tissues with poorly differentiated, lymph node metastasis and advanced TNM tumor stage was significantly higher (P<0.05, respectively). Fibulin-5 was obviously knocked clown by Fibulin-5 shRNA (P<0.05), and Fibulin-5 knockdown significantly inhibited cell proliferation and invasion in MGC-803 cells (P<0.05, respectively). Conclusions: The high expression of Fibulin-5 is associated with the malignant clinicopathologic parameters in gastric cancer and Fibulin-5 knockdown inhibits cell proliferation and invasion in MGC-803 cells, suggesting Fibulin-5 may act as a key factor in the progression of gastric cancer.

Abstract:To investigate the expression of extracellular matrix in neo-adjuvant chemotherapy and control group, and its relationship with clinical significance in non-small cell lung cancer. Expression of extracellular matrix in lung cancer tissues was detected in 73 patients by using immunohistochemistry Envision method. The median survival of 14 patients with low expression of fibronetin was 67month, 57 patients with positive expression was 38month. A significant difference existed between the two groups (P=0.048). The high expression of laminin in neo-adjuvant chemotherapy group were negatively related to clinical response.When analyzed only patients randomized to neo-ajuvant chemotherapy using cox step-wise regression analysis, the TNM stage is the only prognosis factor to survival. Overexpression of some extracellular matrix may be involved in the negative chemoresponse rate. The survival time of fibronectin and laminin negative patients may be longer than positive patients.

Abstract:PURPOSE:Recently, mutations in the epidermal growth factor receptor (EGFR) gene were reported to correlate with EGFR tyrosine kinase inhibitor response in advanced non-small cell lung cancer (NSCLC). In this study, we attempted to detect EGFR mutations in plasma and pleural effusion samples and to make clear its correlations with gefitinib response and survival in NSCLC patients.METHODS:The free DNA was isolated from the plasma of 56 cases and pleural effusion of another 32 cases of advanced NSCLC. Five common types of EGFR mutations were analyzed by LightCycle PCR with Taqman-MGB probes.RESULTS:EGFR gene mutations were found in 22 of all the 88 (25%) NSCLC patients (23.2% of 56 plasma samples, 28.1% of another 32 pleural effusion samples). EGFR mutations were more frequently present in females, never-smokers and adenocarcinomas (P < 0.01). It also showed that patients with EGFR mutations had a significantly better response rate when compared with that of the wild-type patients (P < 0.001). The median progression-free survival (11.2 vs. 2.7 months P = 0.005) and overall survival (21.8 vs. 5.8 months P = 0.003) were significantly higher in patients with EGFR mutations than in patients with wild-type EGFR.CONCLUSIONS:The EGFR mutations in the serum and the pleural effusion from advanced NSCLC patients can be detected with LightCycle PCR using Taqman-MGB probes. The mutations highly predict the efficacy of gefitinib in advanced NSCLC.
Relationship Between Mutations of Epidermal Growth Factor Receptor in the Plasma and Pleural Effusion and Responses to Gefitinib in Advanced Pretreated Non-Small Cell Lung Cancer

Abstract:BACKGROUND:Recently,mutations in the epidermal growth factor receptor(EGFR) gene were reported to correlate with EGFR tyrosine kinase inhibitor(TKI) response.In this study,relationship between mutations of EGFR in plasma and pleural effusion and responses to gefitinib was investigated in pretreated patients with advanced non-small cell lung cancer(NSCLC).METHODS:The circulating free DNA was isolated from the plasma of 53 cases and pleural effusion of another 10 cases and analysed for EGFR mutations by LightCycle PCR method.Relationship between EGFR mutations and response to gefitinib was analysed with Chi-square test.RESULTS:EGFR mutations were found in 17 of 63 cases(27.0%).EGFR mutations were frequently present in females(P=0.024) and non-smokers(P=0.021).Patients with EGFR mutations had a significantly better response rate compared to that of the wild-type patients(P=0.000).CONCLUSIONS:The EGFR mutations exist in the plasma and the pleural effusion from patients with advanced NSCLC.These mutations can be detected with LightCycle PCR method which is highly specific and sensitive and easy to perform compared with the direct sequencing.It will enable us to get a potential implication in the diagnosis and targeted therapy of NSCLC at late stage.

Results: Univariate analysis found that the median survival period was 67 months in 14 patients with low expression of fibronectin and 38 months in 57 patients with positive expression of fibronectin. A significant difference existed between the two groups (P = 0.048). The negative expression of laminin in neo-adjuvant chemotherapy group was related with the increase in clinical response. Multivariate analysis by Cox Proportional Hazards Model found that the TNM stage was the only independent prognostic factor. Negative expression of fibronectin and laminin tended to affect the prognosis of patients, but the difference was not significant. Conclusion: Overexpression of some extracellular matrix may induce decrease in clinical response. The fibronectin and laminin-negative patients may have longer survival time than fibronectin and laminin -positive patients.
The Role of Up-Regulation of Integrin Β1 in Resistance to Gefitinib in Non-Small Cell Lung Cancer

Abstract:Objective: To investigate the effects of up-regulation of integrin β1 on tumor cell proliferation and tumor growth of non-small cell lung cancer (NSCLC) inhibited by gefitinib, a epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI). Methods: Three NSCLC cell lines including PC9, PC9/PCD (transfected with empty plasmid pcDNA 3.1) and PC9/D6 (transfected with integrin β1-expressed plasmid) were treated with 0.5 μmol/L gefitinib. The cell proliferation was detected by MTT assay. The graft tumors from the three cell lines in nude mice were established, respectively, and then they were randomly received 3 mg/kg gefitinib or 1% Tween-80. The tumor growth was observed and the growth curve was drawn. The tumor weight was recorded and the tumor growth inhibition rate was calculated. The expressions of integrin β1, EGFR and pEGFR in tumor tissues were examined by immunohistochemistry. Results: The proliferation rate of PC9/D6 cells transfected with integrin β1 was significantly higher than those of PC9/PCD and PC9 cells (P＜0.05). The graft tumor growth inhibition rates were 61.38%, 93.54% and 95.16% in the PC9/D6, PC9/PCD and PC9 groups, respectively; the differences between the PC9/D6 group and the PC9/PCD and PC9 groups were significant (both P＜0.01). The expression of pEGFR was decreased in the three graft tumor groups after treating with gefitinib; however, the expression levels of pEGFR and integrin β1 in PC9/D6 group were significantly higher than those in the PC9/PCD and PC9 groups (P＜0.01). There was no obvious difference in the expression level of EGFR among the three groups. Conclusion: The up-regulation of integrin β1 may decrease the sensitivity to gefitinib in NSCLC in vitro and in vivo. LGR5 is a Promising Biomarker for Patients with Stage I and II Gastric Cancer.

Abstract:Objective: To investigate Leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5) expressions in gastric cancer and to evaluate its clinical significance.Methods: LGR5 expression was assessed by immunohistochemistry in 257 gastric cancer patients after surgery. The relationships between LGR5 expression and clinicopathological features and patients prognosis were statistically analyzed.Results: The expression of LGR5 was significantly higher in gastric cancers as a cancer stem cell marker than in adjacent normal tissues (P<0.001), and more frequently in patients with intestinal type, well-moderate differentiation and stage I and II (P<0.05). Although we found gastric cancer patients with LGR5 positive expression. had a poorer prognosis, it didn't meet statistical significance (P>0.05). LGR5 negative expression was significantly related to the favorable overall survival in stage I and II gastric cancer patients (P<0.05). Furthermore, patients with high LGR5 expression. tended to be more likely to get progression and have poorer progress-free survival (P<0.05). Muldvariate Cox regression analysis revealed that LGR5 expression was an independent factor of overall survival for the patients with stage I and II gastric cancer (P<0.05).Conclusions: Our results show that LGR5 may play an important role in tumorigenesis and progression and would be a powerful marker to predict the prognosis of patients with stage I and II gastric cancer.
Expression of the laminin genes family and its relationship to prognosis in pancreatic carcinoma

Abstract:Background and study aims: Laminin is an extracellular matrix molecule that is the major component of the basement membrane and plays a key role in regulating various processes. However, the association between the laminin gene family and the prognosis of pancreatic carcinoma has not been systematically investigated. Patients and methods: The role of the laminin gene family in pancreatic cancer was evaluated using data from the TCGA database. The effects of different expressions of members of the laminin gene family on pancreatic cancer survival were compared, and their primary cellular roles were examined. The effects of different expressions of positive family genes on proliferation, metastasis, and invasion, as well as EMT and ferroptosis in pancreatic cancer, were also examined. Results: Based on univariate and multifactorial analysis of pancreatic cancer patients, LAMA3 was identified as an independent prognostic factor for overall survival in pancreatic cancer. LAMA3 was found to be enriched in the actin cytoskeleton, P53 signaling pathway, adhesion molecule junctions, pentose phosphate pathway, and regulatory differences in the cell cycle and focal adhesion. Additionally, high expression of LAMA3 was found to promote cancer proliferation, invasion, and metastasis, facilitate the EMT process, and inhibit ferroptosis. Conclusions: Our results identified LAMA3 was associated with the prognosis of patients with pancreatic cancer and may serve as a prognostic biomarker for pancreatic cancer.
Gefitinib metabolism-related lncRNAs for the prediction of prognosis, tumor microenvironment and drug sensitivity in lung adenocarcinoma

Abstract:The complete compound of gefitinib is effective in the treatment of lung adenocarcinoma. However, the effect on lung adenocarcinoma (LUAD) during its catabolism has not yet been elucidated. We carried out this study to examine the predictive value of gefitinib metabolism-related long noncoding RNAs (GMLncs) in LUAD patients. To filter GMLncs and create a prognostic model, we employed Pearson correlation, Lasso, univariate Cox, and multivariate Cox analysis. We combined risk scores and clinical features to create nomograms for better application in clinical settings. According to the constructed prognostic model, we performed GO/KEGG and GSEA enrichment analysis, tumor immune microenvironment analysis, immune evasion and immunotherapy analysis, somatic cell mutation analysis, drug sensitivity analysis, IMvigor210 immunotherapy validation, stem cell index analysis and real-time quantitative PCR (RT-qPCR) analysis. We built a predictive model with 9 GMLncs, which showed good predictive performance in validation and training sets. The calibration curve demonstrated excellent agreement between the expected and observed survival rates, for which the predictive performance was better than that of the nomogram without a risk score. The metabolism of gefitinib is related to the cytochrome P450 pathway and lipid metabolism pathway, and may be one of the causes of gefitinib resistance, according to analyses from the Gene Set Enrichment Analysis (GSEA), Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Immunological evasion and immunotherapy analysis revealed that the likelihood of immune evasion increased with risk score. Tumor microenvironment analysis found most immune cells at higher concentrations in the low-risk group. Drug sensitivity analysis found 23 sensitive drugs. Twenty-one of these drugs exhibited heightened sensitivity in the high-risk group. RT-qPCR analysis validated the characteristics of 9 GMlncs. The predictive model and nomogram that we constructed have good application value in evaluating the prognosis of patients and guiding clinical treatment.

LINC00116 Is a Novel Prognostic Biomarker of Nonsmall Cell Lung Cancer

Abstract:Purpose: Lung cancer is the most malignant respiratory cancer with an undesirable prognosis. Emerging evidence shows that long noncoding RNAs (lncRNAs) can regulate lung cancer development. Currently, the role of LINC00116, a novel lncRNA, in lung cancer is unknown. This study was designed to evaluate the association and clinical significance between the level of LINC00116 and lung cancer.Materials and Methods: In the present study, 19 paired lung cancer and non-cancerous adjacent tissues were collected from lung cancer patients to measure the expression of LINC00116 by quantitative reverse transcription-polymerase chain reaction and Microarray analysis (Oncomine and CCLE). Clinicopathological features of patients were obtained to analyze their relationships with LINC00116 expression levels. Kaplan-Meier survival analysis was used to analyze the prognosis.Results: LINC00116 expression was upregulated in lung cancer. Furthermore, we found that LINC00116 expression was correlated with pT factor, pN factor, pTNM stage, smoking history, differentiation, and Ki-67 labeling index (p < 0.05). In addition, Kaplan-Meier survival analysis showed that high levels of LINC00116 expression were associated with poorer overall survival (OS), progression-free survival (PFS) and first progression (FP) (p < 0.05) of lung cancer patients.Conclusions: Our study provided evidence that LINC00116 is upregulated in lung cancer tissues and can predict poor survival. It might also be able to serve as a therapeutic target. The Clinical Trial Registration number 2022-020.
Downregulation of LINC00115 inhibits the proliferation and invasion of lung cancer cells in vitro and in vitro

Abstract:BACKGROUND: Lung cancer is a common malignant tumor in clinical practice. Its morbidity and mortality rank first among malignant tumors. However, the pathogenesis of lung cancer has not been fully clarified. This study found that LINC00115 is highly expressed in lung cancer tissues, but the role and molecular mechanisms of LINC00115 in the occurrence and progression of lung cancer are still unclear.METHODS: Fluorescence quantitative PCR was used to detect the expression of LINC00115 in lung cancer tissues and para-carcinoma tissues. Cell counting kit-8 (CCK-8), clone formation, and Transwell assays were used to detect the effects of LINC00115 knockdown on the proliferation, clone formation, invasion, and migration of lung cancer cells. Western blot was used to detect the effects of LINC00115 knockdown on the expression of epithelial-mesenchymal transition (EMT)-related molecules. Finally, a xenograft model in nude mice was used to detect the effect of LINC00115 knockdown on the proliferation of lung cancer cells in vivo.RESULTS: Compared with para-carcinoma tissue, LINC00115 was highly expressed in lung cancer tissue. Cell function experiments showed that knockdown of LINC00115 could significantly inhibit the proliferation, invasion, and migration of lung cancer cells. Western blot results showed that knockdown of LINC00115 could significantly inhibit the expression of the EMT-related proteins N-cadherin, vimentin, and fibronectin, and promoted the expression of E-cadherin. In vivo experiments in nude mice showed that knockdown of LINC00115 could significantly inhibit the proliferation of lung cancer tissues in vivo.CONCLUSIONS: LINC00115 is highly expressed in lung cancer tissues, and knockdown of LINC00115 can significantly inhibit the proliferation and invasion of lung cancer, which provides a theoretical basis for the design of targeted molecules for the subsequent treatment of lung cancer.

Abstract:Aims: This study aimed to determine the molecular markers related to gefitinib sensitivity for guiding the prognosis of lung adenocarcinoma (LUAD) and providing new evidence for promoting the precise treatment of LUAD. background: The World Health Organization stressed the need for molecular markers as diagnostic criteria for lung cancer in 2015 and 2021. Background: Lung adenocarcinoma (LUAD) is a prevalent lung cancer subtype with inferior survival outcomes. However, gefitinib is the first molecular targeted drug approved by Food and Drug Administration (FDA) to treat advanced LUAD. Gefitinib sensitivity-related gene targets for LUAD are rarely studied. objective: This study was designed to determine the potential molecular markers related to the sensitivity of Gefitinib in lung adenocarcinoma (LUAD), and to study their relationship with tumor biological traits. Objective: This study was designed to probe the potential molecular markers related to the sensitivity of gefitinib in LUAD. Methods: The gene expression profiles of LUAD cells in the Genomics of Drug Sensitivity in Cancer (GDSC) database were used for Weighted Gene Co-expression Network Analysis (WGCNA) to select the modules most related to gefitinib sensitivity. The Cancer Genome Atlas (TCGA) database was used to compare the expression of LUAD and para-cancerous tissues. Differentially expressed genes (DEGs) were then filtered and intersected with the highly linked genes in the module relevant to gefitinib sensitivity. Univariate Cox regression analysis was conducted to identify prognostically related genes to LUAD. The correlation between genes and drug IC50 was calculated by Spearman correlation analysis. Quantitative RT-PCR and immunofluorescence detection validated hub genes FAM13B and PFKP expressions. Results: Among the 10 modules divided by WGCNA, the module with the most significant positive correlation and the most significant negative correlation with gefitinib sensitivity were found. FAM13B, PFKP, FGD3, RNASE1, MUC16, GJB5, and GJB3 were hub genes related to gefitinib sensitivity in LUAD. Significantly, the low expressed FAM13 in LUAD tissues positively correlated with immune response. At the same time, overexpressed PFKP in the LUAD cohort was related to an unfavorable prognosis, cell proliferation, and cell cycle. We also found that FAM13B and PFKP expressions were enhanced in LUAD cell lines. Conclusions: This study identified 7 critical genes related to gefitinib sensitivity in LUAD. Functionally, genes positively correlated with gefitinib sensitivity might regulate the progression of LUAD through the immune, cell cycle, and metabolic pathways and showed potential effects in predicting sensitivity to different drugs. These findings help offer a theoretical direction for personalized treatment of LUAD.

Abstract:Background: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors, including gefitinib, are first-line drugs against advanced non-small cell lung cancer with activating EGFR mutations. However, the development of resistance to such drugs is a major clinical challenge. Methods: The role of annexin A5 in resistance to EGFR tyrosine kinase inhibitors was investigated by qPCR and western blot of relevant molecules, by CCK8 and EdU assay of cell proliferation and viability, by annexin V/propidium iodide assay of apoptosis and cell cycle distribution, by JC-1 assay of mitochondrial integrity, and by xenograft assay of tumorigenicity. Results: We found that annexin A5 is upregulated in gefitinib-resistant cell lines, as well as in clinical specimens resistant to EGFR tyrosine kinase inhibitors. Accordingly, knockdown of the gene from gefitinib-resistant cells restores gefitinib sensitivity in vitro and in vivo by downregulating polo-like kinase 1 signal pathway, thereby inducing mitochondrial damage, caspase activation, cell cycle arrest at G2/M, and, finally, apoptosis. Conclusions: The data indicate that annexin A5 confers gefitinib resistance in lung cancer by inhibiting apoptosis and G2/M cell cycle arrest, and is thus a potential therapeutic target in non-small cell lung cancers resistant to EGFR tyrosine kinase inhibitors.
LPAR5 Stimulates the Malignant Progression of Non-Small-cell Lung Carcinoma by Upregulating MLLT11.

Abstract:OBJECTIVE:This study aims to explore the diagnostic and prognostic values of Lysophosphatidic acid receptor 5 (LPAR5) in non-small-cell lung cancer (NSCLC) and its regulatory effects on biological functions of NSCLC cells.PATIENTS AND METHODS:NSCLC and adjacent non-tumoral tissues were collected for analyzing differential levels of LPAR5 by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Clinical information of recruited NSCLC patients was collected for assessing the diagnostic and prognostic values of LPAR5. In vitro regulation of LPAR5 on proliferative and migratory potentials of H1299 and SPC-A1 cells was examined by Cell Counting Kit-8 (CCK-8) and transwell assay, respectively. In addition, in vivo regulation of LPAR5 on the growth rate of NSCLC in nude mice was detected by tumorigenicity assay. The interaction between LPAR5 and its downstream target MLLT11 was determined by rescue experiments.RESULTS:LPAR5 was upregulated in NSCLC tissues than adjacent non-tumoral ones. High level of LPAR5 predicted higher rates of lymphatic metastasis and distant metastasis, as well as worse overall survival and progression-free survival in NSCLC. Knockdown of LPAR5 not only attenuated in vitro proliferative and migratory abilities in H1299 and SPC-A1 cells, but also slowed down in vivo growth of NSCLC in nude mice. MLLT11 was upregulated in NSCLC tissues, and displayed a positive correlation to LPAR5. Overexpression of MLLT11 was able to reverse the attenuated in vitro proliferative and migratory abilities, and the suppressed in vivo growth of NSCLC because of LPAR5 knockdown.CONCLUSIONS:LPAR5 stimulates proliferative and migratory potentials in NSCLC by positively regulating MLLT11, which can be served as an effective diagnostic marker for early stage NSCLC.

Abstract:Laminin is an extracellular matrix multidomain trimeric glycoprotein, that has a potential role in tumor progression. Here, we studied the effects of non-small cell lung cancer (NSCLC) cells interaction on laminin and explored the underlying mechanism of laminin associated NSCLC progression. Culture of A549 and NCI-1299 cells on 2D collagen gels (containing laminin) significantly promoted the proliferative and tumorigenic characteristics, as well as cell invasion of tumor cells in vitro . Consistently, comparing the clinical NSCLC tumor tissues, a poor overall survival was observed in patients with high laminin expression. Mechanistically, the expression of integrin α6β4 was required for the pro-tumor effects of laminin. Meanwhile, we showed that the downstream signaling of integrin α6β4, involved the focal adhesion kinase (FAK)/Yes-Associated Protein (YAP)/TAZ signaling pathway. The activation of FAK/YAP/TAZ signaling pathway induced by laminin was validated in tumor tissues from NSCLC patients. Suppression of integrin α6β4/FAK/YAP/TAZ signaling pathway efficiently suppressed the laminin-induced tumor growth, and strengthened the anticancer effects of chemotherapy, describing a novel target for NSCLC treatment.

Abstract:The purpose of the present study was to investigate the prognostic value of Leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5) in hepatocellular carcinoma (HCC) and its role in promoting HCC metastasis. The expression level of LGR5 in liver tumor tissues and adjacent non-tumor tissues were detected adopting immunohistochemistry (IHC), real-time PCR (RT-PCR) and western blot assays. Chi-square test was used to evaluate the correlation between LGR5 expression and clinicopathological characteristics. In addition, we assessed the relationship between LGR5 and two epithelial-mesenchymal transition (EMT) markers (E-cadherin and N-cadherin) in HCC tissues and cell lines. Our results showed that the expression of LGR5 was significantly higher in liver tumor tissues than in adjacent non-tumor tissues. Moreover, up-regulated LGR5 was associated with larger tumor diameter (> 5cm, P=0.001), higher TNM stage (P=0.021), increased recurrence (P=0.023) and growing metastasis (P=0.030). Besides, we found that the expression level of LGR5 was correlated with E-cadherin and N-cadherin. In conclusion, up-regulated LGR5 in HCC patients is associated with malignant clinicopathological characteristics. LGR5 may promote HCC metastasis through inducting EMT process, and thus can be regarded as a candidate biomarker for prognosis and as a target in therapy.
EPHA5 mutation predicts the durable clinical benefit of immune checkpoint inhibitors in patients with lung adenocarcinoma

Abstract:Immune checkpoint inhibitor (ICI) therapy has shown remarkable clinical benefit in lung adenocarcinoma (LUAD) patients. Genomic mutations may be applicable to predict the response to ICIs. Eph receptor A5 (EPHA5) is frequently mutated in breast cancer, lung cancer, and other tumors; however, its association with outcome in patients who receive immunotherapy remains unknown. In this study, we report that EPHA5 mutations were associated with increased tumor mutation burden (TMB), neoantigen load, levels of immune-related gene expression signatures, and enhanced tumor-infiltrating lymphocytes (TILs) in LUAD. LUAD patients with EPHA5 mutations in the immunotherapy cohort achieved a longer progression-free survival (PFS) time than patients with wild-type EPHA5. Immune response pathways were among the top enriched pathways in samples with EPHA5 mutations. In addition, patients with EPHA5 mutations tended to be more sensitive to certain targeted molecular inhibitors, including serdemetan, lox2, and PF1-1. Collectively, our results suggest that identifying mutations in the EPHA5 gene may provide insight into the genome-wide mutational burden and may serve as a biomarker to predict the immune response of patients with LUAD.

The association between SLFN 5 expression and the prognosis of non-small cell lung cancer

Abstract:Schlafen-5 (SLFN5) is one member of the Schlafens (SLFNs) family, which have significant effect on cell cycle and tumorigenesis. However, it remains unclear about the function of SLFN5 in the Non-small cell lung cancer (NSCLC). The aim of this study is to explore the underlying function of SLFN5 in the NSCLC. The results demonstrated that SLFN5 was weakly expressed in almost NSCLC tissues. Low-expression of SLFN5 protein was significantly associated with a number of clinicopathological variables, including tumor diameter, T classification, N classification and clinical stage. More importantly, the overall survival was obviously improved in NSCLC patients with highexpression of SLFN5 protein. In multivariate analysis, SLFN5 expression was significantly connected with early N classification. In addition, patients with high SLFN5 expression in N0 stage had improved survival. Furthermore, our results found that the knockdown of SLFN5 could increase the expression of migration-associated protein matrix metalloproteinase 9 gene (MMP-9) and the ability of A549 cell migration. Taken together, all data suggest that SLFN5 may be an important factor to improve the prognosis in NSCLC patients. It could be a potential biomarker to identify the prognosis of NSCLC patients.
