The effects of intrauterine growth retardation on the expression of mRNA of renal angiotensin Ⅱ receptor in rats

Abstract:Objective To study the effects of intrauterine growth retardation (IUGR) on expression of mRNA of angiotensin Ⅱ receptor of kidney in rats to provide experimental data for potential mechanisms underlying developmental programming of adult diseases. Methods The IUGR model of SD rats was established by feeding pregnant rats with low protein (6%) diet throughout pregnancy. Only male neonatal rats with IUGR were admitted in this study, their renal expressions of mRNA of AT1A、AT1B and AT2 receptors were measured by real time PCR at 1 day and 4 weeks of age, and their levels of plasma renin, angiotensinⅡ and aldosterone were measured by radioimmunoassay at 4 weeks of age. Results The expression of mRNA of AT1A and AT2 receptors was significantly decreased in rats of IUGR group (P<0.05) compared with control group. The expression of AT2 mRNA was significantly decreased (P<0.05) at 4 weeks of age, the expression of AT1A mRNA was increased compared with control group but no statistic significance (P>0.05). There was no significant difference of levels of plasma renin, angiotensin Ⅱ and aldosterone between control and IUGR groups. Conclusions This study suggests that abnormal renal expression of angiotensinⅡreceptor is possibly one of the molecular mechanisms to affect the development of kidneys and to cause hypertension in IUGR rats.
