THE PROTECTIVE FUNCTION OF PHYTOEATROGEN ON THE MITOCHONDRIAL ULTRASTRUCTURE OF THE HIPPOCAMPAL NEURONS OF THE OVARIECTOMIZED RATS

Abstract:In this study, the ovariectomized (OVX) animal model of Sprague-Dawley rats were used to imitate the pathologic changes of neural degeneration of postmenopausal women. The estrogen and phytoestrogen were given to model rats for observing the protective effect to the mitochondria of hippocampal neurons. The rats were randomly divided into 5 groups: (1) normal control group; (2) OVX 12-day (OVX-12) model group; (3) estrogen treatment group; (4) genistein treatment group; (5) ipriflavone treatment group. After 12 days, the ultrastructure change of mitochondria of hippocampal neurons of rats were observed and made a stereological analysis. The results showed: (1) the mitochondria swelled obviously, like empty-bubble, and the cristas broke and reduced or disappeared largely in the OVX-12 group, but the mitochondria were protected well, and the changes were like those of normal control group in the estrogen and phytoestrogen treatment group. (2) The volume density (Vv), average diameter (D) and average surface area (S) of mitochondria declined obviously (P<0.05), and surface area ratio, number density (Nv), particle dispersity (Cλz) of the three treatment groups aggrandized (P<0.05) comparing with the OVX-12 group. These results suggest that estrogen and phytoestrogen could withstand the degeneration of mitochondria better, which hinted that the mechanism of estrogen and phytoestrogen on the neural degeneration and Alzheimer’s disease after menopause could be explained via protecting the functions of mitochondria.
