Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:A synthetic approach to the previously unknown thieno[2,3-f][1,2,4]triazolo[1,5-a]azepine derivatives was efficiently accomplished by cycloaddition of the bicyclic 1-aza-2-azoniaallene salts 6, derived from 6,7-dihydrobenzo[b]thiophen-4(5H)-one hydrazones 4, to the triple bond of nitriles, followed by ring enlargement. The X-ray crystal diffraction analysis of the picrate 10a provided affirmative proof for the structural assignment made.

Abstract:A range of 2,4-dialkylidenetropinone-derived quaternary ammonium salts smoothly reacted with thiols in the presence of tributylamine, delivering structurally diverse thiolated 2,7-disubstituted tropones in moderate to excellent yields with high site selectivity. The reaction employs readily available feedstocks and reagents, is free of transition metals, tolerates various functional groups, and can be easily scaled up.

Abstract:The synthesis of a series of unprecedented tricyclic heterocycles, 1,2,4-triazolo[3,2-d][1,5]benzothiazepine 6-oxides (5a-i), was accomplished via cycloaddition of the allene-like cations 3, derived from thiochroman-4-one arylhydrazones, to the triple bond of nitriles and ensuing rings expansion annulation. The structure of one product (5h) has been unambiguously determined by X-ray diffraction analysis.

Abstract:Many [1, 2, 4] triazolobenzoheteroazepine derivatives show interesting biological activities. In this paper, the geminal arylazo isocyanato compounds 3 were prepared from chroman-4-ones in three steps, i.e. condensation with arylhydrazine, cycloaddition of the resulting hydrazone with HNCO, and oxidative ring cleavage by KMnO4. Compounds 3 were allowed to react with HBF4, producing the respective 3-spiro substituted 1-aryl-4, 5-dihydro-5-oxo-3H-1, 2, 4-triazolium salts 4. The concurrent rearrangement of compound 4 with insertion of the nitrogen atom into the carbon skeleton provided, after the basic work-up, 2-oxo-[1, 2, 4] triazolo [3, 2-d] [1, 5] benzoxazepines 5a-5h. An unambigous structutal proof was obtained from the X-ray diffraction analysis for compound 5h.

Abstract:A series of novel tricyclic O,N-heterocycles, [1,2,4]triazolo[3,2-d][1,5]benzoxazepin-2-thiones 7 were achieved via acid-induced ring closure of the geminal arylazo isothiocyanate compounds 5 which were derived from substituted chroman-4-ones, followed by feasible ring expansion with simultaneous insertion of the nitrogen atom into the carbon skeleton. Abstract:The palladium-catalyzed three-component cycloaminocarbonylation of 2-formylaryl tosylates with hydrazines and carbon monoxide has been established, which provides an efficient method for synthesis of substituted phthalazinones. Abstract:A one-pot protocol was developed for the synthesis of functionalized benzotropone derivatives via a nucleophilic phthalide ring opening by a 5-lithiated dioxinone derivative, followed by an intramolecular aldol condensation. The method demonstrates exceptional versatility with diverse substrates, yielding a variety of functionalized benzotropones. Subsequent transformations of the obtained benzotropone derivatives were explored for their potential applications.

Abstract:A novel strategy for constructing tropanes via [3+3] annulation of cyclic azomethine ylides with substituted 2-vinylindoles and 2-vinylpyrroles in the presence of in-house Cu(II)/N,O-ligand complex has been developed. The reaction was achieved via a tandem copper(II)-catalyzed Michael addition/BF3 center dot Et2O promoted intramolecular aza-Friedel-Crafts reaction of indoles or pyrroles with non-activated ketimines, providing indole or pyrrole-annulated tropanes in high yields (up to 87%) and excellent stereoselectivities (>99:1 dr in all cases, up to >99% ee).

Abstract:3-(Piperidin-4-yl)-6,7-dihydro-5H-pyrrolo[2,1-c][1,2,4]triazole was synthesized through a four-step process including etherification, hydrazonation, cyclization, and reduction with an overall yield of 39%. The final product was characterized by 1H NMR and ESI-MS/MS. The molecular structures of benzyl (Z)-4-(2-(pyrrolidin-2-ylidene)hydrazine-1-carbonyl)piperidine-1-carboxylate and related compounds were analyzed using DFT calculations at the B3LYP/6-311+G(d,p) level of theory. The results indicated a higher stability of the hydrazone tautomers.

Abstract:We describe here, for the first time, that 1-cyano-3-benzoyl-1,2,3,10b-tetrahydropyrrolo[2,1-a]phthalazine (8), prepared by 1,3-dipolar cycloaddition of 2-phenacyl phthalazinium bromide (6a) with acrylonitrile (7a), can be aromatized by tetrakis-pyridine cobalt (II) dichromate (TPCD) to give 1-cyano-3-benzoylpyrrolo[2,1-a]phthalazine (9a) in good yield. Furthermore, a general and convenient one-pot procedure for preparation of pyrrolo[2,1-a]phthalazines (9a-p) was developed by 1,3-dipolar cycloaddition of phthalazinium N-ylides (6a-c) with alkenes (7a-g) in the presence of TPCD.

Abstract:Abstract: Novel hydrazones 4, 5, and oxime 6 were produced from the reaction of ketone 3 with hydrazine, phenylhydrazine, and hydroxylamine HCl, respectively. One pot multicomponent reaction of chalcones 7a-c, 3-(1H-indol-3-yl)-3-oxopropanenitrile 8, and ammonium acetate in MeOH at reflux temperature gave 2-(1H-indol-3-yl)-nicotinonitrile derivatives 10-12. Additionally, a one-pot reaction of chalcone 7a, malononitrile or ethyl cyanoacetate, and ammonium acetate in AcOH at reflux temperature afforded 6-aminopyridine derivatives 13 and 14, respectively.
