Screening and Identification of Clock-related Genes in Peripheral Lymphocytes of Rat

Abstract:Objective To explore clock-related genes in peripheral blood lymphocytes of rats.Methods We used the method of differentially displayed polymerase chain reaction(DD-PCR) to display the differential genes at two timepoints,and then applied the technique of RNA interference to confirm the relationship between the screened EST and the Clock gene.Results Ten differential(cDNA) fragments were found,among which there were 3 known genes such as catalase,myelin(proteolipid) protein and histone acetylase,4 known EST and 3 novel EST.The levels of expression is all these screened genes or EST were regulated by the Clock gene.Conclusion There are at least more than three clock-related genes in peripheral lymphocytes of rat and their expressions is regulated by the Clock gene as an important component of the circadian oscillator.
