Abstract:This study amplified a 760bp DNA fragment derived from chicken growth hormone (cGH) gene by PCR. It contained a 5′ flanking sequence of 473bp from transcription initiation site, two potential TATA boxes, exon Ⅰ and part of intron Ⅰ of cGH gene. We amplified this fragment from seven strains including 14 chickens. These strains have different growth speed. Male-line broiler, a meat-type strain which has most high growth speed and heavy body weight; female-line broiler, a meat-type strain which has high growth speed, well laying ability and medium weight; Barred Plymouth Rock, a meat and eggtype strain which has medium weight; Red Island, an egg-type strain which has medium growth speed and weight; BAU Brown, a egg-type strain with medium growth speed and weight;White Leghorn, an egg-type strain with light weight and low growth speed; Silky fowl, a strain which has light weight and lowest growth speed but not dwarf. We cloned and sequenced these fragments. Compared with published sequence, there are three differences among these fragments and fragment which was published by Tanaka, M. (1992) as follow: deletion of C at the site of - 34bp, insertion of T between the site of + 160 and + 161bp and substitutions of G to A at the site + 175bp. Comparing the sequences of these fragments each other, we could conclude as follow: there are high conservation in these fragment, and no differences among these strains. It means that the difference of growth speed, and body weight among strains is probably not due to the difference in the promoter region of chicken growth hormone gene.
