Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.

Abstract:The chiral benzofuranone structural motif is a prominent feature in many natural products, which exhibit a broad range of biological and pharmaceutical activities. In the past few years, a survey of chiral benzofuranone derivatives based on asymmetric catalysis reveals an increasing number of papers, which reflects the latest achievements to facilitate the synthesis of sufficient quantities of related compounds as potential medicinal agents and biological probes. Recent advances in this area are summarized and classified according to the structure of the starting substrate.

Abstract:Chiral phosphoric acid-catalyzed couplings of C-alkynyl N,N '-di-(tert-butoxycarbonyl)-aminals with beta-naphthols led to chiral propargylamines in moderate to high yields with high to excellent enantioselectivity, in which the reactions underwent sequential chiral phosphoric acid-catalyzed in situ formation of N-(tert-butoxycarbonyl)-imines (N-Boc-imines) from the aminals, and 1,2-addition of beta-naphthols to the N-Boc-imines. Chiral 1,2-dihydronaphtho[2,1-b]furans and naphtho[2,1-b]furans were prepared with satisfactory results when 10 mol% AgOAc and 20 mol% 2,6-lutidine or 1.2 equiv. of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) were added to the resulting chiral propargylamines solution, respectively.

Abstract:A highly efficient asymmetric synthesis of dihydronaphthalenes is disclosed. The process represents a new addition to the limited asymmetric reactions of isobenzopyryliums, a family of versatile 10π-electron aromatic species. Excellent asymmetric induction is achieved for the first time without an anchoring group in the 4-position or a metal catalyst, both of which were required previously in these reactions. The success is attributed to the unusual chiral counteranion (meanwhile also the nucleophile) generated in situ from the chiral phosphate and the boronic acid as well as the leaving group. Preliminary control experiments provided important insight into the reaction mechanism.

Abstract:An efficient synthetic method for chiral trisubstituted diarylcyclopropanecarboaldehydes has been developed from substituted benzyl chloride and alpha,beta-unsaturated aldehydes. The reactions were catalyzed by chiral amine catalyst under mild condition to afford the chiral diarylcyclopropanecarboaldehydes in good to high yields and up to excellent enantioselectivities. Abstract:A novel chiral diallylic diamine has been synthesized through double addition of allyl zinc bromide to chiral imines in this contribution. Firstly, (S)-N-Boc-2-amino-3-phenylpropanaI was prepared through N-Boc protection and Dess-Martin oxidation. Then, it was condensed with (S)-valine methyl ester to form a chiral imine, which was reacted with allyl zinc bromide in the presence of CeC1(3)center dot 7H(2)O to afford the corresponding chiral homoallylic amine with high optical activity. After removing Boc group, the newly formed chiral homoallylic amine was condensed with 2-thiophenecarboxaldehyde to provide a novel chiral imine with high optical purity. The following asymmetric addition of allyl zinc bromide to the novel chiral imine provided the target compound. The structure and the purity of desired compound and the key intermediates were demonstrated by H-1 NMR, C-13 NMR and MS spectra.

Abstract:A series of 2-amino-3-eyano-4-arylbenzo[h]chromenes were synthesized by the reaction of arylmethlene malononitrile with 1-naphthol in the presence of triethylbenzylammonium chloride (TEBA) in water. The structures of all products were characterized by elemental analysis, IR and H-1 NMR spectra. Compared to the previous methods, this new protocol has the advantages of high yields, inexpensive medium and environmental friendlyness.

Abstract:An efficient and stereocontrolled synthesis of (20S)-camptothecin and an analogue has been developed. The key feature of this synthesis is the organocatalyzed asymmetric α-hydroxylation of the lactone precursor 4 to construct its stereocenter, providing tricyclic hydroxylactone 2 in 90% yield and with 88% enantioselectivity. The precursor 4 was efficiently synthesized from the known pyridine 5 in three steps.

Abstract:A catalytic asymmetric dearomatization reaction of benzyl 1-naphthyl ethers accelerated by a chiral N,Nʹ-dioxide/Co(II) complex is disclosed. The reaction proceeds via an enantioselective [1,3] O-to-C rearrangement through a tight ion pair pathway, providing a wide array of α-naphthalenone derivatives bearing an all-carbon quaternary center in high yields with excellent ee values.

Abstract:2,3-Dihydronaphazarin was prepared from 1,4-dimethoxybenzene and dichloromaleic anhydride. Reaction with bromochloromethane gave 1,8:4,5-bis(methylenedioxy)naphthalene which with NBS, gave 2-bromo-1,8:4,5-bis-(methylenedioxy)naphthalene. Reaction of the Grignard derivative with DMF afforded 2-formyl-1,8:4,5-bis(methylenedioxy)naphthalene. This method had several advantages compared with the reported synthesis, including fewer steps, milder condition and higher yields.

Abstract:An one-pot stereoselective synthesis of 2-aminobenzylidene derivatives from readily available 5-nitro/cyano-activated 2-halobenzaldehydes (2-chloro-5-nitrobenzaldehyde, 2-bromo-5-nitrobenzaldehyde, 2-fluoro-5-nitrobenzaldehyde, 5-cyano-2-fluorobenzaldehyde), the active methylidene compounds (cyanoacetamide and ethyl cyanoacetate), and secondary cycloamines via a novel parallel convergent Knoevenagel-nucleophilic aromatic substitution and nucleophilic aromatic substitution-Knoevenagel condensation cascade approach under mild conditions has been developed with high stereoselectivity and in 52-88% yields.

Abstract:Herein, a visible-light-induced chiral N-heterocyclic carbene (NHC) catalyzed asymmetric amination of ketenes has been developed. This strategy provides a convenient and facile synthetic protocol for the efficient construction of α-chiral amides under mild reaction conditions with moderate to excellent yields and stereoselectivities (up to 95% yields and 97:3 e.r.). Furthermore, chiral 2-phenyl-1-porpylamine derivative and (R)-ibuprofen could be obtained after simple transformations of chiral products.
