Abstract:We studied on EC1118,E01-2005 and PT yeast mutating by ultraviolet,and testing of resistant high sugar ability.The better yeast were screened out to test resistant high ethanol and ferment.The result showed the yeast E01-2005-1 whose capability of fermentation was more super,and the brewing wine's flavor was very nice.

Abstract:A high-yield yeast which can ferment(grow)well under the high sugar density has been screened and bred by UV mutagenization. The result showed when the yeast was irradiated for 30s with 15cm away from it under the dilution ratio 10-6,the yeast would grow best. At the same time,when the initial sugar density was 30% and the temperature was 20~22℃,the mutafacient yeast grew best,it would produce the best favor wine.

Abstract:Strain TJ-18 was screened by the methods of triphenyltetrazolium chloride(TTC),using heating and alcohol resistances from the different source of matrix.By the ultraviolet ray and nitrous acid compounding mutation,a stable high alcohol yield mutant strain UV-N-5 was gained which the alcohol content reached to 8.25%vol,39.83% higher than starting strain.The research of physiological characteristics showed that the yeast could growth normally in pH value 3-9,and kept a good resistance under the 40%-45% sugar concentration at 40℃.UV-N-5 had a resistance to alcohol of which the death rate was respectively 43% and 11% under the 12% and 8% of the alcohol concentration.All these characteristics could meet the requirement of yeast for modern alcoholic fermentation.

Abstract:In this study, a systemic analysis was initially performed to investigate the relationship between fermentation-related stress tolerances and ethanol yield. Based on the results obtained, two elite Saccharomyces cerevisiae strains, Z8 and Z15, with variant phenotypes were chosen to construct strains with improved multi-stress tolerance by genome shuffling in combination with optimized initial selection. After three rounds of genome shuffling, a shuffled strain. YZ1, which surpasses its parent strains in osmotic, heat, and acid tolerances, was obtained. Ethanol yields of YZ1 were 3.11%, 10.31%, and 10.55% higher than those of its parent strains under regular, increased heat, and high gravity fermentation conditions, respectively. YZ1 was applied to bioethanol production at an industrial scale. Results demonstrated that the variant phenotypes from available yeast strains could be used as parent stock for yeast breeding and that the genome shuffling approach is sufficiently powerful in combining suitable phenotypes in a single strain. Abstract:In order to screen out Saccharomyces cerevisiae suitable for table grape fermentation, and compare it with commercial Saccharomyces cerevisiae in terms of fermentation performance and aroma producing substances, differences of fermentation flavor caused by different strains were discussed. In this experiment, yeast was isolated and purified from vineyard soil, 26s rDNA identification and fermentation substrate tolerance analysis were carried out, and the causes of flavor differences of wine were analyzed from three aspects: GC-MS, PCA and sensory evaluation. The results showed that strain S1 had the highest floral aroma fraction, corresponding to its high production of ethyl octanoate and other substances, and it had the characteristics of high sugar tolerance. The fruit sensory score of S3 wine was the highest among the six wines. Through exploration and analysis, it was found that compared with commercial Saccharomyces cerevisiae, the screened strains had more advantages in fermenting table grapes. The flavor of each wine was directly related to the growth characteristics and tolerance of its strains.

Abstract:A yeast producing alcohol with high alcohol tolerance was screened from lees and named as S17.This strain was tentatively identified as Brettanomyces.The mutation of strain S17 by ultraviolet was carried out to improve its alcohol tolerance.The result showed that,with the ultraviolet mutagenesis for 120 s,a positive mutant was obtained,which could tolerate 20%(volume fraction) alcohol,enhancing 1% compared to that of the original strain.

Abstract:The Saccharomyces Cerevisiae YE0 was mutated using high-energy-pulse-electron(HEPE)beam. After ethanol stress and determination of the alcohol yield by gas chromatographs,the mutant YF1 with high alcoholic yield was obtained.The results showed that under the optimized fermentation conditions(34°C as the fermentation temperature,72 h as the fermentation time and 30%as the glucose concentration),the alcoholic yield of YF1 was 15.57%which was 58.23%higher than that of the original strain YE0(9.84%)under the same conditions.The growth rate and lethal temperature of the mutant YF1 were obviously enhanced to the original strain YE0.The mutant YF1 has a great potential application in industrial production of alcohol.And it can also be used as the original strain for further mutagenesis to get the strain of higher alcoholic yield.

Abstract:Fresh blueberry planted in Majiang,Guizhou was collected,then 40 mg/L SO2was added for its fermentation,and 13 strains were separated from the fermented mash.5 yeast strains with good gas production and enjoyable smell were further screened out through the test of ethanol resistance,sugar tolerance,low pH value tolerance,high temperature tolerance,and SO2tolerance.Those 5 yeast strains coupled with commercial yeast 71B and D254 were used for the production of blueberry wine in the experiments.And the results suggested that strain Z2 was the best among them(the alcoholicity of its fermented wine was 7.27 %vol,total sugar content was 4.83 g/L,total acids content was 6.5882 g/L,and total esters content was 0.669 g/L) and it was chosen for consequent test.

Abstract:Four strains of S. cerevisiae are used as material and the resistance for alcohol, SO2 and sugar are studied. The fermentation characteristic curve of S. cerevisiae is obtained and the quality of buckwheat wine is evaluated by comprehensive evaluation. The results show that all of the four strains of S. cerevisiae has good capacity for resistance of alcohol, SO2 and sugar and they can be used for wine brewing. The buckwheat wine fermented by Angel yeast exhibited better aroma, taste and quality than others samples. In the follow-up study, Angel yeast can be used for the fermentation of buckwheat wine.

Abstract:A strain of yeast named SD-01 for fermented drink production is screened from the bread dough with different grades of maturity after three level screening.The content of alcohol and acetoin is determined by three level screening.SD-01 is identified as Saccharomyces cerevisiae on the basis of morphological characteristics, colony morphology, physiological and biochemical tests.The result indicates that the optimal fermentation temperature is 26 ℃,with alcohol of 0.98 g/dL and acetoin of 0.97 g/L.The fermentation technique of metabolism reducing sugar,acid production, alcohol production, acetoin production is researched at 26 ℃.SD-01 has low alcohol, high acetoin and rich organic acids,is feasible for fermented drink production.

Abstract:A thermotolerant alcohol yeast (named UT1) was obtained by UVB mutation breeding and high-temperature culture of an alcohol yeast (named E12), and the main growth characteristics and the resistance capabilities against stress conditions of both the two yeast strains (UT1 and E12) were compared which could be used as references for the selection and the optimization of alcohol fermentation conditions. The results showed that the alcohol yield from UT1 at 40 ℃ reached up to 6.0 %vol, 11.1 % higher than that from E12. Through the comparison of the growth status of both the two strains under different environmental conditions (different temperature, pH values, high alcoholicity, high sugar content, and high salinity), it was reveal that UT1 strain has better tolerance to the change of temperature, acidity and salinity, which suggested that strain with stronger stress resistance is helpful to achieving higher alcohol yield.

Abstract:Three strains (7#,1# and 6# ) with good fermentation performance were screened from seven yeast strains through fermentation experiments. The sulfur resistance, acid resistance, sugar resistance, alcohol resistance and temperature resistance of the three strains were analyzed respectively and two yeast strains (7# and 6# ) which had excellent fermentation performance were selected eventually. The two strains had the following advantages: high yield, good sulfur dioxide resistance (80～120 mg/L), normal growth and fermentation under the conditions of pH 3.3～3.5, sugar content 24 %～27 %, and alcohol content 12 %～14 %Vol, low residual sugar content after fermentation, and good coagulation capacity. The wine fermented by the two strains had good quality and mellow fruit aroma and harmonious taste. The two strains could be used in wine production by mixed fermentation of lemon juice and sugar cane juice.

Abstract:Prepared the monolploid cell suspension of Y11 yeast strain which screened specially for mulberry wine making, then dealt the suspension with ultraviolet radiation and microwave treatment, and further decided the optimal dilution concentration in two mutagenesis factor by flat plated counting method. Described the optimal dose and mutagenized time of mutagenesis model by fatal rate and positive rate. First treated the start strain with ultraviolet radiation, then selected the perferable property strain for microwave treatment. Applied three-class mode for screening excellent yeast strain for mulberry wine making from large quantity flat plates.The results showed that ultraviolet radiation in 20~40s could obtain better mutagene effect(positive between 31.1%~41.2%), on the other hand, microwave treatment within 15s will be better(positive 22.1%). Enhanced mutagensis dose may increased the positive, but the risk of death increased at the same time. The ME44 strain in 2weeks fermentation cycle could obtain 11.7%(v/v) alcohol concentration which enhancede 7% to start strain, and fenmentation starting time and wine character enhancede simultaneity.
Isolation and Screening of High-alchol-yield Yeast with Strong Tolerance

Abstract:63 yeast strains were isolated from the nature by using the culture medium which contained high concentration of alcohol. After four-grade screening, one yeast strain, Cor-01, which had strong tolerance and could produce high-yield alcohol, was finally obtained. The alcohol tolerance of such strian was 20 %(v/v), the sugar tolerance was 22°Bix, and the lethal temperature of such strain was at 55 ℃. Sweet potato starch was used as raw materials for uncooked material fermentation, the ratio of material to water was 1∶2, after 72 h fermentation at 30 ℃, 100 r/min, the ethanol concentration could reach up to 16.2 % in fermented mash.

Abstract:120 Saccharomyces cerevisiae strains was preliminarily screened according to their polysaccharide production rate by phenol-sulfuric acid method.Then three strains with higher polysaccharide production rate were obtained.Then the fermenting properties of the three strains including fermenting power,low-temperature tolerance,SO2 tolerance,and ethanol tolerance were measured.As a result,NE-18 strain was suitable for dry white grape wine production whereas W-15 strain and NX-4 strain were not qualified because their fermenting power or tolerance could not meet the requirements.

Abstract:An excellent yeast strain was screened from ripe mulberry fruits,leaves and orchard soil by means of TTC coloration,aerogenic primary screening,big bottle secondary screening and enlargement test. The results of fermentation test showed that the strain could start fermentation quickly and produced 11.7% alcohol in 7 days. So it could replace the contrast strain for mulberry wine production. The results of morphological APl physio-biochemical identification and 18S rDNA sequence determination showed that the strain isolated was oval like,spore-forming but no-pseudohypha forming in corresponding medium. The results of physio-biochemical and molecular identification showed that it was mostly similar(similarity 97% by sequence blast)to saccharomyces cerevisiae WW-W23,named saccharomyces cerevisiae Sangguo.

Abstract:β-phenethylol,as an important flavoring substance in yellow rice wine,is mainly produced through the metabolism of yel-low rice wine yeast(S.cerevisiae).The increase of the yield of β-phenethylol by yellow rice wine yeast would significantly improve the flavor of yellow rice wine.In this study,S.cerevisiae strains were used as the starting strains for UV mutation,and the positive mu-tant strains were screened out by substrate analogue resistance test,alcohol tolerance test and yellow rice wine imitation liquid screen-ing.Then a positive mutant strain BYC-3 with high-yield of β-phenethylol was selected after multiparous spore purification screen-ing.The yield of β-phenethyl by strain BYC-3 reached up to 238.12 mg/L,2.67 times as much as that of the starting strains,and there was no significant difference in the ethanol-producing capacity between strain BYC-3 and the starting strains. This study provided useful reference for the development of high-quality yellow rice wine products.

Abstract:Although using non-Saccharomyces yeasts during alcoholic fermentation can improve the wine aroma, most of them are not ethanol tolerant; therefore, in 2017, this study screened 85 non-Saccharomyces yeasts isolated and identified from 24 vineyards in seven Chinese wine-producing regions, obtaining Pichia terricola strain H5, which displayed 8% ethanol tolerance. Strain H5 was subjected to ultraviolet (UV) irradiation and diethyl sulfate (DES) mutagenesis treatment to obtain mutant strains with different fermentation characteristics from the parental H5. Compared with strain H5, the UV-irradiated strains, UV5 and UV8, showed significantly higher ethanol tolerance and fermentation capacity. Modified aroma profiles were also evident in the fermentation samples exposed to the mutants. Increased ethyl caprate, ethyl caprylate, and ethyl dodecanoate content were apparent in the UV5 samples, providing the wine with a distinctly floral, fruity, and spicy profile. Fermentation with strain UV8 produced a high ethyl acetate concentration, causing the wine to present a highly unpleasant odor. To a certain extent, UV irradiation improved the ethanol tolerance and fermentation ability of strain H5, changing the wine aroma profile. This study provides a theoretical basis for the industrial application of non-Saccharomyces yeasts that can improve wine flavor.
