The Role of Transferrin Receptor in the Entry of Francisella tularensisis LVS into Macrophages

Abstract:To evaluate the role of transferrin receptor 1 in mouse macrophages infection with Francisella tularensis LVS.A shuttle plasmid pFNLTP6 gro-gfp was transformed by electroporation to F.tularensis LVS.F.tularensis LVS expressing GFP were used to infect murine macrophage J774A.1 cells.Transferrin receptor 1 was detected with mAb and visulization with a goat-anti mouse IgG conjugated to Alexa 594.Knockdown of Tfr1 in J774A.1 cells using siRNA.The transfected cells were infected with Francisella for immunoblot analysis or infection assay.We show here that the recruitment of transferrin receptor 1 on macrophage involved the uptake of bacteria.It was shown by western-blot that the siRNA-Tfrc_1 could knockdown of Tfr1 in J774A.1 cells about 75%.It was determined by infection assay,in knockdown of Tfr1 sample,bacteria number at 1 h infection with Francisella were not different from control(F=1.06,P=0. 3265),but after 24 h of infection the bacteria number were significantly decreased as compared with control(F=24.12,P=0.0006) These findings demonstrate that transferrin receptor 1 involved in the early phase of Francisella infection.It plays an important part for delivering iron to the host cell during infection.Knockdown expression of Tfr1 does not affect bacterial entry processes.Francisella,however,failed to proliferate in macrophages in which expression of the transferrin receptor was suppressed.
